Read More Abstract Background and Objectives The best way to increase the tolerance of plants against stresses is to know the specific characteristics of the cultivars, which is needed both for planting in different climatic conditions and for breeding programs of these cultivars. For this reason, the exact study of the physiological traits of native cultivars of each region in the various stages of growing of grape is necessary. Materials and Methods Sampling was done in five times. During the dormancy season (Winter 2013) three times samplings of stems with buds were done and biochemical parameters of proline, electrolyte leakage and soluble sugar were measured. The aim of this study was to investigate some of the effective parameters of cold tolerance in five cultivars of grapevine including “Bidaneh Sefid”, “Bidaneh Ghermez”, “Lal”, “Sahebi” and “Fakhri” in a randomized complete block design (RBCD) with three replications. In the growing season (spring 2013) two times samplings were collected from the leaves of cultivars and biochemical parameters such as proline, malondialdehyde (MDA), soluble sugars, photosynthesis pigments, total protein, activity of antioxidant peroxidase, polyphenol oxidase and ascorbate peroxidase enzymes were measured. Results Result showed that there were significant differences between the cultivars in both winter and spring and in all considered characteristics. The winter cultivar of 'Fakhri' with minimal electrolyte leakage in all three times of sampling (33.12 µSiemens/gr Fw in January, 23.17 µSiemens/gr Fw in February and12.89 µSiemens/gr Fw in March) was more prepared to tolerate environmental stresses than other cultivars. In the spring, in both sampling times of spring season, cultivar of “Bidaneh Ghermez” with the least amount of malondialdehyde (3.7 µmol/gr FW in late April and 4 µmol/gr FW in early May) and the highest proline (10.27 µmol/gr FW late April and 7.39 µmol/gr FW in early May), and protein (respectively 0.26 mgr/gr FW late April and 0.23 mgr/gr FW in early May) were more prepared to tolerate environmental stresses than other cultivars that were examined in this study. The cultivar of “Lal” with the highest level of malondialdehyde (9.52 µmol/gr FW in late April and 4.52 µmol/gr FW in early May) showed lower relative stress endurance compared with other studied cultivars. In the winter, cultivar of 'Fakhri' had higher stress forbearance than other cultivars. Also, in the spring, cultivar of “Bidaneh Ghermez” had more and “Lal” had the lowest stress endurance than other cultivars that were examined in this study. Discussion Considering all the traits studied, it can be stated that in winter tolerance of seeded cultivars is more suitable than seedless cultivars. Seedless cultivars are more sensitive to temperature variations and generate severe reactions.

Normally about one week after harvesting, a considerable loss occurs in picked fruits. Plant nutrition, during growth and developmental stages influence the quality of many fruits. Calcium is one of the most important elements that can affect quantity, quality, storability and market of many horticultural crops. Although South Khorasan province is located in arid areas, it has lime soils with enough calcium for plant growth and development. However, due to low mobility of calcium from soil to plant, calcium deficiency is common in horticultural plants. Thus, pre-harvest spray with minerals like calcium salts can be useful for reducing postharvest losses during handling and storage. Therefore, the aim of this research was to evaluate the effect of different calcium salts spray on physicochemical properties and quality of fresh jujube fruit. Materials and Methods Twenty jujube trees, eight years old, were sprayed with calcium salts twice, mid of June and early in July 2016 in a commercial orchard in Birjand, South Khorasan province. Experimental treatments were distilled water (control), calcium chloride (0.5 and 1%) and calcium nitrate (0.5 and 1%). Sprays were done at two stages of fruit growth and development, cell division (3 mm fruit diameter), and 20 days later in full cell enlargement stage. Fresh fruit was harvested at a crisp mature (white-red) stage early in August. The experiment was conducted in a completely randomised block design with four replications, and each replicate included 500 g of fresh fruit. Data were analysed using SAS program (ver. 8) and comparison of means was made with LSD test at 1 and 5% probability. Results Physiochemical properties of harvested fruit including fresh fruit weight, dry matter, fruit length and diameter, firmness, total soluble solids, pH, carotenoids, total phenol, and calcium content were evaluated. The results showed that calcium nitrate application at 0.5% significantly increased fruit fresh weight, dry matter and fruit length and diameter compared to control. Also, fruits that were sprayed with calcium chloride at 0.5% concentration had higher firmness, carotenoids and total phenol. The highest total soluble solids and calcium content values were obtained in treated fruit with calcium chloride (1%). Although different calcium salts spray had no significant effect on juice pH. Discussion In general, it can be concluded that physical fruit parameters were affected by calcium nitrate, while calcium chloride spray had a greater impact on chemical and nutritional quality aspects of fruit. Although calcium chloride at 0.5% was more effective on chemical factors, however, it was not significantly different with calcium chloride 1% treatment. Among different treatments, calcium nitrate at 0.5% had the highest effect on physical properties of jujube fruit. Therefore, it can be concluded that spraying of jujube fruit trees during fruit growth and developmental stages with both calcium salts at 0.5% concentration have beneficial effects as they improve the most important physical and chemical parameters of fresh jujube fruit.

Read More Abstract Background and Objectives Sprouting is one of the main reasons of wasting about one half of potatoes during the storage. The existing methods to control sprouting in long term storage (for example postharvest applications of chlorpropham) can do the job as well, but the incidence of quality problems and undesirable side effects in such potatoes, and more importantly, the growing interest in organic food products, have challenged the application of these methods. Materials and Methods The peppermint extracts used in the experiment were obtained from dried peppermint leaves in 48 hours, at room temperature using 1:1 distilled water-alcohol solution, and with the normal air pressure. The peppermint extracts were dried for 10 days and then transferred to a dark glass bottle by means of petroleum ether (as solvent) and were kept at 2°C until being used for later use. Herbal extracts of peppermint were used at concentrations of 0, 0.5, 1, 2 and 4 g/l, with regular intervals of 2, 4 or 6 weeks during the 5 months of potato (Agria cultivar) storage while sprouting was monitored. Direct contact between tubers and the peppermint extracts was not allowed. To provide the needed oxygen for the tubers respiration, the containers apertures were opened for 15-20 minutes every four weeks and were sealed again. A group of tubers was only treated by 37g chlorpropham (5% purity). To do this, the powder was poured into the container, completely sealed and slowly shaken for 10 minutes. The sealed container was kept at room temperature for 48 hours. All experiments were repeated 3 times. Data was analyzed by a completely randomized design (CRD) with a factorial experiment. Results According to the results, by increasing the concentration of mint from 0.5 to 2 g/l, sprouting potato was controlled more effectively and meaningfully. More concentration in the control of potato sprouting was not significant. Use of this extract at a concentration of 0.5 g/l stimulates the germination of tubers compared to the control. Extensive sprouting of tubers was delayed for up to 4 months compared to the control treatment too. Application of peppermint extract caused %92 and %96 decrease in potato germination and total weight sprouting respectively. Organoleptic evaluation of the boiled and fried potato samples, treated with the mint extract and chlorpropham, did not show significant differences. Discussion Use of 2 g/l mint extract will be able to decrease waste potato tubers in storage up to 6 percent. Replication of every 4 weeks extracts application is absolutely essential for the best result. Organoleptic evaluation of the boiled and fried potato samples, treated with the mint extract and chlorpropham, received more points compared to control.

Read More Abstract Background and Objectives Olive (Olea europaea L.) belongs to Oleaceae family. Olive fruit has been used for oil and table olive. Olive is one of the most important fruit trees around the world. Iran is one of the regions of the world with different wild olive genotypes distributed in different provinces. Olive genotypes are important genetic resources for Iran. These genotypes can be selected as promising genotypes and used in breeding programs. The objective of this study was to evaluate adaptability of 8 olive genotypes from Gorgan and Taroum. Materials and Methods This experiment was conducted to evaluate adaptation ability of 8 olive genotypes in Tarrom olive research station during 2007- 2011. Trees were planted under a complete randomized block design with a 6 × 6 distance. Vegetative and reproductive characters of olive trees were measured. The collected data were analyzed with MSTATC and the means were compared with Duncan's multiple test. Results Vegetative results showed that tree height, wide, shoot season growth and trunk cross sectional areas were significantly different at 1% between olive genotypes in Tarrom olive station. Inflorescence number per shoot, flower number per inflorescence, perfect flower percent, and final fruit set were significantly different at 1% statistical level. Fruit weight was significant between olive genotypes and it varied between 2.3 and 9.8g. G4 genotype had the biggest fruit and flesh weight. Fruit yields were significant between genotypes so that T2 and B1 had the highest and lowest yields respectively. The oil percent in dry matter was significantly different between genotypes. All genotypes had higher than 40% oil in dry matter. The highest oil percentage (52%) was recorded in G4 genotype. Generally, G4 olive genotype could be a promising genotype due to the large fruit (more than 9 grams), relatively high yield (about one ton) and high oil content (more than 52% of the dry matter). Discussion Wild olive genotypes are very important gene pole for any purposes in each county. Iran has different wild olive genotypes in different regions like Golestan, Gilan, Kerman, Kermanshah, Ilam and so on. Identification and evaluation of adaptability of local olive genotypes are very important for new olive cultivar selection and breeding programs. As mentioned above, the evaluated olive genotypes show different responses in Taroum olive research station. Different vegetative and reproductive characteristics were recorded on evaluated olive genotypes. Generally, results showed that G4 olive genotype was superior based on fruit traits, oil and yield.

Read More Abstract Background and Objectives Management of organic fertilizers utilization is of the main components of organic crops production which can greatly affect the yield and quality of plants. Due to the short time course of leafy vegetables production, farmers commonly use nitrogenous chemical fertilizers for the nourishment of plants. The results are huge nitrate accumulation on the edible parts of the vegetables and hence the deteriorating health side effects. So, owing to the necessity of vegetables in the daily diet, resorting to the production methods to reduce the nitrate accumulation and even preserve and add up the nutritious quality of vegetables is of great interest. Materials and Methods This experiment was conducted to evaluate the effects of deferent levels of cow, vermicompost and poultry organic fertilizers (0, 10, 20 and 30% in volume) and urea (102, 204 and 306 g Kg/ha) on nitrate accumulation (morning and evening sampling time) and Fe2+, Zn2+ and potassium concentration and total phenolic content as well as on some growth characteristics of spinach as RBCD design with three replications in Azarbaijan Shahid Madani University during 2016. Results The results revealed that the highest morning nitrate accumulation (2435 mg Kg-1 Fwt) and that of evening (2052 mg Kg-1 Fwt) belonged to 306 kg/ha urea treatment. Leaf length was effected by 20% (3.6 cm) and 30% (8.8 cm) of poultry manure. The highest data for TSS and root fresh and dry weight were attained by 10 and 20% of cow manure. The treatments, 20 and 30% of vermicompost, cow manure and 30% poultry manure increased the leaves fresh and dry weight. All levels of poultry and vermicompost manure had positive effects on potassium content. The highest amounts of Fe2+, Zn2+ and total phenolic content were recorded in 10, 20 and 30% cow manure and vermicompost manure Discussion Nitrate accumulation in vegetables is dependent upon several intrinsic and extrinsic factors of which, mineral nutrition is of crucial importance. Considering the positive response of spinach to the application of organic fertilizers, it seems that using these organic fertilizers would greatly decline the chemical fertilizers input as well as reduces the environmental side effects of chemicals and virtually goes to the production of safe organic products under sustainable production systems.

Read More Abstract Background and Objectives Taxanes are a family of diterpene alkaloids which is currently known as the most important anticancer compounds to suppress and inhibit cell growth, differentiation and proliferation in indefinitely known cancer cell lines. The most well recognized member of the taxane family is taxol (paclitaxel), a minor cytotoxic component derived originally from yew bark (Taxus brevifolia). 10-Deacetylbaccatin III (10-DAB), which is a homolog of taxol, can be extracted at high yields from the needles of Taxus baccata and is currently considered the most appropriate precursor for semi-synthesizing taxol and its analogue, taxotere (docetaxel). The natural source of both compounds is the inner bark and needles of several Taxus species, especially T. baccata and T. brevifolia. Plant cell culture is an environmentally sustainable source of taxol and offers several advantages as it is not subjected to weather, season or contamination. The aim of the present study was to evaluate taxol and 10-DAB content of two Taxus species and their ability in production of both taxanes through cell culture. Materials and Methods The plant materials of T. baccata and T. berevifolia were collected from Shahid Beheshti University (35° 48' N, 51° 23' E at an altitude of 1785 m) and botanical garden of Faculty of Agriculture, University of Tehran, Karaj (35° 48' N, 50° 59' E at an altitude of 1238 m), respectively. Callus culture of both species was initiated from internodal segments on B5 medium supplemented with plant growth reaulators. Cell suspension culture was established on the medium supplemented with 2.0 mg L−1 naphtalene acetic acid (NAA) and 0.1 mg L−1 benzyl amino purine (BAP). Cell viability was determined by Evan’s blue staining test. After 21 days, the amount of taxol and 10-DAB was analysed by HPLC. Results The results showed that mother stocks of T. baccata and T. brevifolia contain 45 and 27 µg/g DW of taxol and 40 and 4 µg/g DW of 10-DAB, respectively. Cell growth was estimated by measuring fresh weight (FW) and dry weight (DW). Results showed that the maximum growth was reached after 21 days of cultivation. Cell cultures of T. baccata and T. brevifolia produced 540 and 20 µg/l taxol and 1 and 110 µg/l of 10-DAB, respectively. Taxol production in T. baccata cell culture was 26-fold higher than in T. berevifolia cell culture. Discussion As far as our literature survey could ascertain, growth and taxol production in T. baccata cell cultures have previously been reported. Although, T. baccata cells are more potent for the production of taxol, T. brevifolia cell culture can be considered for the production of 10-DAB, a homolog of taxol, as an appropriate precursor for semi-synthesizing taxol and its analogue, taxotere (docetaxel). However, it is essential to study and quantify the effect of selected key medium components on growth as well as product accumulation and strike a balance between the two to enhance the yield and productivity.

Read More Abstract Background and Objectives Iris spuria subsp. Musulmanica is a rhizomatous herb growing as wild in Iran, Turkey and, Caucasia. This species is important from ornamental and medicinal perspective and could be used as flowering pot plant, cut flower and landscape plant. A few of Iris seeds germinate under natural conditions. Therefore, an experiment was designed to improve germination of Iris seeds using different treatments that could be accessible for floriculture purpose. Materials and Methods This experiment was carried out based on completely randomized design with 10 treatments and 4 replications (each replication contains 25 seeds). Treatments were control without treatment, priming in hydroxide sodium 20M for 10 minutes, priming in sodium hydroxide 15M for 20 minutes, W100-10: in water 100°C for 10 minutes, W80-15: in water 80°C for 15 minutes, priming in sulfuric acid 98% for 10 minutes, priming in sulfuric acid 98% for 15 minutes, stratification for 30 days, stratification for 60 days, and stratification for 90 days. For all treatments, germination variables including germination, germination rate, mean germination time, seed vigor index, mean germination daily, shoot length, root length, shoot fresh weight, root fresh weight, shoot dry weight, and root dry weight were measured. Results Treatment effect on germination variables was significant (p<0.05). The results improvement of that treating seeds with 20 mM of hydroxide sodium solution for 10 minutes led to improve the seed germination parameters like germination percentage, germination rate, seed vigor index, shoot and root length, fresh and dry weight of root and shoot compared to control. In three levels of stratification treatments, germination percentages were less than control treatment. Also, the results showed that treating seeds with sulfuric acid for 10 minutes improved as partially seed germination and other germination variables in comparison with control treatment. Discussion Previous studies revealed that the seed coating played an important role in seed dormancy of Iris species. In this study, scarification treatment with hydroxide sodium resulted in increased germination which may correspond to the reduced seed mechanical inhibitory and the improvement in gas and water exchange. Moreover, sodium hydroxide solution facilitated the seed germination via the elimination of seed coat and increasing the seed capsule Intercellular space which finally promoted the exchange of materials between seed and its surrounded environment. Like too many Iris species, a non-deep morphophysiological mechanism may play the most important role in dormancy of I. spuria subsp. Musulmanica.

Breeding programs in eggplant focus mainly on fruit quality. Production of commercial triploid cultivars and emission of seeds in fruits may increase acceptability by consumer and increase fruit quality. Induced tetraploidy and cross between di and tetraploid parental line is a conventional method used in different fruits and vegetables like watermelon. The current study aim to find how may induced tetraploidy affect pollen grain quality, its fertility, seed and transplant vigor in different eggplant cultivars. Materials and Methods Aiming to investigate autotetraploidy in four qultivars of Solanum melongena L., Keshtzar, Yalda, Chahboland and Passargad on fertility, number of days till fruit-set, germination rate and percentage and successful transplanting percentage, a survey was done in a completely randomized design. Tetraploid plants were derived from seed maceration/shoot tip treatment (0.5, 1 and 2 percent of colchicine). To ensure ploidy level, chromosome counting of root tips and flow-cytometry was done during the experiment. To quantify germination percentage and rate, 15 seeds of each cultivar in each ploidy level were placed in petri-dishes in three replications. The seeds of each ploidy form were sown in seedling trays and after seedling period, plugs were transplanted in field for evaluation of characteristics in different ploidy levels. Growth and development rate of plant in seedling stage observed since seeds were cultivated. As soon as hypocotyl appeared on the surface, they were counted. Then seedling emergence and rate were calculated. To test viability of pollen grains, they were stained using acetocarmen. In order to find best parental lines, 18 plants from each cultivar and in each ploidy level were cultivated in the greenhouse condition in three replications. Then all plants were crossed competley (direct and reciprocal). Di and tetraploid plants were tested to find out which one would be better as which parent. Results Number of days till fruit-set, characteristics of seed and seedling and growth rate in seedling stage were significantly (P≤5%) influenced by changes on levels of poloidy. Pollen grain staining with acetocarmine for evaluation of fertility percentage in pollen grains showed that with an increase in ploidy level, fertility may significantly increase (P≤5%). In an effort to triploid plant production, none of the crosses between di and tetraploid plant could bear fruit. Discussion With the overall results, despite slower growth, can be tetraploid plant, due to improved germination and seedling emergence, survival and reproductive capabilities, higher transplanting success in this category of plants, induced increase in the number of chromosomal bundles can be considered as a good way to create genetic variation and prepare parental lines with desirable traits for use in the breeding programs.

Read More Abstract Background and Objectives Recently, the use of cut flowers has increased, and some of these cut flowers (Lily, Rose etc.) are among the most used for decorative purposes. With this boom in popularity has also come a rise in the need for commercial use, and very little academic literature exists on how to best utilize the preservative solutions (here 8-HQC and essential oils) to prolong cut flowers vase life keeping their full quality. The vase life of Lily is usually short due to vascular occlusion. The use of synthetic chemical fungicides to protect cut flowers raises environmental and human health concerns because of chemical residue routinely found on food meant for consumption. Thus, biocides showing insignificant toxicity to human and organisms are more desirable and less likely to cause negative environmental effects. Materials and Methods The chemical composition of essential oils extracted from the endemic Iranian plants Satureja hortensis L., Thymus vulgaris L. and Citrus limon L. was studied in order to evaluate their efficacy as a substitute for synthetic chemical that was used as preservative-solutions. The effects of essential oils (250 mg L-1) individually and 8-hydroxyquinoline citrate (8-HQC) with sucrose 3% were evaluated. Cut flowers were pulsed for 12 h in different preservative solutions, followed by storage in dry and wet conditions (in distilled water) at 8˚C for 15 days, and then all treated cut flowers were kept in room temperature to evaluate commercial vase life. At the end of commercial vase life, parameters such as relative fresh weight, amount of chlorophyll, phenol, anthocyanin, protein and carbohydrate, vase life and antioxidant enzyme activities were measured. In this experiment, flowering stems were arranged using a completely randomized design. Results Limonene was identified as major component of C. limon L. essential oil, whereas Carvacrol was the major component in S. hortensis L. and Th. vulgaris L. Results showed that applying all essential oil treatments or 8-HQC increased positive characteristics and vase life compared with control. However, among all these treatments, the flowers treated with Thyme essential oil plus 3% sucrose or 8-HQC were kept in wet storage condition showed best vase life. Based on our results, new antimicrobial agents such as limonene, thymol and carvacrol in combination with 3% sucrose had a positive effect on the vase-life and relative fresh weight. The senescence of cut flowers is associated with a series of highly regulated physiological and biochemical processes, including breakage of water balance, degradation of photosynthetic pigment, decrease in metabolic constituents, and loss of membrane integrity. In our experiment, Thyme essential and 8-HQC oil treatments were reduced degradation of leaf protein and chlorophyll content which was seen in the controls. Essential oils derived from plants such as T. vulgaris are particularly valuable because of their antibacterial and antioxidant properties. Discussion In conclusion, using various concentrations of essential oils and 8-HQC in preservative solutions showed promising prospects for the utilization of natural essential oils or plant extracts in extending lily vase-life. The proposed mechanism of action, e.g. inhibition of the growth of bacteria in the vase water or inside the xylem vessels of the flower stem needs further elucidation.

Read More Abstract Background and Objectives In order to introduce the best method of planting between Cretan and furrow and also spraying nanoparticles of calcium on the quality and quantity of Gladiolus flowers (Gladiolus hybrida cv. Magma), a study was conducted at the research farm. Nanoparticles release nutrients slowly and continuously because of having nanomembranes so that we can adjust the speed of releasing nutrients from the capsuls. Using this fact, the efficiency of nanoparticles would improve. Cretan method is the most common method of planting of Gladiolus in Iran. Materials and Methods Corms were planted in two ways; Cretan and furrow planting. The density of them was 24 per square meter (four rows with the distance of 15 cm). Nanoparticles of calcium at three levels (0, 1 mg/litre, and 2 mg/litre) were sprayed on the plants before the harvest once a week for three weeks. Considering the importance of planting methods and performance of nanoparticles of calcium, a factorial experiment was conducted based on randomized complete block design with three replications. Data were analyzed using SAS software. Results In this study after the harvesting, most of the features of flowers were measured including: the inflorescence length, the inflorescence weight, the weight index of cut flower (fresh weight of the cut flower/ the length of the cut flower), the shoot diameter, the shoot length, the diameter of the first and the second floret, the number of florets, the number of corm, the corm weight, a, b, and the total chlorophyll, the relative water content, and the vase life. Significant differences were observed at the 5% level for all of the traits except the weight index of the cut flower and the relative water content. The cut flower weight, the cut flower length, the inflorescence length, the florets diameter, the number of florets, the number of corm, and the vase life were increased in the furrow planting method. The importance of the nanoparticles of calcium treatments on vase life was significant. The Number of corms was also significant in the furrow planting method. Discussion The results of this study showed that the quality, the yield and the vase life in furrow planting were better than those in the Cretan. The foliar feeding calcium nanoparticles resulted in survival improvement in post-harvest flowers. In Khuzestan, which is capable of producing commercial Gladiolus flowers in autumn and winter, the furrow planting can be used to obtain flowers with higher quality and quantity instead of using the cretan planting.

So, reducing of respiration rate and delaying of senescence process are required for increasing of storage life. Gamma amino butyric acid (GABA) is a non-protein amino acid with low content in the cell in normal state. No study has taken into account the effect of GABA on the storage life of sweet cherry fruit and also, increasing of shelf life by natural compounds seems essential to maintain the quality. Therefore, this study was conducted with the aim of improving the post-harvest quality of sweet cherry fruit, increasing biochemical and antioxidant properties and improving fruit appearance using GABA with the purpose of replacing it with synthetic chemicals. Materials and Methods Fruits of sweet cherry cv. Tak Daneyeh Mashhad were harvested at commercial maturity from a commercial orchard in Urmia (Iran) and transported to laboratory immediately. GABA was used in three concentrations (5, 10 and 20 mM). Titration method was used for determination of Titratable acid (TA) and pH was determined by pH meter. TSS was determined by refractometer. Total antioxidant capacity was determined by ferric ions reducing antioxidant power assay (FRAP). Total phenolic content was determined by Folin ciocalteu method and total flavonoid content was determined by the aluminum chloride colorimetric method. Results The results showed that TA content remains constant with treatment of GABA during storage. GABA treatment leads to an increase in total antioxidant activity, total phenol, total flavonoid and activity of catalase and guaiacol peroxidase enzymes in the treated fruits compared to control fruit after 30 days of storage. The pH value in treated fruits compared to untreated fruits was decreased during storage. Also, GABA at 10 mM level compared with other concentrations had the highest effect on total antioxidant capacity after 30 days of storage. Discussion The increasing of organic acids in treating fruits can be probably due to increasing of antioxidant capacity by GABA treatment, hence the damage to membrane was prevented and the organic acids were used less frequently. Treatment of fruits with GABA decreased the pH during storage, which may be due to its role in reducing of respiration rate in harvested fruits. The mechanism of the effect of GABA on increasing of total phenol and flavonoid content may be due to stimulating the production of Phenylalanine ammonia lyase (PAL) which can trigger phenylpropanoid pathway and hereby, syntheses of phenolic compounds such as flavonoids compounds were occurred. Catalase is one of the key enzymes to protect the cells and cause the conversion of hydrogen peroxide to water and oxygen. Therefore, probably the GABA treatment via the increasing the antioxidant system such as catalase leads to scavenge free radicals thereby protecting plants against oxidative stress.

Read More Abstract Background and Objectives In recent decades, global warming causes climate changes which triggers physiological and nutrition disorders in plants. Iran is a main pistachio producer in the world. Pistachio cultivar is mainly grafted into seedling rootstocks are which obtained from especial cultivars and wild type pistachio. The changes in the vegetative growth and yield in pistachio orchards have been obviously observed in the years with so warm or so cold weather conditions. The purpose of this research was evaluating the effects of temperature on the growth of three pistachio rootstocks. Materials and Methods In order to study the effect of temperature on the growth and physiological characteristics of three commercial pistachio rootstocks, a factorial experiment on the basis of a completely randomized design (CRD) was carried out with three replications. The first factor was three rootstocks including: Pistacia vera cv. Badamie-zarand, P. vera cv. Ghazvini and P. vera var. Sarakhsi and the second factor was four temperature conditions including: 10, 15, 20 and 25 degrees Celsius. Pistachio seedlings after planting up to 15-leaf stage were kept in the greenhouse condition and then the pots were transferred to growth chambers which adjusted to defined temperature treatments. The plants were kept in temperature treatments for fifteen days. At the end of experiment, growth parameters including fresh and dry leaf weight, fresh and dry weight in shoot and root and leaf area and physiological parameters including stomatal conductance, relative water content of leaves and leaf chlorophyll index were measured. The statistical analysis was done using SAS 9.1 software and mean comparison was done by using the lowest significant difference test (LSD). Results The results showed that with increasing environmental temperature, an increase in the number of leaves, stem height, dry weight of leaf, stem and root, leaf area was observed in pistachio rootstocks. There were significant differences among rootstocks in most of the growth parameters. Badamie-Zarand was more vigorous than the other rootstocks in low temperature. The highest SPAD index was observed in high temperature environment (20, 25°C) beside lowest SPAD index level was observed at Low temperature environmental (10, 15°C). Minimum and maximum stomatal conductance was respectively obtained at 10 and 25 °C in Sarakhsi rootstocks which reveals the sensitivity of this rootstock to temperature fluctuations. In contrast, stomatal conductance variations in the Ghazvini rootstocks are lower than the other rootstocks when exposed to different environmental temperature. This may show the tolerance of this variety to changes of environmental temperature conditions. The leaf RWC in three pistachio rootstocks at different ambient temperatures shows a significant difference. At 15 and 20°C, Ghazvini rootstock has the highest RWC and Badamie- Zarand rootstock has the lowest levels of RWC. The highest relative water content was obtained at 10°C in Sarakhsi rootstock and at 25°C in Badamie-Zarand rootstock. Positive correlation between root dry weight and stomatal conductance reflects the rootstock compatibility with ecological conditions. Discussion Badami Zarand rootstock has more vegetative growth in low temperature environment and more adaptation to a wide range of temperature in comporison to two others rootstock and also can be recommended to nursery as a rootstock for planting.

Read More Abstract Background and Objectives Tomato is one of the most important vegetable in Iran, not only because of its economics importance, but also for the nutritional value of its fruit, mainly due to the fact that they are an excellent source of natural colors and antioxidant compounds. Water quality and fruit maturity stage has a significant effect on tomato fruit quality. In the present research, we studied the response of tomato grown at different salinity levels, as well as the changes that take place during different maturity stages, in order to improve the management and harvesting of tomato and obtain fruit of a higher nutritional value. Materials and Methods In this study, to evaluate the effect ofsalinity and fruit ripening stage on physical, biochemical properties and shelf life of tomato fruits in hydroponics system, an experiment was carried out as factorial based on randomized complete block design with three replications at Moghan College of Agriculture and Natural Resources, during 2015. Fruits from plants grown under three saline treatments (2, 4 and 8 dS m-1) were harvested at three ripening states (mature green, purple and red colors). The nutrient solution was prepared based on full strength of Hoagland's solution containing: 5.6 mM Ca (NO3)2, 4 mM KNO3, 1 mM KH2PO4. Characteristics such as fruit fresh and dry weight, total soluble solid, firmness, flavor index, titratable acidity, soluble protein, ascorbate peroxidase activity and shelf life were measured. Results The results showed that total soluble solids, juice electrical conductivity and dry matter percentage of fruits significantly increased with increasing salinity in the nutrient solution and progressed the ripening stage, but fruit water content decreased by 9%. The highest weight (147.7 g) and volume of fruits (153 cm3) were obtained in plants grown under 2 dS m-1 salinity and fruits harvested at the purple stage. Titratable acidity increased with increasing of salinity in the nutrient solution, but its content decreased at the red color stage. Firm fruit with greater shelf life was observed under 2 dS m-1 salinity and at the mature green stage. Soluble protein content in fruit was increased by increasing salinity levels in the nutrient solution, especially at the ripening stage. The application of 4 dS m-1 salinity at mature green stage increased ascorbate peroxidase activity up to 62.5% compared to 8 dS m-1 salinity and red color fruits. Discussion These results suggest that the improvement in fruit quality induced by the salinity is achieved by some biochemical changes and the reduction of fruit water content. Increasing of salinity in the nutrient solution can be useful for improving fruit quality, especially, when fruit is harvested at the red stage. However, with increasing salinity in the nutrient solution up to 4 dS m-1 and fruit harvest in the red color stage fruits with better quality and more suitable weight can be produced.

Weeds cause 23 to 30% reduction in yield of wheat annually. Winter wild oat is the most serious grass weed of wheat fields of Iran. Chemical method is the common way to weed control in wheat fields. ACCase inhibitors such as clodinafop-propargyl, diclofop-methyl and Fenoxaprop-P ethyl + Mefen pair di ethyl are widely used for control of grasses in this regard. Continuous application of herbicides with the same mode of action has resulted in developing herbicide resistant biotypes of weeds such as Avena ludoviciana. The objectives of the current study were: 1) to screen the collected accession of A. ludoviciana suspected to resistance to FOP herbicides, 2) to determine the resistance factor of resistant biotypes and 3) mapping distribution of resistant biotypes. Materials and Methods In order to survey Avena ludoviciana resistance to clodinafop-propargy, diclofop-methyl and Fenoxaprop-P ethyl+ Mefen pair di ethyl herbicides in wheat fields of Gonbad-e Kavus, several experiments were conducted in 2014 at Gorgan University of Agricultural Sciences and Natural Resources. (115 populations). The seed bioassay in Petri dish and pot experiment was conducted to determine resistance to acetyl coenzyme Acaboxylase inhibitors (ACCase), respectively. The seed bioassay experiment consisted of screening the suspected populations with discriminating concentration and concentration-response bioassay for resistant populations. Results In 48, 27 and 21 populations of A. ludoviciana were resistant to clodinafop-propargyl, diclofop-methyl and Fenoxaprop-P ethyl + Mefen pair di ethyl herbicides. Resistance accessions indicated varying degrees of resistance to tested herbicides. Based on EC50, the lowest and highest resistance to Clodinafop-propargyl herbicide was shown in Accessions of 74 and 6 with resistance factors of 1.99 and 31.42, respectively. Also for Diclofop- methyl herbicide, EC50 values from 2.88 for accession of 103 (with 1.34 resistanc factor) to 16.02 for accession of 91 (with 7.45 resistance factor) were different. In addition, EC50 estimatedfor Fenoxaprop-P ethyl + Mefen pair di ethyl herbicide was different from 0.124 to 0.86 for 20 and 6 accessions, respectively. Checking the distribution map of resistance accessions showed these accessions had uniform distribution in this city. Discussion Based on these observations, the existence of cross-resistance to herbicides was proved in some of the collected accessions. The results of this study can be used to correct programs designed to manage weeds herbicide resistance and prevent of the development of the plants in other areas.

Read More Abstract Background and Objectives After the potato and tomato, onion production is in the third place in world, but planting it in the fall season is faced with the problem of bolting. Although flower production is one of the main stages of seed production, flowering branches formation by consuming foods stored in the bulb reduces the quality and quantity of products. Critical size of plants for stimulating the production of flowering branches depends on genotype (Brewster, 1985). Temperature requirement for stimulating flower production varies between 5 and 12 degree Centigrade (Khokhar et al., 2007; Wiebe, 1990; Brewster, 1982). Thus, studying how this phenomenon, effective environmental conditions and its inducing factors for identifying bolting resistant genotypes is considered important. Materials and Methods The seeds of six Iranian landraces genotypes of onion including Lorestan, Qom, Tehran, Kerman, Hormozgan and Sistan (received from Iranian Gene Bank, Karaj), 10 indigenous genotypes collected from different regions of the center and south of Iran, as well as two foreign commercial cultivars were planted in pots in the greenhouse condition. After reaching the eight leaf stage, plants were transferred to the cold room. The average temperature of cold room was 8 °C and photoperiod was 11 hours brightness to simulate winter condition. To evaluate cold requirement for bolting, two different treatment times (45 and 70 days) were considered. After that treatment, pots were transferred to the greenhouse. Some morphological, phonological and physiological traits were measured. Results According to results of analysis of variance, differences of all traits between all studied genotypes were significant showing high variability in genotypes. Also, the effect of genotype and treatment for plant height, flowering percentage, total leaf soluble solids and total leaf soluble protein was significant. In this study, Kerman genotype was identified as the most resistance genotype to bolting in both treatments. Then Ghale Reiysi, Golden and Badrood genotypes, with the small flowering percentage showed the highest resistance to bolting. Badrood genotype had the highest sensitivity to bolting. Discussion Kazemi Shahandashti et al. (2011) suggested that cold treatment inhibits cell division so the temperature decrease leads to stoping the growth and changing the synthesis of proteins. However, in some plants during the cold treatment, adaptability to cold is normal and higher height in some genotypes can be attributed to cold adaptability of them. These results generally reflect the diversity between genotypes in terms of flower production and indicate that in line with other studies (Darabi, 2010), the flower production reduced weight, length, diameter and volume of bulb. In general, increase in total soluble solids can be considered as a physiological index for resistance to bolting.

Read More Abstract Background and Objectives Salinity and salt accumulation in the soil surface is one of the most important abiotic stresses that limit agricultural crop production in the arid and semi-arid regions of Iran. Thus, finding an alternative production technique or materials to alleviate stress condition is a research priority. One way to decrease the harmful effects of salinity is the foliar application of some chemicals such as Gamma-amino butyric acid (GABA) to increase plant tolerance to salinity. GABA is often accumulated in plants in response to live and non-live stresses such as drought, salinity, oxygen deficit, heat shocks and contamination of pathogens. Materials and Methods The current study was carried out to monitor the GABA effects on tomato physiological changes under salinity stress. The plants were grown in hydroponic system and received 0 and 50 mM NaCl as control and salinity stress, respectively. GABA at three concentrations 0, 10 and 20 mM was applied as a foliar application in either control or salinity treatments. Physiological characteristics of relative water content (RWC), membrane stability index (MSI), proline, total soluble protein (TSP) and enzymes activity, chlorophyll a, chlorophyll b, total chlorophyll and total soluble carbohydrates (TSC) were measured. Results The results showed that salinity stress and GABA were effective on the physiological characteristics of tomato. GABA can increase the tolerance of plants to environmental stresses, including salinity stress. MSI, chlorophyll a, total chlorophyll, carotenoids in salinity treatments were lower than control while TSC, proline, peroxidase and SOD increased. RWC, MSI, proline, TSC of GABA treated plant were greater than untreated in either salinity or control conditions. Also, SOD and peroxidase activity elevated in GABA treated plant under salinity stress. Discussion GABA increased antioxidant capacity in the plant and thus might eliminate radicals and prevents the destruction of cell membrane tissue, including the chloroplast membrane. In general, the results showed that in salinity stress conditions, GABA application as a compatible osmolytes might improve the tomatoes physiological performance and alleviate salinity stress.

Read More Abstract Background and Objectives Preharvest and harvest conditions can affect quality and biochemical composition of fruit during storage. Pomegranate (Punica granatum L.) is a non-climactric fruit and there is no clear physiological index to determine the proper harvest time. Lack of standard maturity index results in prolonged period of picking pomegranate fruit within the season. The effects of different physical or chemical treatments on postharvest quality of pomegranate fruit have been investigated in many studies, but our knowledge on the pre-harvest and harvest aspects that influence pomegranate fruit postharvest physiology is scarce. The objective of the present experiment was to determine how qualitative indices of pomegranate fruit in ‘Bagmalek’ (Khuzestan, southwest of Iran) are affected by maturity stages at harvest and storage duration. Materials and Methods A factorial design including three harvesting times and three storage durations with three replications was used for statistical analysis. Fruits of ‘Malas Post Zard’ as an important pomegranate cultivar were harvested from a commercial orchard in ‘Bagmalek’ (150 Km Ahwaz, Khuzestan, southwest of Iran). Fruits were harvested at three harvest times at 14-day intervals (14 September, 29 September and 13 October 2011) and stored for 40 and 80 days at +7 oC. Some physical (weight, surface color, decay and shriveling) and biochemical characteristics (ascorbic acid, total soluble solids, titratable acidity, pH, anthocyanin index and browning index) were evaluated. The factors were evaluated at harvest time and after 40 and 80 days. Results Regardless of storage durations, pomegranate fruit showed significant variations in total soluble solids, titratable acidity, pH and anthocyanins. Interactions of the harvest time and storage duration (for 40 and 80 days) were significant on gravity, surface color changes, shriveling, ascorbic, acid, pH, titratable acidity and fruit juice pigments (anthocyanin index and browning index). Results showed that the period 29 September until 13 October 2011 and storage duration for 40 days are the best treatments to maintaining postharvest quality of ‘Malas Post Zard’ fruits in ‘Bagmalek’ (Khuzestan, Iran). Discussion Results showed that quantity and quality indices of the pomegranate could be improved through the selection of proper harvesting time. Harvest of pomegranate fruit at different maturity stages altered fruit appearance quality indices and biochemical characteristics under ‘Bagmalek’ geographic condition, and at early harvest time (14 September), it reduced fruit quality but did not make its consumption unacceptable. More research needs to assess the effect of different climatic parameters such as temperature and light intensity on the postharvest quality of pomegranate fruits in Khuzestan.

Where vineyards have access to a permanent and unlimited water source, irrigation can be managed so that water stress is imposed during certain periods of time to increase fruit quality and to control canopy. This study was conducted to investigate the effects of different levels of topping times and preharvest cut-off irrigation treatments on the qualitative and Quanitative characteristics of Askari grapes in Dena (Sisakht) region during 2013-2015. Materials and Methods ‘Askari’ vines were 18 years old on their own roots, trained as a head system. The vines were spaced 2.5 × 3 m. They were pruned as spur pruning which left 60 buds (4-node spurs) on each vine. Vineyard had a drip irrigation system. Shoot topping done at 10th buds of the last cluster. An experiment was arranged as factorial that the first factor included different levels of topping dates with four levels (control, after fruit set, unripe stage and veraison stage), and the second factor involved the preharvest irrigation cut-off date treatments on four levels (Control, 70, 80 and 90 days after full bloom) in a randomized complete blocks design with four replications. Factors relating to the determination of the quality of fruit juices included %TSS, pH, TA%, vitamin C and TSS/TA ratio respectively. Also, bunch weight and yield were measured. Results Results of the analysis of variance showed that the effect of topping and irrigation cut-off date treatment were significant on the TSS, TSS/TA, pH and Vit-C content (P≤0.01). But the interaction effects were significant only on the TSS/TA and Vit-C (P≤0.01). Finally, mean comparison showed that the highest vitamin C was observed at veraison stage and irrigation cutoff dates treatments at 80 and 90 days after full bloom. Discussion In some of vineyards, high vigour vines also have too much canopy shading which is detrimental to the light microclimate and has also been shown to lead to increase of TA and decrease of TSS and TSS/TA ratio. Therefore, canopy manipulation is used successfully in grape production to balance the vegetative and reproductive growth of vines. Differences in vine water status before veraison has been shown to have no effect on the onset of veraison, while withholding water after veraison can delay the accumulation of soluble solids under severe water deficits. Certainly, light and temperature are the most important environmental conditions that impact the growth and quality of grapes and unshaded conditions affect better coloring and increase TSS/TA ratio.

Read More Abstract Background and Objectives Lilium ledebourii is one of the wild species of Lilium genus, growing in northern Iran. This species has high potential for export. Plant tissue culture techniques are widely used in plant propagation, and using these methods can effectively provide micro-propagation of this plant in the large scale. High percentage of regeneration is necessary for plant protection, used in the breeding programs. Materials and Methods The experiment was factorial based on completely randomized design with four replications and was carried out in tissue culture Lab of Mohaghegh Ardabili University in 2015. For this purpose, segments of explant that were treated with ultrasound were cultured on MS medium supplemented with different concentrations of NAA and BA alone and/or in combination with each other. This experiment conducted Concentrations of NAA at four level Zero, 0.01, 0.1 and 1 mgl-1 and Concentrations of BAP at four levels Zero, 0.01, 0.1 and 1 mgl-1 and Ultrasound at four levels zero, 5, 10, 20 and 30 seconds. In order to remove possible contamination from the mediums, all media were autoclaved for 30 minutes at 121 °C. At the end of the experiment, the number of bulblet produced, root length, fresh weight of bulblet and etc were recorded. The cultures were kept at 20°C or 25°C under 16 h photoperiod or in darkness. Results The results showed the highest number of bulblets produced in 0, 0.01, 0.1 mg L-1 NAA and 5secend ultrasound, 0.1 mg L-1 NAA and 0, 5, 20 and 30 second ultrasound compared with control. The highest frequency of bulblet regeneration was produced in MS medium supplemented with 0, 0.01, 0.1 mg L-1 NAA and 5 second ultrasound. On the other hand, 0.1 mg L-1 NAA and 5 second ultrasound increased number and weight of bulblets. Different concentrations of NAA had also significant effects. Discussion On the other hand, ultrasound increased the number and weight of bulblets. Mechanical stress and micro streaming by acoustic cavitation might be considered as the most possible cause of the various physiological effects of ultrasound on cells. Low-energy ultrasound can modify cellular metabolisms or facilitate the uptake of nutrient, and make them easily through the cellular walls and membranes. Finally, it seems that ultrasound in combination with growth regulators have the potential to produce the highest average number of bulblets in the scale explant, using plant growth regulators in combination with an abiotic stress in in vitro.

In order to select suitable clones for commercial cultivation in Dezful climatic conditions, in this study morphological and biological production traits of 58 clones of Satureja rechingeri were evaluated. Materials and Methods After initial screening of plant populations, 58 genotypes were selected based on their adaptation into field conditions. To evaluate the morphological and biological production traits of these clones in Dezful climatic conditions, an experiment was conducted based on randomized complete block design with three replications. Morphological and biological production traits of 58 clones were evaluated in the second year of cultivation at the flowering stage. Results Analysis of variance showed significant differences for all studied traits (p< 0.01) among 58 clones. The most important traits varied in terms of plant height (27.83-71.50 cm), fresh weight (20.26-289.71 g/p), flower and leaf weight (4.05 to 83.62 g/p), and plant oil yields (0.07 to 1.54 g/p). Flowers and leaf dry weight had a significant and positive correlation with plant height, number of the main branches and lateral branches, leaf width, plant diameter, fresh weight, dry weight and essential oil yield. Cluster analysis grouped the 58 clones within two main groups. Factor analysis showed that three main factors justified 70.62 percent of the whole variation. Clones of Z28, Z19, K48, E7, H82, F14 and G48 were superior with regard to weight of dry leaves, flowers and essential oil yield. Discussion Superior S. rechingeri clones could be used for commercial cultivation in Dezful climate conditions. The most important biological production traits, yield of leaf and flower fraction, essential oil content and yield showed significant variation between clones. These variations could be used to breed high-quality and desirable cultivars for use in food and pharmaceutical industries.

Read More Abstract Background and Objectives Chemical variability among populations according to their geographical and bio climatic distribution imposes that conservation strategies of populations should be made appropriately, taking into account these factors. Perovskia, with the common Persian name of "Brazemble", a small genus from Lamiaceae family, is distributed in various regions of Asia, as Iran, Afghanistan and Pakistan. The genus is represented in Iran by only three species as P. abrotanoides, P. atriplicifolia and P. artemisoides. The plant is an aromatic shrub which mainly grow in mountains at an altitude of 2200 to 4200 m from Northeastern across center to Southeastern of Iran. All parts of the plant are aromatic, but Perovskia is not edible. The plant is used in Iranian folk medicine as an analgesic in rheumatic pains, treatment of leishmaniasis, fever and headache. The present study, was planned to evaluate variations in essential oil composition and contents of different Plant Organs from P. abrotanoides. Materials and Methods In this study, different plant organs (leaf, flower and stalk) of P. abrotanoides were collected from natural habitat in North Khorasan province and their essential oil content and composition were studied. The experiment was arranged in a completely randomized design with three replications for the essential oil contents. Essential oils were analyzed by Gas chromatography (GC) and Gas chromatography–mass spectrometry (GC-MS) in respect to their chemical composition. Results Essential oil contents of leaf, flower and stalk were 1.0, 2.3 and 0.7% (w/w), respectively. The total number of compounds identified and quantified was thirty-nine in leaf, thirty-two in flower, and thirty-five in stalk, representing 99.4, 99.8, and 98.7 % of the total essential oil, respectively. The major compounds of the essential oil were α-bisabolol (2.6-29 %), 1,8-cineole (11.4-16.5 %), camphor (10.1-21 %), α-pinene (1.9-16.2 %) and δ-3-carene (7.4-11.3 %). Results showed that leaf essential oils of the plant characterized with high content of oxygenated sesquiterpenes (41 %). Oxygenated monoterpenes, were rich in flower (42.1 %) and stalk (49.5 %). The main volatile compound identified in the leave was α-bisabolol, which reached a concentration of 29%. The content of essential oil in the flower was higher than that in the leave and stalk. Discussion Results of the current study show that the main compounds and content percentage of essential oil were different in each plant parts. By using the hydrodistillation, 32 to 39 compounds were identified representing 98.7% to 99.4 % of the total oil components. The essential oil analysis revealed that α-bisabolol, 1, 8-cineole, camphor, α-pinene and δ-3-carene, were main essential oil constituents in all organs. The variation observed for essential oil content was 0.7% to 2.3% in stalk and flower, respectively. Chemical variation of P. abrotanoides essential oils from different plant parts, which is significant for conservation and breeding programs, can be considered by medicinal plants breeders and pharmaceutical industries for breeding and processing uses.

Read More Abstract Background and Objectives The storage life of onion is an essential factor to marketing this product in the winter season. The storage life of onion depends on different factors of which withholding of irrigation before harvest is the most important. On the other hand, Brassinosteroids (BRs) are widely distributed in the plant kingdom, with pleiotropic effects, as they influence varied developmental processes like growth, germination of seeds, rhizogenesis, flowering, stem elongation, pollen tube growth, leaf epinasty, ethylene biosynthesis, proton pump activation, gene expression and photosynthesis. Therefore, studying the effects of withholding irrigation and Brassinosteroied on quantitative and qualitative characteristics of onion seems to be necessary. Materials and Methods In order to evaluate the effect of withholding irrigation and Brassinosteroied hormone before harvesting on yield and postharvest life of Baluchistan onion, a split plot experiment in a randomized complete block design with three replications was conducted in Iranshahr’s Institute of Agricultural Research and Education in 2015-2016. This experiment was performed to evaluate three factors, water withholding at three levels (one, two and three weeks before harvest time) and spraying Brassinosteroied in four levels (0, 0.5, 0.75 and 1 μM). Different traits such as Bush height, diameter, length, weight, edible layer of bulb and neck diameter, post-harvest life, dry matter percentage and yield were measured. Results Agronomic traits and post-harvest life of onion increased by increasing the hormone concentration to 0.75 μM. The highest yield observed in 0.75 μM of Brassinosteroied treatment was about 42 t/ha and in the treatment with one week of withholding irrigation before harvesting was about 39 t/ha. The lowest yield observed in without hormone treatment was about 33 t/ha and in the treatment with three weeks of withholding irrigation before harvesting it was about 36 t/ha. The maximum post-harvests observed in two weeks of withholding irrigation before harvesting and 0.75 μM of hormone treatments were about 60 and 50 days, respectively. The lowest post-harvest lives observed in one week of withholding irrigation before harvesting and without hormone treatments were about 33 and 43 days, respectively. Discussion Withholding irrigation can decrease the amount of water in bulbs and increase post-harvest life because of reducing post-harvest diseases. On the other hand, Brassinostroides can induce growth and photosynthesis rate and alleviate the adverse effects of abiotic stress in plants. These are important reasons to increase quantitative and qualitative traits in this product.

Read More Background and objectives: According to a breeding program carry out in Citrus and Subtropical Fruits Research Center (Ramsar), ‘Noushin’ and ‘Shahin’ mandarins were created from crossing between Clemantin mandarin×Salustiana orange and Clemantin mandarin×Hamlin orange, respectively. The fruits of this new varieties on Sour orange rootstock did not study yet, completely. Therefore, the aim of this study was to investigate the Noushin and Shahin physico-chemical characteristics on Sour orange rootstock during two years. Materials and methods: ‘Noushin’ sampled from October to December during three months. ‘Shahin’ fruits were collected from October to February (5 times) as monthly. Various physico-chemical characteristics evaluated including fruit lenght, width, thickness; arithmetic, geometric, equivalent and harmonic diametrics means; aspect ratio, sphericity, surface area, true volume, apparent volume, volume error, density, peel thickness, Easy of peeling, weight, juice percentage, seed number, peel color indices (L*, a*, b*, hue angle, chroma and CCI), total soluble solid (TSS), titratable acidity (TA), technological index (TI) during experiment. Results: The results showed that fruit size, arithmetic, geometric, equivalent and harmonic means of both varieties had not significantly (p<0.01) changes from the first to final time of harvesting. There was a positive correlation between fruit sphericity and arithmetic, geometric, equivalent diametric means. Fruit of ‘Shahin’ had high aspect ratio, less sphericity and low density (<1). Fruit density showed an increasing trend during the harvest time. The green colour of peel in ‘Noushin’ and ‘Shahin’ changed to orange colour at the last week of November and December, respectively. Based on citrus colour index (CCI), no cultivars necessary to de-greening after the last week of November. The TSS:TA ratio was optimum for harvesting in late of October (9.81) in ‘Noushin’ and at the last week of December (8.32) in ‘Shahin’. Although the technology index in both varieties had not significantly (p<0.01) changes from the first to final harvesting but it was high in ‘Shahin’ with 50 percent nearly. The peel thickness in ‘Noushin’ and ‘Shahin’ was 3 and 2 mm respectively which had not significantly (p<0.01) changes during harvesting time. The seed number in ‘Noushin’ was 19 to 27 and in Shahin was 15 to 35 that revealed both of them belonged to high seed content category. Conclusion: According to TSS:TA ratio index, ‘Noushin’ as an early ripening variety can harvested in the last October and ‘Shahin’ as mid ripening variety can harvested in late of December. Both of varieties is sutaible for the north of Iran climate. Based on juice percentage and TI data, the amount of juice with about 50% was higher in ‘Shahin’ that sutaible for prosessing industries. Due to empty space inside the fruit skin and flesh of ‘Noushin’ and low dansity (<1), fruits are floating on washing water during prosessing. The skin colour was green in both of varity in late of October. Becase of TSS:TA ratio is sutaible at this time in ‘Noushin’, there fore growers can harvested ‘Noushin’ variety with green skin colour in late of October but ‘Shahin’ must be remained on tree until late December.

Effect of hormonal compositions on micropropagation of fifteen cultivars of Gerbera (Gerbera jamesonii Bolus ex Hooker f.)

Read More Background and objectives Gerbera is one of the most important ornamental plants that used as cut flower and potted plant. The common methods of propagation does not have performance for supplying the global demand of this ornamental plant. So the most common method for commercial propagation of gerbera is micropropagation. Material and methods In this study, effect of different hormonal treatments and cultivars on micropropagation and rooting of gerbera shoot tip explant was evaluated. Shoot tip explants were first washed with running tap water for 30 min then they were surface sterilized by dipping in 1.5% sodium hypochlorite solution for 5 min and rinsed with sterile distilled water, followed by immersing in 0.1 % mercuric chloride solution for 5 min. After Sterilization with mercuric chloride solution, capitulum was washed with sterile distilled water. Subsequent washing was done with sterile distilled water three times. Sterilization with sodium hypochlorite solution and mercuric chloride and final rinse with sterile distilled water were done under laminar air flow hood. In the proliferation phase, MS medium containing 1 mg/l BA or KIN in combination with 0.1 mg/l IAA or hormone-free medium, 3% sucrose, 8 g/l agar was used. For rooting of propagated plantlets, ½ MS medium containing 1 mg/l IBA, IAA, 2IP or ½ MS hormone-free medium, 3% sucrose, 8 g/l agar was used. After 4 weeks different parameters such as number of plantlet, height of plantlet, number of main root, root length, number of secondary root were measured. Cultivars that was used in this experiment are as follows: 1: ‘Aventura’, 2: ‘Barones’, 3: ‘Mayfair’, 4: ‘Cacharelle’, 5: ‘Real’, 6: ‘Sorbet’, 7: ‘Dalma’, 8: ‘Goldy’, 9: ‘Panama’, 10: ‘Lancaster’, 11: ‘Dune’, 12: ‘Bellavue’, 13: ‘Blinddate’, 14: ‘Applause’, 15: ‘Sunway’. Results and discussion The results showed that in all studied cultivars, the medium containing BA lead to producing the highest number of plantlet. So using the medium containing BA in propagation phase is recommended for obtaining the highest number of plantlet. Also the results showed that the rooting of gerbera plantlets was affected by cultivar and suitable hormonal composition for rooting of each cultivar was different. However, the appropriate acclimation of produced plantlets is so important. Although the length and number of roots in ½ MS medium was at the average of other treatments, but propagated plantlets from this medium, had a suitable acclimation capacity. Moreover, additional cost for utilization of rooting hormones was reduced and therefore commercial production cost decreased. So application of this medium for rooting of different gerbera cultivars is recommended. Finally acclimation of rooted plantlet was done in cocopeat and perlite medium, with 95% success.
