Read More AbstractIntroductionRose is one of the most important ornamental plants that is widely cultivated in many countries. The optimal growth and development of commercial cut rose varieties is essential to supply water needed for plants. If the water level is reduced at different stages of production, it can have damaging effects on the vegetative properties of the cut rose. Therefore, accurate estimation of water requirement and adaptation of irrigation program based on water requirement of the plant and its growth conditions can improve water use efficiency, avoid stress and control production. Materials and MethodsIn order to evaluate different levels of irrigation requirement in combination with crop media on vegetative characteristics of rose CV. Samurai, a greenhouse experiment was conducted as split plot based on randomized complete block design with four replications in greenhouse conditions of Ferdowsi University of Mashhad. The needed irrigation of the plant 120%, 100%, 80%, 60% and sub plot consisted of three different substrate levels (Perlite, perlite_cocopeat 1:3, perlite _Vermiculite 1:3). The traits included a stem length, branch diameter, flower bud length and diameter, fresh and dry weight of flower bud, number of flowering branches, chlorophyll a, b, relative leaf water content (RWC). Results and DiscussionResults revealed that among the experimental treatments, water stress treatment had significant effect on all measured traits except flower bud length. However, culture medium treatment and interaction of media water stress did not affect all measured physiological and biochemical parameters except for branch length and flower bud diameter. The results of this experiment showed that the 120% and 100% water regimes significantly increased the measured physiological and biochemical traits (shoot length, fresh and dry weight of flower shoot, shoot diameter, number of shoots, RWC, chlorophyll a)included in this study. However, the 60% aqueous regime had adverse effects on the parameters considered while the highest amount of chlorophyll b was obtained in the 60% aqueous regime. In the present study, the results indicate that water stress has adverse effects on all vegetative characteristics of cut roses, therefore, it is suggested that the aquatic requirement of the plant be accurately estimated in order to achieve high quality roses. Also, the flower bud length traits are not significant at water stress levels, because plant roots are in close contact with water under cultivated conditions and respond to water stress by improving the electrical conductivity in the plant. Therefore, it is recommended that due to the negative effects of water stress on the quantitative and qualitative traits of rose, special attention should be paid to the problem of accurate estimation of water requirement and optimal water supply. Therefore, it is important to improve the vegetative and physiological traits of roses in non-soil conditions using appropriate cultivation for future studies in order to achieve desirable results. ConclusionOptimal water supply results in improved vegetative and physiological traits in the plant. Therefore in no-tillage conditions of rose samurai, 100% (control) and 120% irrigation requirement are recommended as the most appropriate levels of irrigation regime in Mashhad region.

Read More AbstractIntroductionZantedeschia, native to southern Africa, herbaceous flowering plants in the family Araceae, with common names such as Arum lily, Trumpet lily, Pig lily, or Miniature Calla Lily. Calla lilies are among the most colorful flowers with a unique flower form. It is one of the most important pot, cut flower and outdoor plant in the world. GA3 is sometimes used to stimulate growth and flowering of plants. The aim of this study was to investigate the effects of GA3 and different weights of rhizome on growth, development, as well as qualitative and quantitative properties, of Zantedeschia in the greenhouse. Materials and MethodsThis research was carried out to evaluate the effect of different concentrations of GA3 on micro-tuber of Z. pentlandii cv. Picasso. Micro-tuber in three sizes and average weights included; large (S1=6.20g), medium (S2=3.20g) and fine (S3=1.20g) soaked in GA3 solution with concentrations 0, 250, 500, 750 ppm for 30 minutes. Experiments were carried out in a factorial arrangement with randomized complete block design with three replications. 10 micro-tuber cultured in bed with compounds of 60% coco peat and 40% perlite in greenhouse conditions. From the time of planting to harvesting, the ability of micro-tuber sprouting, leaf length, leaf width, stem length, number of flowering stems, plant height, flower diameter (spathe), number of buds, bud length, the diameter and weight of the micro-tuber after harvest were evaluated.Results and DiscussionThe results showed that the maximum length of leaf (100% more than S3GA0) length of flowering stem, diameter and length of flower (spathe) were observed in S1 tubers treated with 500 ppm (S1GA500) and the least amount was obtained in control treatment (S3GA0). The fastest germination was in S1 tubers with 750 ppm (S1GA750 36% earlier than S3GA0) and the earliest flowering was in S2 tubers with 500 ppm treated (S2GA500) which have flowering 28% earlier than S3 tubers with 250 ppm (S1GA250) but non-GA treated tubers in all three sizes have none flower. The highest weight and diameter tuber belonged to S1 tubers with 500 ppm and the lowest weight in S3 tubers without treatment. S3 tubers did not enter the flowering stage. It was shown that the GA3 treatment on Zantedeschia ‘Picasso’ tubers caused earlier flowering and the formation of a greater number of flowers and leaves. GA3 decreased the time between germination and appearance of flower. In simple effects study, large tubers (S1) and GA500 treatment were superior to other treatments in almost all traits. Also, the method of soaking of tubers with GA3 can be used in production scheduling, early efficiency and selection of superior populations in breeding programs. ConclusionThe best growth and flowering indices were S1 tubers with treatment of 500 ppm. S1 tubers with GA3 (500 ppm) treatment were superior in all traits to other treatments. The method of soaking of calla tubers with GA3 can be used in production, increasing efficiency and selecting superior populations in breeding programs.

Read More Background and ObjectivesMicrografting is a relatively new technique for propagation of plants. This technique has been used on woody species to produce viruses-free plants, rejuvenation, and reinvigoration, analysis of grafting compatibility and incompatibility and clone's propagation. Pistachio (Pistacia vera L.) is one of the most important commercial trees grown in arid and semi-arid regions of Iran where it is of vital importance to the permanent farming. Increase of pistachio cultivation areas during last three decades caused a continuous growing demand for pistachio planting materials in Iran. The benefits of applying in vivo and in vitro micrografting to the pistachio are obvious, especially when considering the need to improve the genetic quality of the planting stock of this slow-growing species while increasing crop potential. The objectives of this study were to determine the influence of scion storage and grafting time on clonal propagation of pistachio from elite-mature trees. Materials and methodsIn order to evaluate the effect of scion storage and grafting time on grafting success of in-vivo micrografting of Pistachio, an experiment was achieved an factorial based on compeletly randomized design (CRD) during 2017-2018 under greenhouse condition. In the experiment, the success of micrografting in "Ahmad aghaiee " Pistachio on 14-days "Badami-Riz-Zaran" rootstocks were examined. The scion of "Ahmad aghaiee" were prepared from shoot tips of mature trees at three different times (February, march and may) and grafted immediately or after one month storage at 4 °C using inverted cleft grafting method. 21 days after grafting, some traits such as graft union percentage, scion growth, time to graft union, scion leaf number and total soluble sugar were recorded.ResultsThe results showed that the highest graft union percentage (100%) was observed on the grafted plants of February, immediately after grafting. Based on the results, soluble sugars were significantly increased in freshly harvested scions in February and March. The results also showed that the maximum time (18 days) for time graft union was obtained in April with the stored scion whereas the lowest time graft union (11 days) was observed in February with scion taken at the same time. In addition, we found that the highest scion growth rate was due to freshly harvested scions and grafts in February.DiscussionIt was found that February was the best time of micrografting with scion taken at the same time because in this period carbohydrate content of the scions was higher than other months. Also, pistachio contain high amount of phenolic compounds. Exudation of phenolic compounds from the cut surfaces and their oxidation by polyphenoloxidase and peroxidase enzymes cause discoloration of the tissues which results in poor micrografting.

Read More Abstract Background and Objectives Red flesh apple belongs to Malus niedzwetzkyana of Rosaceae family. It has high levels of important phytochemicals like antioxidants, flavonoids and anthocyanins in its cortex, which, in addition to attractiveness, is a source of benefit compound. The aim of the present investigation was to optimize efficient protocols for micro propagation of red flesh apple in order to use the results in future breeding strategies. Materials and Methods During the early spring, stem cuttings of growing red flesh apple trees were surface-sterilized in 70% ethanol and 50% sodium hypochlorite. Then, sterilized samples were placed in the test tubes containing MS medium supplemented with 2 µM BAP. At the proliferation stage, interactive effects of BAP (0, 2, 4, 6 mM) and GA3 (0, 3, 6 mM) on various aspects of proliferation (multiplication rate and vegetative growth characteristics such as number of produced shoots per explant, shoots length, leaf numbers, and the fresh and dry weight of explants) were investigated. At rooting stage, the effect of different concentrations of IBA or NAA (0, 1, 2, 3,4 µM) on the percentage of rooting, number of roots per explant and length of roots were compared. At acclimatization stage, the plantlets were transferred into pots with a volume of 200 ml containing sterile compound at the greenhouse condition. Results The results showed that there was a significant difference between the effects of various concentrations of BAP and GA3 on micro propagation of red flesh apple. The best result in proliferation (7.9 of axillary shoots after 8 months) was obtained from explants that were cultured in the medium containing 4 mM BAP and 3 mM GA3. At rooting stage, the maximum rooting percentage (85.2%) was obtained in the medium containing 3 mM IBA. In addition, rooted explants from IBA treatments were successfully acclimatized (100%) in the greenhouse, but rooted explants in NAA treatments were weak in acclimatized stage (40%). Discussion Measuring the growth rate is the major economic parameter for successful plant production at in vitro conditions, which is usually characterized by the number of axillary shoots, number of regenerated leaves and increase in the length of shoots and fresh and dry weight. We obtained the highest growth rate of red flesh apple in the culture medium containing 4 μM of BAP. The efficiency of BAP as an important cytokinin in apple proliferation has been reported in other studies as well. The results of the present investigation showed that highest rooting percentage was obtained from culture medium containing 3 μM of IBA. In addition, morphology of roots varied in callus formation and longitudinal growth and the samples rooted in IBA treatments had stronger roots than the rooted samples in NAA treatments. It seems that exposure to NAA instead of IBA stimulates indirect organogenesis and produced undesirable side effects on roots, such as callus formation, which can cause plantlets weakness during the period of acclimatization.

Read More Abstract Background and Objectives Salinity stress is one of the major environmental factor limiting plant growth and productivity. The adverse influence of salinity stress is evaluated on all plant levels. Ascorbic acid is a small, water-soluble antioxidant molecule that acts as a primary substrate in the pathway for detoxification and neutralization of superoxide radicals and singlet oxygen. Ascorbic acid has been shown to play multiple roles in plant growth, such as in cell division, cell wall expansion, and other developmental processes. The objective of the present experiment was to investigate the effects of the exogenous application of ascorbic acid on the growth parameters and photosynthesis attributes of tomato plants. Materials and Methods The experiment was conducted as a factorial in a completely random design with three replications in greenhouse conditions. Salinity stress was applied by 0, 25, 50, 100 mM NaCl in modified Hoagland solution. Ascorbic acid treatments (0, 2.5 and 5 mM) were sprayed on the leaves every three days until the end of experiment. Leaf, stem, and root fresh and dry weight, total fresh and dry weight, yield per plant, net photosynthesis, Stomata conductance, transpiration, internal CO2 concentration, chlorophyll a, b and total chlorophyll, proline and total soluble sugar content were evaluated. Results In the present experiment, salinity stress sharply reduced the leaf, stem, root and total fresh and dry weight, yield per plant, net photosynthesis, stomata conductance, transpiration, internal CO2 concentration, chlorophyll a, b and total chlorophyll content whereas the exogenous treatment of AsA appreciably delayed the loss of all studied traits. Foliar application of ascorbic acid under both stress and non-stress conditions improved the growth parameters and photosynthesis attributes. Salinity stress also increased total soluble sugar content and proline accumulation in the leaf of the tomato plant. Application of ascorbic acid under both non-stress and salinity stress conditions decreased total soluble sugar content and proline accumulation and reduced the effects of salinity stress. Discussion This experiment showed that treating the tomato (Solanum lycopersicum L. cv. Mobin) with 5 mM ascorbic acid substantially influences several metabolic processes, leading to the increased growth. The protection of the plant against salinity stress by using of ascorbic acid was possibly to be caused indirectly as a result of increasing amount of chlorophyll which plays an essential role in photosynthesis process and then formation of carbohydrates. Thus, it might be concluded that exogenously applied ascorbic acid has low external osmotic potential and ion toxicity and it might be effective in amelioration of the adverse effects of salt stress.

The results of various studies have shown that the ecological advantage of intercropping is due to the efficient use of environmental resources. The purpose of mixed cultivation experiments, especially forage plants, is to increase the yield of forage in quantitative and qualitative terms, maintain the ecological balance and stabilize the system. Materials and Methods In order to compare the forage yield and determine the best planting ratio in mixed cropping of Triticale, Barley, Crimson clover (Trifolium incarnatum L.), and Vicia sativa, an experiment was conducted based on the complete randomized block design with sixteen treatments and three replications in paddy fields of Rice Research Institute of Iran (Rasht) during 2012-2014 for two years. Experimental treatments included pure planting of Triticale, Crimson clover, Vicia sativa, and Barley, planting mixtures of Triticale and Crimson clover with ratios of 70:30, 50:50 and 30:70, planting mixtures of Triticale and Vicia sativa with ratios of 70:30, 50:50 and 30:70, planting mixtures of Barley and Crimson clover with ratios of 70:30, 50:50 and 30:70 as well as planting mixtures of Barley and Vicia sativa with ratios of 70:30, 50:50 and 30:70. The studied traits included plant height, number of stems, number of tillers, number of plants per m2, fresh forage yield, dry forage yield as well as land equivalent ratio (LER) values for fresh and dry forages. The obtained data were analyzed by SAS software and mean comparisons were performed using LSD test. Results The results of analysis of the variance showed significant differences between the experimental treatments in most traits. Mean comparison showed that planting ratio of 30% Vicia sativa + 70% Triticale had maximum fresh and dry forage yields with averages of 48844 and 15269 kg. ha-1, respectively. Most treatments had LER of greater than one. Planting ratio of 70% Triticale and 30% Vicia sativa showed LER values of 1.31 and 1.54 for fresh and dry forage yields. The treatment is recommended for planting in paddy fields of Guilan area due to its relative advantage with respect to other planting ratios. Discussion Considering that the main objectives of crop forage production in rice paddy fields are the optimum use of paddy fields, sustainability of rice production, production of forage for livestock, and increasing farmers' incomes, the cultivation of all treatments tested as pure and mixed crops in rotation with rice has no problem with regard to the interference of harvesting of the forages with the time of cultivating the seedling rice.

Read More Abstract Background and Objectives Tomato (Solanum lycopersicum L.) is one of the vegetable crops of a global economic importance. The reduction of genetic variation in tomato through domestication and breeding has resulted in the need for conservation and utilization of all existing genetic resources. Local populations are among the most important genetic resources. They are often heterogeneous and composed of different genotypes which are mostly homozygous. Therefore, assessment of their diversity requires a different approach of common methods. The objective of this study was estimating the phenotypic variation among and within the tomato local populations of Kalaybar area using Nei’s genetic diversity index. Materials and Methods Plant material consisted of 8 tomato local populations collected from Kalaybar area in the East Azerbaijan province.The experiment was carried out as a randomized complete block design with three replications. A total of 36 plants of each population in three blocks were planted with the distance of 140 cm between rows and 80 cm between plants. Seven traits, including fruit yield and some of its related traits, was measured. All continuous traits were divided into three classes with equal width and the frequency of classes was used to estimate total diversity index (HT), phenotypic diversity within each population (Hs) for each trait. Then, mean diversity index () for all traits, proportion of phenotypic diversity among populations (GST) for each trait, and mean diversity index for each population across all traits () were calculated. Results The total diversity index (HT) for each trait ranged from 0.46 to 0.53 with an average of 0.49. Most of values were around the mean. All traits had relative high value of mean diversity index ( > 0.40). The mean phenotypic diversity among populations (GST) was 0.10. For the majority of traits, little diversity was observed among populations. The greatest value of GST (0.16) was relevant to the mean of fruit weight per plant. Most of population had similar magnitudes of mean phenotypic diversity within each populations () across all traits (0.42 to 0.49). The fruit yield (0.51) and mean of fruit weight per plant (0.51), the number of fruit per plant (0.56), the number of node on stem (0.54), the number of floret (0.54), the number of days to flowering (0.44), and the number of days to fruit (0.54), had the highest diversity index (Hs) values in Gharavanloo, Rostamkandi, Sheykhlan-sofla and Kalantar, Sheykhlan-olya, Alisharloo, and Sheykhlan-olya populations, respectively. Discussion The results showed a relatively wide phenotypic diversity present in tomato local population of Kalaybar area for most of the studied traits. The highest diversity was within the populations and there was little variation among the populations. The traits that contributed the most to within populations’ heterogeneity differed from population to population. However, the Sheykhlan-olya population had the highest fruit yield (4.83), the number of fruits (18.4), the number of nodes on stem (20.7), the number of flowers (83.5), and it was concluded that population could be used as a genetic source for future tomato breeding programs in Kalaybar region.

Read More Abstract Background and Objectives Apricot is one of the most important horticultural crops because of its attractive taste and nutritive value. Different cultivars of apricot contain highly variable content of ascorbic acid, cartenoids, and phenolic compound,which are good sourecs of antioxidant. Fuit quality is an important trait for concumers and producers. The purpose of this work was to estimate the effects of pre-harvest application of polyamines on quality characteristics of apricot fruits at the end of fruit ripening process. Materials and Methods The experiment was carried out as a factorial based on the randomized complete block design (three concentrations of spermidine at three harvesting time) with four replications. The selected branches of trees were sprayed as a pre-harvesting application method by three concentrations of spermidine (0, 2, and 3 mili molar) 7 days before commercial harvesting time. Fruits were harvested during harvesting times (3 days interval) and then some fruit characteristics such as total soluble solids (TSS), total ccidity (TA), vitamin C, firmness of fruit tissue, pH of fruit extract, total phenol, flavonoid, and total antioxidant capacity were measured in the laboratory of flower biology and fruit growth development at the department of horticulture, faculty of agiculture, university of Tabriz. Results The results showed that the effects of different harvesting times were significant regarding all traits. The best results were observed in 3 mM spermidine and this treatment improved all the measured characteristics of fruit. The highest value of total antioxidant capacity (3.01%), vitamin C (3.29%), and fruit firmness (3.27%) were found in 3 mM spermidin at three harvesting times. In some characteristics such as TSS and TA, there was no significant difference between 2 and 3 mM spermidine, but among treated and non-treated fruit a full significant difference was observed. We assume that the second harvesting time can be the best harvesting time regarding fruit quality parameters. During fruit ripening, the amount of TSS and TA increased, while firmness, total phenolic, and flavonoids compounds and total antioxidant capacity decreased gradually. Discussion Fruit physicochemical properties are very important for understanding the product behaviour during the harvesting, transporting, packaging, and storing of fruit crops. The results of this study showed that the pre-harvesting application of polyamines delayed the ripening and increased the quality of fruit during the different harvestings times.

Read More Abstract Background and Objectives Iris Germanica has become the most well-known flower for landscape planning due to its colorful flowers, reproduction by creeping rhizomes, flat leaves, and a trio of flower parts, resistance to calcareous soils and adverse environmental conditions, and low watering requirements. Given the pharmaceutical-ornamental importance of I. germanica, this research was conducted to find new hybrids and their significant traits. Diversity and innovation in the field of flowers and plants are good, the world is presented with hundreds of colors and varieties and given the importance of I. germanica, the management to achieve hybridization between the new hybrid was performed. Materials and Methods Three genotypes of I. germanica with brown and blue (female parent) and yellow (paternal line) colors were selected for hybridization out of a total of 15 progenies and three parents to be evaluated in a randomized complete block design with three replications. The research was conducted in Ornamental Plants Research Center (OPRC) in Mahallat from 2014 to 2016. Quantitative traits included leaf width, peduncle length, peduncle thickness, flower diameter, outer petal width, inner petal length, inner petal width, bush length, flowering branch diameter, and crown diameter. The statistical analysis was conducted in the SAS and Excel software packages. According to the instructions of UPOV, 10 quantitative traits and some statistical parameters such as simple correlation coefficients, estimates of broad sense heritability, phonotypical and genetically coefficient of variations were measured. Results Results showed significant (P≤0.01) differences between genotypes in all studied traits, indicating wide variations in traits except for the flower diameter. The correlation coefficients of quantitative traits among genotypes (parents and progenies) showed the most positive and significant correlation between the diameter of flowering branch and crown diameter, peduncle length, and inner length of petal. The highest broad sense heritability was related to the outer petal width (96.28), inner petal length (93.66), and bush length (93.40) and the lowest was related to peduncle length (42.58). The findings indicated that hybrid NIOP8 had the highest stem diameter. It was also superior in crown diameter, flower branch diameter, and peduncle diameter versus the other hybrids. The results indicated that hybridization has a significant effect on the traits. Discussion The hybrids NIOP5 and NIOP8 outperformed the other hybrids and parents significantly in the length of inner petal, the width of the inner petal, and leaf width.

Effect of Harmel Extract on Bacterial Population in Vase Solution and Vase Life of ‘Stanza’ and ‘Pink Elegance’ Cultivars of Gerbera Cut Flower

In cut flowers, water deficits are created when transpiration is more than water absorption. Therefore, petal wilting and neck bending increase in these flowers. These conditions are mainly due to occlusion of the vascular bundle, which can occur in different ways, such as the activity of pathogenic bacteria and other microorganisms in vascular bundle. In recent years, due to the adverse effects of chemical antimicrobial compounds on human health and the environment, many studies have been conducted to find replacement materials to maintain the quality and vase life of cut flowers in postharvest. Therefore, in this study, the effect of different concentrations of harmel aqueous-alcoholic extract (Peganum harmala L.) on bacterial population and vas life of ‘Stanza’ and ‘Pink Elegance’ cultivars of Gerbera cut flower were studied. Materials and MethodsThe effect of concentrations 0, 0.25, 0.5, 0.75 and 1% of the aqueous-alcoholic extract of Harmel plant was studied in a factorial experiment based on completely randomized design with four replications. At harvest time, 3, 6 and 9 days after harvest (at 20 °C and 70% relative humidity), the effect of treatments on the bacterial population of the vase solution and end stem, vase life and some characteristics of ‘Stanza’and ‘Pink Elegance’ Gerbera cut flowers were studied. ResultsThe results showed that the inhibitory effect of harmal extracts on gram-positive bacteria was higher than gram-negative bacteria. Also,‘Stanza’cultivar had better quality and longer vase life than ‘Pink Elegance’ cultivar. By increasing the vase life of flowers, the quality of these flowers decreased. In the first place, however, concentrations of 0.25% and then 0.5% of the harmal extract performed better than other treatments in various aspects: first, they reduced the bacterial solution, stem end, and electrolyte leakagein stem and petals; second, they preserved quality (discolored of vase solution, discolored of petals and stem), relative fresh weight, stem diameter, totalsolublesolids, catalase activity and phenylalanine ammonia lyase activity; and finally, they increased vase life of gerbera flowers in both cultivars. Concentrations of 0.75% and 1% increased allelopathic substances in cut flower. While these treatments decreased the bacterial population in vase solution and stem end, they did not have a significant effect on the quality and vase life of gerbera cut flowers. DiscussionThe results of this study showed that the different concentrations of harmal extract significantly reduced the growth of microbial agents, including the bacterial population, but by increasing the concentration of the harmal extract, the allelopathic effects of this plant extract on gerbera cut flowers were harmful and did not affect their vase life and quality. Therefore, the concentration of 0.25% then 0.5% reduced the bacterial populations in vase solution and stem end of cut flowers and preserved the quality and vase life of ‘Stanza’ and ‘Pink Elegance’ cultivars. Therefore, the use of low concentrations of the harmel aqueous-alcoholic extract is recommended to reduce the microbial population of the vase solution and maintain the quality and vase life of the gerbera cut flowers.

Read More Abstract Background and Objectives Tomatoes are considered the favorite and most widely consumed vegetables in the world since their fruit is rich in minerals, vitamins and antioxidant compounds. Tomato fruit is harvested after physiological maturity at different color stages based on consumption type. Mature unripe tomato fruit could be stored for long time and be ripened according to market demand, but this tropical product is susceptible to low temperature, which imposes serious constraints in postharvest period and supply chains. Several techniques or chemical treatments have been used to decrease low temperature deleterious effects. Gamma-mino butyric acid (GABA), as an efficient tool, could increase plant resistance to chilling injury (CI) during postharvest life. This study was carried out to evaluate the effect of pre-harvest GABA spray on postharvest quality changes and chilling of tomato fruit during cold storage. Materials and Methods In this study, the effect of pre-harvest application of GABA at three concentrations (0, 5 and 10 mM) on postharvest quality and chilling of tomato cv. Newton was evaluated. The spray was carried out 2 weeks before harvest. Fruits were harvested after physiological maturity when the fruit color changed from pink to red. During storage period at 4 °C and 90-95% relative humidity, some fruit quality attributes such as weight loss, firmness, chilling injury of tomato fruit, soluble solid content (SSC), titratable acidity (TA) and fruit bioactive constituent were analyzed at days 0, 5, 10, 15 and 20. Results Chilling in control fruit was 25% at final assessment, while in 10 mM GABA treatment it was only 8.33% at the same time. GABA treatments decreased the rate of weight loss and retained fruit firmness during storage. Also, vitamin C and titratable acidity in GABA treatments were higher than those of untreated fruit. Fruit lycopene content increased during fruit storage in both treated and untreated fruit, but the value was greater in GABA treatments than the control. SSC increased during fruit storage, but it was greater in control than the treatments. While superoxide dismutase enzyme activity decreased in both treated and untreated fruit, the rate of decline in control fruit was greater than GABA treatments. Discussion Pre-harvest foliar application of GABA ameliorated chilling injury induced by cold storage, maintained quality properties of tomato fruit, and increased lycopene content. GABA treatments probably lessened deteriorate effect of low temperature through keeping fruit cell integrity and higher bioactive constituent levels.

Due to low efficiency of propagation trough cutting, tissue culture is a good and fast method for propagation of disease-free plants of GF677. In vitro multiplication efficiency in GF677 is widely dependent on the type of culture medium and growth regulators. Therefore, the aim of this study was to determine the conditions required to optimize micropropagation methods for GF677 rootstock from nodal explants. Materials and Methods This experiment was conducted during 2015 at the laboratory of plant tissue culture at Agricultural and Natural Resources Research Center of Kermanshah, using a factorial in a completely randomized design with three replications. Sterile nodal explants were cultured onto different media of MS, WPM and B5 supplemented with benzyl adenine (BA) at concentrations of 0.25, 0.5, 1, 2 and 4 mg/L and indole-3-butyric acid (IBA) at concentrations of 0, 0.1, 0.25 and 0.5 mg/L. Elongated shoots of GF677 were cultured on MS medium supplemented with 0.25, 0.5, 1.0 and 2.0 mg/l IBA and 0.0, 0.1, 0.2 and 0.5 mg/l BA for rooting. Factorial analysis of variance was carried out and differences between means were scored with LSD tests. Results The effect of different culture media (MS, B5 and WPM) and plant growth regulators (BA and IBA) on a number of shoots per proliferated explant of GF677 proved that the highest rate of adventitious shoot initiation, percentage of egeneration, shoot length and diameter, number and length of nodal, leaf number and multiplication was obtained in MS medium containing 1 mg/l BA + 0.5 mg/l IBA. Cytokinin stimulates the initiation and activity of axillary meristems, which result in shoot formation. This study showed that the number of shoots increased as concentration of BA increased to a certain amount. As concentration of BA increased to 1 mg/l, the number of shoots increased, too. It seems that there is a positive correlation between concentration of BA and the number of shoots to a certain concentration of BA. At concentrations higher than 1 mg/l BA the number of shoots increased. One of the possible reasons can be the reductive effect of higher concentrations of BA. Apparently, a certain amount of BA is required to obtain the best effect. Higher concentrations of BA cause formation of high amount of callus, which is not appropriate in tissue culture. The results of this experiment confirmed the positive influence of the growth regulator on the number of roots per shootlet. Among the various plant growth regulators used, the highest rate of rooting (33%) and the number of roots per shootlet (1.62) were obtained on MS medium containing 1.0 mg/l IBA and 0.5 mg/l BA. The concentration of auxin during rooting period strongly influenced the quality of root system during the rooting period. Rooted plants were transferred to a combination of terrestrial environments, including perlite, sand and soil in the ratio of 1: 2: 1, respectively. Among rooted plantlets that were acclimatized and transferred to the potting mix successfully, 90% survived and grew naturally after strengthening and transferring to the soil. Discussion The shoot multiplications are influenced by the media and growth regulators. The MS medium gave the best results for the proliferation of cultures from explant among the tested media (MS, B5 and WPM). Growth regulators compounds have significant effects on different traits and these changes depend on type and concentration of hormone. Both cytokinin and auxins are important in micropropagation for GF677 rootstock.

Since it contains various secondary metabolites and shows noticeable activity against different Gram-positive and Gram-negative bacteria, nettle has been identified as an appropriate choice for wider applications in the food and pharmaceutical industries in the recent years. This plant is native to Asia, Europe, Africa and North America and grows naturally in different regions of Iran. Although nettle is an important medicinal plant worldwide, there is no information about optimization of its tissue culture conditions. This study was carried out to investigate the effects of different plant growth regulators, explant types and light condition on the callogenesis properties of nettle plant under in vitro condition. Materials and MethodsA factorial experiment was carried out on the base of completely randomized design with three replicates. Different explants including young leaves, old leaves, nodes and seeds of nettle plant were cultured in the solid MS medium supplemented with 30 g l-1 sucrose and different concentrations of plant growth regulators of 2,4-D (1, 2 and 3 mg l-1) and BA (0.1, 0.5 and 1 mg l-1). Callus-related characteristics, including time of callus induction, callogenesis percentage, and their color and texture were analyzed. Analysis of variance was carried out by SAS software ver. 9.3.1 and mean comparison was conducted using Duncan multiple range test. ResultsAccording to the results, callus induction was significantly influenced by explant type. The highest callus induction was attained from young leaves, nodes and old leaves, respectively. The results also showed that callus induction and its properties were significantly influenced by light condition. Preservation of cultures in dark condition resulted in higher callus induction than light condition. In addition, the effect of plant growth regulators was significant on callus induction and its properties. The highest percentage of callus induction was obtained from young leaf explants in the medium containing 1 mg l-1 2,4-D and 1 mg l-1 BA in dark condition followed by node explants in the same plant growth regulator treatment. However, medium supplemented with 1 mg l-1 2,4-D and 0.1 mg l-1 BA showed to be more effective for old leaf explants. Based on the results of this research, young leaf explants in the MS medium containing 1 mg l-1 2,4-D and 1 mg l-1 BA in dark condition are suggested for callus induction from nettle plant. DiscussionType, color and amount of produced callus in different plant species are highly dependent on the genotypes, compositions of tissue culture media as well as the levels of both endogenous and exogenous plant growth regulators. Auxin and cytokinin are two main plant growth regulators playing important roles in callogenesis of different plant species. There is an interaction between the level of plant growth regulators and color of produced callus. Based on the results of present study, it seems that in the nettle plant, node explants endure a higher level of plant growth regulators and hence produce a brighter callus than the leaf segment explants when they are treated with high concentrations of these chemicals.

Hook f.) is known as the world's fourth cut flower and silicon (Si) is considered the second most abundant element (28%) of the Earth's crust.Silicon deposition in most tissues and organs, such as plant cell wall, the space between the cells of the epidermis, roots, leaves, and reproductive organs causes resistance to environmental stresses, insects, and pathogens into the plant. Greenhouse whiteflies (Trialeurodes vaporariorum) are the most important pests in several crops such as gerbera, which have herbivorous insects with piercing-sucking mouthparts and have 4 to 15 in their generation. Materials and MethodsConsidering the numerous environmental risks posed by using pesticides in pest control especially under greenhouse conditions,this study was conducted to compare whiteflies control with imidacloprid pesticide and nano-silicon fertilizer (NSF) as a quasi-essential element in plant nutrition and their impacts on improving resistance to pest. This research was designed as a pot experiment in a soilless greenhouse with polyethylene (PE) coated and controlled environmental conditions. Flower quality components, including flower diameter, flowering stem height and vase life, relative water content (RWC), vase solution uptake, the total phenol content,leaf Ca++ and Si+ content and whiteflies contamination (egg, nymph, pupa, and adult) were evaluated using a factorial experiment based on completely randomized design with three replications in a hydroponic greenhouse. ResultsThe results showed that NSF caused a significant increase of flower diameter, stem length and vase life of flower compared to imidacloprid. Also, NSF increased total phenol content. Leaf calcium and Si concentration in treatment with NSF showed a significant increase compared to other treatments.In addition, the percentage of whitefly population significantly decreased under applying NSF and imidacloprid compared to the control plants. Spraying imidacloprid despite whitefly control led to the reduction of flower diameter and total phenol content. Also, imidacloprid significantly increased flowering stalk height and the contamination of whitefly nymph with respect to NSF. DiscussionAccording to the results of the present research, foliar application of NSF not only reduced the environmental impact caused by using chemical pesticides but also led to the control of whiteflies population and improved the quality of gerbera cut flower. Therefore, we recommend the use of NSF as a fertilizer-medicine in greenhouse cultivation of gerbera 'Stanza'.

Read More Abstract Background and ObjectivesPomegranate (Punica granatum L.) is a fruit tree that originated from Iran and has been cultivated since ancient times. Classical propagation methods of pomegranate like stem cutting and air-layering have seasonal timing and a higher risk of pathogens contamination. Micropropagation by tissue culture could be considered a method for production of steady and pathogen free plants in this species. However, factors such as lack of rooting and browning due to the oxidation of phenolic components under invitro culture can limit its micropropagation efficiency. To overcome these problems, the effects of various factors, including media, iron-chelating agent, and plant growth regulators, were examined on the pomegranate propagation through in vitro conditions. Materials and MethodsTo obtain sterile explants, stem cuttings of the pomegranate cultivar Shishe Cap Ferdos were treated with different disinfection treatments. Then, the effect of DKW, MS and WPM basal media and two types of iron chelating agents (EDDHA and EDTA) were evaluated to decrease the secretion of phenolic compound from explant and subsequent browning. In the next step, the effect of plant growth regulators treatments, including 1-2 mg/l of BA in combination with 0.2 mg/l of various auxins (IAA, NAA or IBA) on shoot development and proliferation was studied. After that, rooting of regenerated plantlets was explored at WPM medium supplemented with 0.5 or 1 mg/l IAA, IBA or NAA. Finally, pomegranate plantlets were transferred to different soil mixtures, including eat moss and perlite (2:1), cocopeat and perlite (2:1), peat moss and vermiculite (2:1) and cocopeat and vermiculite (2: 1) for acclimation. ResultsThe results indicated that sterilization with 2% sodium hypochlorite for 15 minutes followed by washing with 1g/l carbendazim for 10 minutes, and finally culturing the explants in the medium containing 1 g/l carbendazim had the best effect on reducing explant contamination. Among different basal media and iron chelating agents, WPM resulted in the lowest explant browning. Also, EDDHA was more efficient compared to the EDTA so that explant browning in WPM media containing Fe-EDDHA reduced by 55.8% compared to DKW containing Fe-EDTA. In addition, WPM basal medium supplemented with 2 mg/l BA and 0.2 mg/l IAA induced higher shoot proliferation in node explant of pomegranate. For rooting treatments, after 45 days, WPM supplemented with 1 mg/l IAA induced higher rooting percentage, root length and number in pomegranate shoots. Among different acclimatizing treatments, plantlets that grew on peat moss and perlite (2: 1) had the highest leaf number and stem height. DiscussionOur results suggested that the medium composition and chelating agent affected pomegranate explants browning. EDDHA appears to have a positive effect on reducing browning by chelating copper, which is a phenol oxidase cofactor. Also, EDDHA was more efficient compared to the EDTA.

Optimization of in vitro Callus Induction and Somatic Embryogenesis of Horseradish (Armoracia rusticana) Supplemented with 2,4-D and IAA

Read More Abstract Background and Objectives Horseradish has a male sterility that produces a small amount of seed with a low germination. It then propagates with root cuttings, which is also subject to a limited number of mother rootstocks. Therefore, the mass proliferation of this plant is important through tissue culture, especially somatic embryogenesis protocols. Somatic embryogenesis is a method to develop embryo via plant somatic cells during in vitro culture. The purpose of the present experiment is to find the best method for callus and somatic embryogenesis inducing horseradish, which is carried out in two B5 and NL media in both solid and liquid phases. Materials and Methods This research was carry out in the tissue culture lab of the department of horticultural sciences at gorgan university agricultural sciences and natural resources from 2016 to 2019. Calli are developed on callus induction media (B5 + 1 µM 2,4-D + 2 µM kin, NL + 1 µM IAA + 2 µM kin), and then transferred to somatic embryo induction phase (B5 + 3 µM 2,4-D + 4 µM kin, NL + 3 µM IAA + 4 µM kin) for 8 weeks. Elimination of IAA, 2,4-D and kin from induced calli done onto realization phase, and globular, heart and torpedo embryos observed 4 weeks later. Results Based on the results, the best surface sterilization was achieved by 70% ethanol for 30 seconds and 70% aqueous sodium hypochlorite (v/v) for 60 minutes. The callus and somatic embryo were observed in both solid and liquid B5 and NL media. In the study of callus induction, according to the measurement of appearance quality and the process of callus formation, the growth and increase of callus in solid phase are higher than that of liquid phase. The highest callus induction record is seen in solid B5 medium containing 2 µM Kin and 1 µM 2,4-D. Generally, calli on embryo induction phase are bright green to yellowish and compact consisting of separate clusters of somatic embryonic cells. The results of somatic embryogenesis revealed that globular, heart, torpedo and total embryos in B5 containing 1 µM 2,4-D are more than NL containing 1 µM IAA. Discussion Optimizing the growth of somatic embryos is important for mass propagation of plants that face problems in seed germination or have a long growth period. Due to the success of the production of horseradish somatic embryos, the results of this experiment can provide the possibility of production and propagation of horseradish plants in a short time. Further studies are required to determine the somaclonal variation of horseradish mature somatic embryos and plantlets.

Effect of Salicylic Acid on the Quality Characteristics of Apple Fruit (Golden Delicious and Red Delicious) During Storage Period

Read More Abstract Background and Objectives Apple has a long-term supply throughout the year because of its high storage capacity. Sometimes keeping this fruit in the cold storage room for a long time reduces the quality and marketability, thus making it unattractive for consumers. Recently, there has been an increasing interest in the use of natural compounds to maintain fruit quality and extend its shelf life. Salicylic acid (SA), a widely distributed compound in plants, belongs to a group of phenolic compounds. It could be considered an endogenous plant growth regulator involved in the regulation of physiological processes and disease resistance mechanisms. Materials and Methods To solve this problem, the effect of different concentrations of salicylic acid on the storage of golden and red delicious apple fruits was examined at harvest, 45, 90, 135 and 180 days after storage. Treatments included no washing, distilled water, and salicylic acid 1, 2, and 3 mM for half and 1 hours in cold storage room at 1 ± 2° C. At the end of each storage period, traits such as fruit firmness, vitamin C, TSS/TA, phenol, mealy fruit, flavor (panel test), chilling index and antioxidant capacity were evaluated in three storage periods (0, 90 and 180 days after storage). A factorial experiment based on a completely randomized design was conducted. In both cultivars, harvest index coincided with 170 days after full bloom. Results Results showed that chilling injury, mealy and fruit ripening reduced by salicylic acid treatment and flavor maintained. Antioxidant capacity and phenol content increased by salicylic acid treatment during the storage, but no effect on vitamin C and fruit firmness was observed. During storage, maximum of chilling injury and flavor was related to yellow apple fruit and the maximum of tissue and skin firmness, vitamin C and antioxidant activity belonged to red apple cultivar. The use of 2 mM salicylic acid for half an hour was more effective than other treatments in preserving the quality and storage of fruit of both cultivars. Discussion The results of this study showed that at the end of the storage period the amount of mealy fruit and flavor increased, while skin and tissue firmness decreased significantly. The effects of salicylic acid include a decrease in mealy fruit and chilling injury, as well as an increase in phenolic compounds and maintenance of the antioxidant activity, flavor and TSS/TA, which ultimately increased shelf life of apple fruits. High quality and storage capacity were related to red rather than yellow apple.

Read More Abstract Background and Objectives Gerbera (Gerbera jamesonii L.) is one of the most important cut flowers in the world, and its production is increasing. One of the major problems in cultivating gerbera is its bent neck and short vase life. With the ever-increasing development of flowering industry and the necessity of improving the quantity and quality of flowers, it is necessary to use the beneficial elements in the production of flowers. Calcium is one of the most important nutrient elements that plays a major role in plants. It has a crucial task in the stability of cell wall and cell membrane as well as the development of cells. The balance between cations and anions, the activation of certain enzymes and the regulation of osmotic pressure are among other functions of this element. Calcium, also, plays a role in root development and prevents injuries caused by mechanical and thermal damages such as wind. Nanochelate technology has been able to significantly solve chelate problems. The present study was conducted to investigate the effects of various concentrations of calcium nanochelate on quantitative and qualitative characteristics and vase life of Gerbera. Materials and Methods In this study, three concentrations of calcium nano-chelate (0, 2 and 3 g/l) on three cultivars of gerbera cut flower (Intense, Amlet and Cabana) were examined. Factorial experiment was conducted in a randomized complete blocks design with three replications. In this experiment, morphological, physiological, biochemical parameters and postharvest life of these cultivars were evaluated. The related characteristics of the flower included the dry weight of the scape, the number of inflorescences per plant, the height of the flowering stem, the diameter of the scape and the diameter of the inflorescence. The percentage of electrolyte leakage or membrane stability index, chlorophyll and carotenoids content, calcium content of the plant, vase life and water absorption were investigated. Results The results showed that morphological traits improved with increasing the concentration of calcium nano-chelate. Also, chlorophyll and stomatal conduction increased, but ion leakage decreased. Increasing the concentration of calcium nano-chelate increased the vase-life and water absorption in gerbera flower. The highest number of inflorescence, scape diameter, scape dry weight, inflorescence dry weight and vase life were obtained with 3 g / L of calcium nano-chelate. Discussion In this research, we found that gerbera treatment by calcium nano-chelate was significantly effective. Among the treatments, the concentration of 3 g/l was recognized as the best treatment by increasing the vase life and improving the growth, physiological and biochemical characteristics of gerbera cut flower.

Read More Abstract Background and Objectives Descurainia sophia L. is one of the most valuable medicinal plants in the pharmaceutical industry because its seeds are used for the production of mucilages. Methanol spraying, as a biological stimulant on three carbonaceous plants, can increase carbon dioxide stability and reduce respiration, resulting in more dry matter in these plants. Materials and Methods This experiment was carried out to investigate the effect of methanol spraying on seed yield and mucilage variation of D. sophia based on randomized complete block design with five treatments and three replications in a field experiment in Khoy, Iran. The treatments consisted of the control treatment (distilled water without methanol application) with 4 concentrations of methanol spraying (containing 10, 20, 30 and 40%) carried out during the growing season three times at intervals of 12 days (36, 48, 60 days after planting). After the last spraying at the time of full maturity, the plants in each plot were taken individually. The measured morphophysiological characteristics included plant height, plant leaf number, plant stem number, plant dry weight, number of pods per plant, number of seeds per pod, 1000 seed weight and seed yield. Also, the measured phytochemical traits included seed ash content, mucilage percentage and yield, and inflation index. Results Results showed that foliar application of different concentrations of methanol had a significant effect on number of plant stems, number of plant leaves, plant dry weight, number of pods per plant, number of seeds per pod, grain yield, seed mucilage yield, seed ash, seed inflation index (p ≤ 0.01), plant height and 1000 seed weight (p ≤ 0.05). Foliar application of different concentrations of methanol had a significant effect on all morphological and functional components. To be more precise, methanol spray had a significant effect on plant height, number of stems, plant leaf number, shoot dry weight, number of pods per plant, number of seeds per pod, 1000 seed weight and seed yield, but the effect of year on none of the measured traits was meaningful. Also, the interaction between the year and methanol spraying on the traits of plant leaf number, number of seeds per pod and 1000 seed weight had a significant effect, but the remaining traits were not affected considerably. Combined analysis of variance showed that the year effect had no noticeable effect on seed ash. While methanol concentration had a significant effect (p≤ 0.01) on the seed ash content, the interaction of methanol in different years of experiment was not significant on this characteristic. Discussion Regular methanol spraying, as an active bioavailable and a new method in the production of medicinal plants, improved seed yield and mucilage of Descurainia sophia seed. Generally, the highest values for the evaluated properties were obtained in the induction of 40% volumetric methanol solution.

Read More Abstract Background and Objectives Strawberry is one of the important small fruits worldwide because of its high economical value. Mazandaran is ranked second among Iran’s strawberry prodcing provinces with about 14581 tones of strawberry yearly. This research aimed to evaluate the effect of different silicon sources on the reproductive and vegetative growth of strawberry in an outdoor hydroponic system. Materials and Methods This experiment carried out in a factorial format with two factors consisting of growth media (double medium: 1 cocopeat + 1 perlite, and triple medium: 1 cocopeat + 1 perlite + 1 rice husk) and silicon spray (control, 50 mg/L Si from sodium silicate, 100 mg/L Si from sodium silicate, 50 mg/L Si from potassium silicate, 100 mg/L Si from potassium silicate). It was performed in Sari Agricultural Sciences and Natural Resources University at 2016. Different characteristics including silicon concentration in the leaf, aerial fresh and dry weight, leaf number, fresh and dry weight of crown, fresh and dry weight of root, yield, weight of single fruit, number of the produced fruits in each plant were evaluated. Results Potassium silicate 100 increased silicon concentration of strawberry leaf significantly. Leaf number of the plant and aerial dry weight in triple medium were significantly higher than the double medium. High rate of potassium silicate in the triple medium caused lower crown diameter in comparison with potassium silicate 50 in the same medium. However, in the double medium, the potassium silicate 50 was the best treatment for crown diameter. Both fresh and dry weight of strawberry roots were the highest in the double medium and potassium silicate 50 treatment. Single fruit weight was maximum in the double medium and potassium silicate 100, which was not significantly different from potassium silicate 100 in triple medium. Fruit dry matter in the double medium and potassium silicate 50 treatments was more than other treatments. Fruit size was affected by medium, so that fruit size in the double medium was more than the triple medium significantly. Discussion Sodium toxicity in sodium silicate treatment, low rate of silicon in potassium silicate 50 treatment, and lack of rice husk in the double medium could all negatively affect different physiological aspects of strawberry growth; or at least, they hindered achieving positive results. In summary, it can be concluded that the double medium and potassium silicate 100 can be recommended for higher yield and big strawberry fruit, but the triple medium and potassium silicate are useful treatments for vegetative growth specially for more crown diameter and root growth.

Read More Abstract Background and Objectives Chrysanthemum (Chrysanthemum morifolium R.) is one of the most popular cut flowers in the world. Stem end blockage and water stress are two problems in shortening the vase life of cut Chrysanthemum. Essential oils are noble alternative substitutes for silver and chemical compounds because of their antimicrobial activities and environmentally friendly nature of the extracts. A pomegranate peel extract (PGE) was evaluated as a natural antifungal preparation for the control of postharvest rots. Materials and Methods This study was conducted to investigate the effect of pomegranatepeelextract of three cultivars (‘Shahvar dane sefid’, ‘Rabab dane ghermez’ and ‘Malas Poost Siah Yazdi’) in four concentration of 0, 15, 25 and 50 mg/l and Rosmarinous and Artemisia essential oils in four concentrations of 0, 25, 50 and 75 mg/l on vase life of cut chrysanthemum in a completely randomize design (CRD). Evaluated characteristics were consisted of vase life, stem end bacteria, petals water content, fresh weight loss, electrolyte leakage, protein content and peroxidase enzyme activity. Results According to the results, flowers treated by 15 mg/l pomogrante peel extract of ‘Shahvar dane sefid’ showed the highest effect on vase life of chrysanthemum with 34.10% higher than control. The lowest population of bacterial in stem end of flowers was obtqined by 25 mg/l Rosmarinus essential oil with 70.14% lower than control. By increasing the extract and essential oils concentrations, bacterial population in stem end of cut flower significantly increased, while peroxidase activity significantly decreased. The lowest fresh weight loss was observed in stems treated with 75 mg/l pomegranate peel extract of ‘Shahvar dane sefid’, 15 mg/l Pomogrante Peel Extract of ‘Rabab Daneh Ghermez’, 25 mg/l Artemissia essential oil and 25 and 50 mg/l Rosmarinous essential oil. The highest petals water content (75.66%) was obtained in 75 mg/l Rosmarinus essential oil. The lowest petals water content (44.66%) was obtained in 75 mg/l Artemisia essential oil. The lowest electrolyte leakage was observed in cut stem floweres that were treated by 15 and 25 mg/l pomegranate peel extract cv.’Malas Poost Siah Yazdi’. Discussion Generally, compared to control, all treatments were able to extend vase life and decrease fresh weight and microbial stem end of cut flowers. Also, among three pomegranate peel extracts, extracts of cv. ‘Shahvar dane sefid’ showed the highest effect on extending vase life, decreasing the fresh weight, electrolyte leakage and stem end bacteria, and thus, recommended for extending the vase life of cut flowers.

Read More Abstract Background and Objectives Antioxidants can harness the oxidative stress caused by free radicals by releasing single electrons and hydrogen ions. Myriad plants including vegetables and fruits express enzymatic and non-enzymatic antioxidant properties which are used by humans to maintain health.. In this study we aim to compare the level of antioxidants and the amount of total phenolic and flavonoid compounds at three different stages of fruit ripening including immature, semi-ripen and fully matured fruits in two species of raspberry after a period of storage at freezer. Materials and Methods Fruits of Rubus persicus and R. caesius were collected at different maturity stages including immature, semi-ripen and fully matured stages from natural habitats. The extracts of fruits were stored at -20 °C for 6 months. The antioxidant activities of extracts were determined using DPPH free radical purification and ferric iron regeneration ability (FRAP) methods in June, 2013 in Research Laboratory of Science and Technology Park of East Azerbaijan province. The content of phenolic and flavonoid compounds was also quantified according to Folin–Ciocalteu’s phenol and aluminum chloride methods, respectively. The antioxidant activity and total phenolic and flavonoid contents were compared between species at three maturity stages in a completely randomized factorial design with three replicates. Results Both methods applied to examine the antioxidant activity revealed a high mean value in extracts from fully matured fruits compared with immature and semi-ripen fruits. The magnitude of antioxidant activity was significantly higher in R. caesius than R. persicus except that the amount measured by DPPH free radical purification level was similar between species at only fully matured stage. A significant difference was observed in both total phenolic and flavonoid contents between immature, semi-ripen and fully matured stages in both species (P < 0.05) except that the total flavonoid content was similar between semi-ripen and fully matured fruits in R. caesius. Discussion Results showed that the highest antioxidant capacity was found in the fully matured stage of fruits in both species and thus the fully matured fruits might be capable of counteracting and even neutralizing free radicals. The results also showed that different species had different level of antioxidant activity. The level of total phenolic and flavonoid content at fully matured fruits was also higher than immature fruits in both species. Our results suggest that the antioxidant potential of raspberry can have several medicinal applications and play an important role by reducing the oxidative stress.

In Vitro Regeneration of Ornamental Crocus (Crocus vernus L.) by Using Plant Growth Regulators

Read More Abstract Background and Objectives Crocus flower, which belongs to Iridaceae family, is one of the most important ornamental bulbous plants. The propagation of Crocus vernus L. by corm is not commercially affordable due to transmission of fungal and bacterial diseases as well as the long dormancy period which takes 4 to 5 months. Thus, in vitro culture of Crocus vernus is a suitable solution for commercial reproduction. Plant tissue culture technique would be a better alternative for improving quality and production. Materials and Methods In order to micropropagate ornamental saffron, the effect of different culture media on direct organogenesis experiment was conducted in factorial arrangement in a completely randomized design with 3 replications in the tissue culture lab of the Department of Horticulture at Agricultural Sciences and Natural Resources University of Khuzestan from 2015 to 2016. The explants used in this experiment were latheral and terminal buds which were evaluated in 10 hormons treatments and control in the Murashige and Skoog medium (MS). To eliminate surface contamination, the explants were first immersed under water for 30 minutes after placing the corms. Next, they were placed in high temperature Benlate fungicide solution (55° C) for 30 minutes. Then, they were put in 70% ethanol (ethyl alcohol) for 30 seconds and 15% Sodium hypochlorite solution for 6 minutes. The explants were finally washed three times with sterile distilled water under the air flow bench. The effects of plant growth regulators including BAP (0.5, 1.0, 2.0 mg/l), TDZ (0.5, 0.75, 1.0 mg/l), Kin (0.5, 1.0, 2.0 mg/l) and 0.1mg/l IBA in direct regeneration were investigated. Results The results showed that the kind of plant growth regulators (PGR) and the explant type are very important in regeneration of Crocus vernus so that we can not secure any shoots without PGR. In direct organogenesis experiment, the maximum number of multiple shoots (15.84) was obtained from apical bud explant in MS medium supplemented with 5/0 mg/l TDZ along with 5/0 mg/l BAP, 5/0 mg/l KIN and 1/0 mg/l IBA, after 10 weeks, that had a significant difference with other treatments at 1% Duncan test. We observed 2.o mg/l BAP along with 1/0 mg/l IBA had the greatest effect on the number of corms. Discussion The results showed the shoot regeneration is controlled by the ratio of cytokinin with auxin. It should be mentioned that cytokinin plays a role in the synthesis of RNA, stimulation of the production of protein, and the activity of some enzymes. The results also indicated that the use of cytokinin along with auxin has a beneficial effect on shoot regeneration.

Read More Abstract Background and Objectives To achieve optimal fruit set, economic productivity and targeted crossings in seedlings and grafted walnut orchards, the flowering time of both male and female flowers and their overlapping is important. Since the walnut ovule longevity is limited, fertilization should occur at the shortest possible periods to lead to fruit set. Therefore, the evaluation of effective pollination periods and the needed time for the growth of pollen tube and ovule fertilization are important for successful fertilization. The aim of this study was to investigate the compatibility and the effect of pollen on some selected genotypes of walnut as pollinizers on the fruit set, fruit composition and the quantitative and qualitative characteristics of the progeny derived from MSG15, MKG23 and MKG24 genotypes as maternal parents. Materials and Methods This research was conducted in 2016-2017 in the research garden of Malayer University in Hamedan province. In this study, female parents including MSG15, MKG23 andMKG24 were pollinated with pollinizers of MKG4, MKG5, MKG10, MKG13, MKG14, MSG15, MKG23 and MKG24. Some of the characteristics of the fruit and the kernel were also evaluated in the offspring resulting from controlled pollination. Results Based on the results, the cross between female parent genotype of MSG15 with pollinizers of MKG4 (%79.33), MKG5 (%81.67) and MSG15 (%79.67), female parent genotype of MKG23 with pollinizers genotypes of MKG5 (%83.67) and MKG10 (%62.67), female parent of MKG24 with pollinizer genotypes of MKG5 (%81.67) and MKG14 (%81.33) showed the highest fruit set. Also, female parent of MKG15 with pollinizers of MKG13 (%38.33), MKG23 (%37.33) and MKG24 (%38.33), female parent of MKG23 with pollinizer MKG24 (%40.00), female parent of MKG24 with pollinizers MKG10 (%42.33) and MKG23 (%42.33)showed the maximum flower abscission. Moreover, the highest percentage of kernel (%61.9) was found in combination cross of female parent of MSG15 with MKG5 pollinizers. The lowest fruit weight with husk (250 gr) and kernel fresh weight (50.5 gr) was found in combination cross of female parent MKG23 with pollinizer of MKG24. In addition, the lowest percentage of kernel (%28.98) was totally obtained in combination cross of female parent MSG15 with pollinizer of MKG24. Discussion The highest percentage of fruit was obtained in hybridization of maternal parent MSG15 with pollinizers MK4, MKG5, MSG15, and MKG23 as seed parents with pollinizers MKG5, MKG10, and MKG23 mother's parents with paternal parents MKG5 and MKG10. According to the results of this research, it is possible to use compatible and suitable genotypes introduced in this study in other walnut breeding programs.

Read More Abstract Background and Objectives Culture medium and growth regulators type can influence the growth traits and for determining the most effective medium and hormone compositions an extensive studies are needed; otherwise the culture medium may have a negative effect on the growth traits. In this research the effects of two culture media (B5 and MS) supplemented with two growth regulators (IBA and BAP) with different concentrations (0.5, 0.9, and 1.5 for IBA and 0.4, 0.8, and 1.2 mg/l for BAP) alone or in combination was evaluated. Studied traits were number of leaf per shoot, shoot length, number of regenerated shoots based on completely random plan with 5 replications. Materials and Methods Explants provided in the beginning of spring from mother plants growing in open environment. Shoot tips and axillary buds (0.3-0.4 cm long) used as the explants. After washing with tap water, explants were immersed in ethylic alcohol for few seconds (3-4 seconds) then immersed in mercuric chloride (0.005 W/V) for 5 minutes which supplemented with few drops of tween 20 and finally washed three times with sterile distilled water. Disinfected explants cultured on medium under air laminar flow bench. Growth chamber was adjusted to 16 hours light and 25 to 27ᵒC. Results The results of variance analysis demonstrated significant differences between media, hormone level and interaction between hormone concentrations for traits studied. The results showed the longest shoot (6.39 cm) and the highest shoot number (13.65) obtained in MS medium supplemented with 0.8 and 0.4 mg/l BAP respectively. In this experiment the highest root number (8.41) and root length (14.75 cm) was observed in B5 and MS medium containing 0.9 and 1.5 mg/l, IBA. Discussion Overall the most shoot proliferation of this rootstock occurred in MS medium. New shoots in MS medium showed normal and better growth comparing to shoots obtained in B5 medium, that can be one of the results of higher amount of nitrogen (ammonium nitrite) in MS medium. In B5 medium with all hormone treatments weak growth observed for this plant. In fact this medium was made for herbaceous plants and this is one of the main reasons why penta rootstock did not showed good growth symptoms on this medium.
