Read More Background and ObjectivesGrape is one of the fruit trees that need to well management to produce economical production. Farmers often do not properly prune vine grape. Grape cultivars show different response to pruning severity. Perlette cultivar cultivation is increased recently. The main objective of this research was to determine the severity and number of buds per shoot on yield and productivity coefficient of perlette grape cultivar. Materials and methodsA factorial experiment based on randomized complete block design with three replications was conducted with three charges levels 42, 60 and 72 and three buds number per shoot include (6-2 buds), (5-2) and (4- 2) during 2012 in Garaban agro industrial complex. Some different traits on flower and fruit quantity and quality were measured. ResultsResults of mean comparison showed that Charge, number of buds per shoot and their interaction effects were significant on number of cluster, cluster weight, yield and productivity coefficient. The interaction effect between charge and the number of buds on the yield per plant showed that the highest yield (about 12 kg) was obtained by charge 60 and long pruning with 6 buds and the lowest yield (about 5.6 kg) was in charge 42 with 4 buds on the branches. The highest cluster weight of about 250 grams was obtained in charge 60 with 4 buds on the branches and the lowest cluster weight (182 g) found in charged 72 and 4 buds on the branches. The interaction effect of charge and the number of buds per shoot was significant on productivity coefficient at 1%. The highest productivity coefficient (0.9833) was found in the charge 42 and 5 buds per shoots and the lowest was in charge 60 with 4 buds per shoots. Generally 60 charge was recommended for this cultivar as mixed pruning with two replacing buds and a shoot with 6 bearing buds.DiscussionIncreasing the number of clusters possibly depend on increasing the charge level, which is one of the yield increasing factors. Berries weight loss and berries abscission lead to cluster weight reduction and yield was affected. Thus, the high charge of 72 buds per plant caused yield loss due to small clusters. This phenomenon is possibly due to lack of adequate nutrition by the vine for cluster growth. Yield reduction in 42 charges was due to a reduction in the cluster production number. Yield was increased with a decrease in pruning intensity, which represents an increase in the number of buds on the fruiting branches. The long pruning with 6 buds per cane and 60 charges was superior to the others. This pruning severity was determined based on fruit yield and quality. Proper pruning lead to balance on vegetative and reproductive growth and quantitative and qualitative yield will be increase.

Read More Abstract Background and Objectives Strawberry (Fragaria × ananassa Duch.) is one of the most popular soft fruits worldwide. It is a rich source of health promoting compounds such as essential macro/micro-nutrients, polyphenols, anthocyanin, vitamins and amino acids, while showing a high antioxidant capacity. Preharvest applications of calcium lactate and potassium sorbate have shown to be promising as a tool to improve the quality and extend the shelf life of various fruits. To our knowledge, however, little information is available regarding the effect of calcium lactate and potassium sorbate on strawberry fruits. Thus, the objective of the present study was to determine the effect of foliar application of calcium lactate and potassium sorbate on important quality characteristics and antioxidant properties of ‘Armas’ strawberries. Materials and Methods This research was performed at a commercial orchard on 2-year old strawberries in Marivan, Iran in 2015. The experiment was arranged as factorial based on randomized complete block design with two factors and four replications. The first factor included different levels of calcium lactate (0, 0.5 and 1%) and the second factor included different levels of potassium sorbate (0, 0.5 and 1%). Foliar spraying was performed in three stages: three weeks before commercial harvesting, at initial coloring, and one week before harvest. Fruit were harvested at commercial maturity and immediately transported to the laboratory at University of Zanjan and subjected to physicochemical analysis. Differences among means of data were analyzed by Duncan’s test at p ≤ 0.05 (n = 3). All statistical analyses were performed with SAS version 9.4. Results The results showed that the combination of calcium lactate and potassium sorbate (0.5 Percent concentration) treatment significantly (P £ 0.01) increased the total anthocyanin content of the whole fruit, reuslting into further attractive fruit color and market than the control. It also increased antioxidant properties up to a level of 57.07%. On the other hand, the application of this treatment (calcium lactate and potassium sorbate 0.5 Percent) significantly increased the total flavonoid content, total soluble solids, flavor index (1.1 Percent) and calcium lactate (1 Percent). Potassium sorbate increased total phenol, total acidity, and fruit acidity significantly compared to other treatments and control (P £ 0.05). Discussion In general, the results of the present study showed that a preharvest application of calcium lactate and potassium sorbate increased firmness, nutritional quality, and antioxidant potential of strawberry fruits. Thus, the use of these two materials is recommended to improve the qualitative and antioxidant properties of strawberry fruit.

Read More Abstract Background and Objectives Khuzestan Province is one of the most important areas that provide vegetables throughout winter and spring in Iran. The vegetables are mainly picked up in the afternoon, so it cannot be supplied to the market. In this time of the day, the minimum amount of nitrate is accumulated in leafy vegetables, with a gradual increase throughout the night. On the other hand, its highest level is in the early hours in the morning. The purpose of this study is to evaluate nitrate and nitrite accumulation in vegetables produced in different areas of Ahwaz during winter and spring. Materials and Methods This experiment was conducted in a randomized complete block design arranged in a split-split-plot design with three replications in the Department of Horticulture, College of Agriculture, Shahid Chamran University of Ahvaz. In the treatments viz. first to third harvest dates (1): February 6, 2016, (2):March 4, 2016 and (3): April 7, 2016 respectively were the main factors and nine vegetable species (including coriander, parsley, mint, fenugreek, chard, spinach, leeks, dill and watercress) kept in sub plot, three vegetable production areas in Ahvaz were arranged in sub plot in sub plots and three fields were selected in each of these areas. Before and after cultivation, all planting, harvesting and harvesting operations were carried out on the same field in the selected fields. The concentration of nitrate was measured by the method introduced by Cataldo et al. (1975) and the nitrite concentation was measured via the method of Abu-Dayeh (2009). The data were analyzed using SAS software and the mean comparison was calculated using Duncan's multiple range tests at a probability level of 5%. Results Nitrate and nitrite contents of the produced vegetables were different depending on various regions, harvesting time and vegetable species. The highest nitrate content was observed in parsley fromthe first harvest in the southern region of Ahwaz (4.4 mg/kg-1 dry matter) and the lowest amount of nitrate was recorded in watercress from the third harvest in the northern region of Ahvaz (1.4 mg/kg-1 dry matter). Also, the highest nitrite content was observed at the first harvest in the westhern region of Ahwaz in mint and the lowest amount of nitrate was recorded in the second harvest in the northern region of Ahvaz in chard. In winter, the environmental conditions in Ahwaz are suitable for the growth and development of leafy vegetables (Boroujerdnia et al., 2007); however, soil conditions provide manganese uptake by plants. Moreover, high light intensity is another factor which can decrease nitrate in leafy vegetables. The number of cloudy days in Ahwaz is very low; therefore, the number of sunny days with high radiation intensity is high in winter. This phenomenon greatly reduces the content of nitrate in leafy vegetables. Discussion The amounts of nitrate and nitrite content in various species of vegetables produced in three regions of Ahvaz during the cold season were much lower than the standard figures. Therefore, we can conclude that the quality of vegetables produced in Ahvaz is very high in terms of nitrite and nitrate contents.

Read More Abstract Background and Objectives An important requirement of agronomic planning is to evaluate different plant nutrition systems in order to achieve high yield and quality, especially in medicinal plants. With the correct method of increasing soil fertilization and plant nutrition, it is possible to reduce the erosion and conservation of biodiversity, preserve the environment, and increase the efficiency of inputs. This research was carried out with the aim of improving the quantitative and qualitative performance of lemon balm with bio fertilizers such as mycorrhiza fungi, biophosphate fertilizer, and vermicompost. Materials and Methods The experiment was conducted in spring and summer of 2016 in a field covered by Agriculture Jihad of Guilan, located in Khoshkabijar District of Rasht, Iran. In this research, the experiment was conducted considering three factors: factorial contains mycorrhizal inoculation (M1 = non-inoculated and M2 = inoculated), phosphate biofertilizers (p < sub>1 = 0, p < sub>2 = 50 and p < sub>3 = 100 kg ha-1) and vermicompost (V1 = 0, V2 = 5 and V3 = 10 tons ha-1) layout based on a randomized complete block design with 18 treatments and three replications. Also, one plot was allocated to control in each replication and only chemical fertilizers (use of 90 kg ha-1 of fertilizer of urea, triple super phosphate 90 kg ha-1 and potassium sulfate 90 kg ha-1) and data obtained from control plot, mycorrhizal, phosphate biofertilizers, and vermicompost were used based on a randomized complete block. Results According to the results of this experiment, the highest number of leaf yield, essential oil percentage, biological yield, essential oil yield, total chlorophyll and carotenoids in inoculation with mycorrhiza was obtained. The application of 100 kg of phosphate biofertilizers had a significant effect on all measured biofertilizers. Fertilizer treatment of 100 kg ha-1 had the best result in all measured traits. Also, the best result was obtained with the use of 10 tons of vermicompost per hectare in biological yield (4808.2 kg ha-1), essential oil (0.13%), and total chlorophyll (1.54 mg gr-1FW). There were positive and synergic interactions between factors. For example, positive interaction between inoculation of mycorrhiza and phosphate biofertilizers on leaf yield and essential oil yield, and synergic interaction between mycorrhiza and vermicompost on biological yield were obsereved. Discussion The results of this study show that the treatment of inoculums with mycorrhiza, consumption of 100 kg of phosphate biofertilizers and 10 tons of vermicompost per hectare could provide the best situation to achieve the highest quantitative (5290 kg ha-1) and qualitative yield (0.26% essential oil) of lemon balm in a sustainable agricultural system.

Read More Abstract Background and Objectives Mineral nutrition and post-harvest handling are the major factors affecting secondary metabolites content and composition of plants. Berberis integerrima is one of the most important medicinal plants listed in all pharmacopeias. The bark, fruits and roots of this plant are commonly being used by traditional and modern medicine disciplines. They are well documented for their anti-diabetic, anti-cancer, and antimicrobial activities with berberine as the main active constituent. Also, the shrub has ornamental uses in the landscape and the fruit is used as a food additive. In addition, anthocyanin of the fruit is useful as a natural coloring agent in food industries. There are few studies on the agronomic demands and post-harvest handling of this plant. The experiment was conducted to assay the effects of nano-zinc foliar application and postharvest plant drying temperature on some phytochemical traits of this highly valuable plant. Materials and Methods The treatments were applied to assay the phytochemicals content variations in roots, fruits and aerial parts of the plant (2 years old) by two separate experiments as factorial based on RBCD. In the first experiment, the effects of nano-zinc foliar spray on the yield, anthocyanin content, Zinc content and TSS at the different mentioned organs were studied. In the second experiment, the effects of diverse post drying temperatures (25, 50 and 750C) on total phenolics content, flanonoids content, hydrolysable tannin and berberin content in Berberis integerrima were assayed. Results Zn content, TSS content and anthocyanins were affected by the independent effects of foliar spray and plant part. Plant dry weight was impacted by the organ type. Phenolics, flavonoids and alkaloids contents were affected by the interactional effects of the foliar spray , the organ type, and the drying temperature. The highest figure for the contents of alkaloids (30 mg g-1DWt) and flavonoids (26.9 mg g-1DWt) were from the root × foliar spray by 6 mgL-1 Nano-Zinc at 250C drying regime. The maximum amount of total phenolics was attained by the aerial parts × foliar spray by 6 mgL-1 Nano-Zinc at 250C and fruits × 3 and 6 mgL-1 foliar spray at 250C. For hydrolysable tannins (1.9%), the root and the aerial parts × 250C showed the largest figure. Discussion The overall results revealed the positive effects of nano-zinc foliar application on the studied gross secondary metabolites content in Berberis integerrima. The post-harvest drying temperature influenced the metabolites content as well, and the optimized temperature in the experiments was 250C.

Relationship of Color Indices with Some Chemical Characteristic in Red and White Apple Cultivars

Read More Abstract Background and Objectives Apple is one of the most important common fruit trees in temperate regions of the world with a lot of phenotypes and genotypes. Fruit surface color is one of the important factors in evaluating the qualities and appearance of fruits. This is the first component of acceptance by the customer, which indicates anomalies in agricultural products. Materials and Methods For this purpose, a study was conducted in 2016 in a completely randomized design with 12 varieties of native and imported apples in three replications. In this research, qualitative characteristics including anthocyanins for skin and flesh fruit, total sugar, flavor index, pH and EC and color indexes including brightness index (L), redness index (a), yellowness index (b), Hue (H), and Chroma (C) were measured for each apple cultivar. Results The results showed the highest indexes of a and C of skin. Moreover, the index of a related to the flesh of fruit in Mazand 2 had the highest levels of Hue skin, brightness and Hue flesh in Gala cultivar. The highest amount of brightness and yellowneass of fruit skin and flesh was observed in Golden delicious cultivar. While the highest amount of anthocyanin in skin was found in Red delicious cultivar. the highest amount of anthocyanin in flesh was found in Mazand 2. Furthermore, the highest level of flavor index was found in Shahrod 10 and the maximum amount of total sugar was detected in Red delicious cultivar. The utmost EC juice and the highest juice pH were observed in Gol Ghermez and Golab Kohanz cultivars, respectively. The highest positive correlation was found between anthocyanin of skin and anthocyanin of fruit pulp in the red flesh apple (0.99). Based on the results of this research, the red index and the fruit flavor index correlated with skin Hue, brightness of skin correlated with yellow color of skin, and fruit EC correlated with sugar content of the fruit. Discussion Finally, the highest correlation was found between anthocyanin in skin and the red flesh apple. In other apple cultivars, a significant correlation was found between index (a) and the yellowness of the apples (0.92). Further positive correlation was set between index C and anthocyanin content and flavor index. A positive correlation was observed between H and L indexes and the yellow color of the apple. Index b indicated a significant negative correlation with anthocyanin of skin and flesh of apple. The highest red content in red flesh apples indicated positive correlation with a and c indices, which has already been confirmed by the results of other researchers.

Read More Abstract Background and Objectives The effect of different physical and chemical treatments on quality of pomegranate fruit at postharvest has been studied, but little information is available in the case of pomegranate fresh aril postharvest as a ready-to-eat product. Coating is one of the treatments to preserve the postharvest quality of the pomegranate fresh aril. Since consumer health is impotant in the selection of postharvest treatment, the use of compounds extracted from plants as wax or coating agent is a safe treatment for fresh arils. Extracted gel from leaf tissue of Aloe vera L. is an herbal gel that can be used as coating agent. The objective of the present experiment was to evaluate the effects of Aloe vera gel on the postharvest quality of pomegranate fresh arils. Materials and Methods Pomegranate Fruits (known as ‘Malas Poost Zard’) were harvested from a commercial orchard located in Baghmalek, Khuzestan. Fresh arils were removed manually from fruits and treated with Aloe vera gel (0, 2.5, 5 and 10%). Fresh arils were stored at 7 oC, RH = 90% for 4, 8 and 12 days. Some biochemical characteristics including ascorbic acid, total soluble solids, titratable acidity, total phenolics, color intensity, anthocyanins and browning index were evaluated. Results Results showed that the interaction of Aloe vera gel concentration and storage duration had a significant effect on ascorbic acid, titratable acidity, total phenolics, color intensity, anthocyanins and browning index. All concentrations of Aloe vera gel decreased total soluble solids. The highest concentration of Aloe vera gel (10%) significantly increased anthocyanins/browning ratio. The quality of pomegranate fresh arils significantly decreased with the increase in the duration of storage. During storage condition of fresh arils, Aloe vera gel (2.5 – 5%) improved the color intensity of fresh arils after 4 days. Also, results showed that all concentrations of Aloe vera gel helped to preserve higher ascorbic acid after 8 and 12 days. Discussion Aloe vera gel improved the quality of pomegranate fresh arils. Aloe vera gel contains the different antioxidant and nutrients which may be responsible for the positive effects on quality of pomegarnate arils. Results showed that Aloe vera gel is a promising herbal product for coating pomegranate fresh arils. More research needs to be undertaken on the efficacy of Aloe vera gel on postharvest quality of pomegranate fresh arils and other minimally processed products.

Read More Abstract Background and Objectives Olive tree (Olea europaea L.) is one of the most ancient tree crops which has been domesticated in the Middle East for about 6000 years. The romans enlarged its cultivation from the Greek islands throughout the Mediterranean basin, mainly along the African and European coasts as well as in the areas where large volumes of water have improved the climate. In the past, cultivar distinguishing approaches were based on morphological characteristics of leaf, fruit and stone. Recently, morphological traits with isozyme and DNA- based markers have been applied to the classification of cultivars. The aim of this study was the evaluation of morphological and molecular similarities between the new and the known cultivars of olive trees in the Fadak garden in Qom province in Iran. Materials and Methods In order to analyze genetic variations of olive tree genotypes in Qom province, the olive genotypes (Manzanilla, Sevilana, Mahzam, Arbequina, and Conservalia) and two superior cold resistant genotypes (Fadak and Tooba) were studied using morphological traits based on distinctness, uniformity and stability (DUS) guideline in the Fadak garden (Qom province, Iran) in 2013. In addition, a total genomic DNA was extracted from the leaves, and the PCR was carried out by a set of six inter simple sequence repeats (ISSR) markers for amplification. Then, amplification products from ISSR primer tests were characterized on 2% agarose gels immersed in 1X TBE buffer. The gels were stained with ethidium bromide. The PCR fragments were scored for the presence or absence as 1 and 0, respectively. Results In this study, analysis of variance was performed on 12 quantitative traits. Results showed there were significant differences (p < 0.01, p < 0.001) between all the traits and Fadak and Tooba separated from other genotypes by comparing the means of the tests (Duncan multiple range method). According to the biplot graph (the first two components) in the PCA method by quantitative traits, Arbequina, Manzanilla and Tooba were included in one group, and Fadak and Tooba were distinctly separated from other genotypes. Also, according to the biplot graph (the first two components) in the PCA method by qualitative traits, the distance between Fadak and Tooba was low. In the molecular analysis, all ISSR markers (six primers) showed polymorphism. The total band range was 8-21 (with 14.66 as the average) and the PIC varied from 0.231 to 0.447 (0.309 average). Based on the PCA method, Tooba was utterly separated from other genotypes. Discussion In this study, Fadak and Tooba were completely different from other varieties using morphological traits and ISSR molecular markers. Fadak was found to be similar to Sevilana and Conservalia. There was a high similarity between Fadak and Tooba. Also, no similarity was observed between the results of morphological and molecular traits.

Read More Abstract Background and Objectives Melissa officinalis is a medicinal plant belonging to the Lamiaceae family. The essential oil of this plant is used in many fields. Although more than 100 types of chemical substances have been identified in this plant, the most important compositions of its essential oil include citral, citronella, geraniol and linalool. Heavy metals are one of the main causes of non-biotic stress for living organisms due to increased use in the field of industrial and agricultural development and its high accumulation and toxicity. Cadmium is an unnecessary heavy metal, which, due to its high mobility and low concentration, easily enters the food chain from the soil. Cadmium induces oxidative stress by stimulating the synthesis of free oxygen radicals in the plant. In this study, the effect of cadmium chloride on oxidative stress induction in lemon balm was investigated. This experiment was carried out in 2016-2017 at Shahrekord University. Materials and Methods This research was conducted in a completely randomized design with three replications in vitro condition. We investigated the effects of different concentrations of cadmium chloride (0, 10, 20 and 40μm) on the biochemical parameters of the lemon balm. In this study, sterile stems of lemon balm propagated on the medium (1/2 MS) were used. After 60 days, seedlings were removed from solid MS medium and the roots were cut and cultured in liquid medium of 1/2 MS with different concentrations of cadmium. The cultivation was carried out in Erlenmeyer flask (250 cc) and was kept in an incubator shaker device. Sampling for the experiment was conducted one week after the growth of the stems in the medium. Results Based on the results obtained in this study, fresh weight of shoots grown in 10 and 20μM of cadmium chloride increased compared to the control. This heavy metal significantly reduced the amount of chlorophyll a, b and carotenoids, while the total chlorophyll content decreased only at 40μm concentration relative to control. Cadmium significantly increased hydrogen peroxide levels in all treatments. Also, it increased the activity of catalase and superoxide dismutase enzymes. Malondialdehyde increased in all treatment concentrations, but this increase was not significant at 40μm of cadmium chloride. Chloride cadmium reduced the absorption of molybdenum and iron, yet increased the adsorption of manganese and zinc. Discussion The results of this study revealed that the cadmium chloride in lemon balm shoots may have the potential to accumulate compounds such as hydrogen peroxide which, as a messenger molecule, produces antioxidants and can help the plant tolerate stressed conditions. It seems that two concentrations of 10 and 20 μm of cadmium chloride have no toxic effects for the stems and the plant has managed to withstand this tension with defense mechanisms.

Read More Abstract Background and Objectives Induced mutations have been used in a wide range of horticultural crops for more than half a century. This approach has opened up great opportunities for biotechnologists and plant breeders to produce new varieties with several new traits including resistance to disease, and tolerance to adverse environmental conditions or even produce new horticultural characters in some crops. The optimization of dosage of the physical and chemical mutagenic agents for inducing mutations is the first step in using this technique in plant breeding. This study was thus conducted to determine the optimum dosage of two mutagenic agents including gamma radiation as a physical agent and ethyl methanesulphonate (EMS) as a chemical agent in kiwifruit callus in in-vitro conditions. To that end, simple sequence repeat (SSR) markers were used to detect the mutations in the regenerated plantlets. Materials and Methods Having induced the callus in tissue culture media containing naphthaleneacetic acid, 6-benzylamino purine and meta-Topolin, the produced calli were exposed to different dosage of gamma radiation (0, 10, 20, 30, 40, 50 Gy) or different EMS concentrations (0, 0.4, 0.6, 0.8, 1, 1.2 % for 60 min). Then, the callis were transferred to the shoot regeneration media. The number of dead calli, and the size of live calli as well as the number of shoots generated in live calli were then recorded. Afterwards, the regenerated shoots were transferred to elongation media, and the DNA was extracted. The SSR markers analysis was followed on the randomly selected plantlets to detect the SSR markers using a Genetic Analyzer. Results As expected, with increasing the dosage of gamma radiation, the number of dead calli increased and the callus growth was thus retarded. The lowest number of dead calli was, indeed, detected in the control and the 50 Gy dose resulted in the highest number of dead calli. The morphology of shoots generated in the treated calli and the SSR marker analysis showed that 50 Gy dose could be used as the optimum radiation dose to produce an acceptable mutation rate in kiwifruit callus. In turn, EMS as a chemical mutagenic agent showed a different pattern in increasing the number of shoots regenerated with increasing dosage, however the same trend happened with gamma radiation in the number of dead calli and live callus size. Considering the effect of EMS on callus and the number of shoots regenerated per callus, a treatment with 1% EMS for 60 min can be used to produce a high density mutant population in kiwifruit. The highest mutation rate (12.5 %) was observed in the 50 Gy gamma ray treatment with changes in 12 pairs of SSR markers among 25 loci. For EMS, this value was 2.6 % in 1% concentration treatment. Discussion To detect the induced mutations using Gamma ray and EMS, the SSR markers can be applied. However, for the EMS mutation detection, more studies are needed to evaluate the efficiency of other techniques such as single nucleotide polymorphism (SNPs).

Read More Abstract Background and Objectives In recent years, dust has been identified as a major source of adverse environmental and agricultural effects in west and southwest of Iran. Reducing light penetration, decreasing photosynthesis, changing stomatal performance, and reducing flowering are some of the adverse effects of air pollution on plants. Strawberries have a shallow root system, high leaf area, and are sensetive to drought stress. Kurdistan province is the major producer of strawberries in Iran. Since in Kurdistan dust occurs mostly in the spring and during the strawberry flowering and fruit production, we investigated the interaction effect of dust and drought stress on some physiological characteristics of two strawberry cultivars (cvs. Paros and Queen elisa) during 2013-2014. Materials and Methods The experiment was conducted based on a completely randomized design with four treatments (control, -1.2 MPa soil water potential as drought stress, dust and dust + drought stress) and three replications. In this experiment, leaf relative water content, membrane stability index, proline, soluble carbohydrates, lipid peroxidation, hydrogen peroxide, peroxidase and ascorbate peroxidase activity were evaluated. Results Results showed that relative water content and membrane stability index decreased in response to dust and drought in both cultivars. Also, the amount of malondialdehyde, hydrogen peroxide, and activity of antioxidant enzymes increased by dust and drought treatments. Most increase was obsereved in the proline and total soluble carbohydrates, hydrogen peroxide, malondialdehyde, and the activity of antioxidant enzymes was obtained by dust + drought stress. Drought stress + dust had the highest levels of POD and APX. There was a significant difference between the two cultivars, so that Paros cultivar had higher activity of POD and APX compared to queen elisa. Dust and drought treatments reduced shoot and root dry weigth in both years and both cultivars. Discussion Probably due to the stress caused by dust, the total leaf soluble sugars increased in the first year. In this condition, the plant can continue to absorb water from the soil by increasing the soluble carbohydrates through osmotic regulation. In the second year, the long-term reduction in photosynthesis led to a reduction in total soluble carbohydrates, due to prolonged exposure to dust or damage to light absorption pigments resulting from dust accumulation and its negative effects on stomatal function. Dust and drought stress decreased relative water content (RWC) through decreasing water absorption by the strawberry plants, and the decrease of water led to the increase of H2O2. With the increase of H2O2 in dust and drought stress, membrane lipid degradation increased and led to the increase of malondialdehyde (MDA). The increase of MDA indicates cell wall destruction, which is accompanied by the decrease of memberan stability index MSI). MDA increased in all treatments in Queen Eliza cultivar, which could indicate the higher sensitivity of this cultivar to dust and drought stress. In this study, Paros showed higher antioxidant enzyme activity accompanied with lower H2O2 content and consequently a lower MDA, thus, more resistance to drought and drought stress. In conclusion, dust had negative effects on evaluated characteristics, and drought stress exacerbated these effects. Adverse effects of dust and drought were observed on dry matter production in both years. Reducing the dry matter of the roots and the shoot might be due to increased free radicals, reduced leaf water content and, consequently, loss of photosynthesis of the leaf.

Date is an important horticultural fruit in Iran mainly produced in southern regions. Barhee is considered one of the most important date palm cultivars. The fruit of cv. Barhee can be harvested and consumed at different maturation stages including Khalal, Rutab and Tamar. In the present study, the biochemical alterations of date fruit cv. Barhee during development and ripening stages were studied. Materials and Methods The experiment was carried out in a randomized complete block design with three replications. Sampling was done at Date Palm Research Institute Collection Orchard located in Ahvaz. First, fruits were harvested in five different developmental stages including late Kimri, Khalal, mainly Khalal, mainly Rutab, fully Rutab and Tamar. The fruits were then transferred to Quality Analysis Lab of Department of Horticultural Science at Shahid Chamran University of Ahvaz. Selected uniform fruits were used for the analysis of different phytochemical compounds. Chlorophyll, carotenoid, starch, soluble sugars, phenolic compounds, flavonoids and antioxidant capacity were analysed using spectrophotometric methods. The type and concentration of sugars, organic acids, and phenolics were assessed using HPLC. Also, vitamin C content was determined by titration againt DCIP dye. Results The results showed that soluble sugars increased significantly during ripening stages, and glucose and fructose were the main detected sugars for Barhee cultivar. The amount of sucrose was negligible. The acidity of the fruit increased at initial growth stages and then decreased gradually till Rutab stage. At the end of ripening process and at Tamar stage, the amount of organic acid content increased to 2.45 mg/g FW. Among the detected organic acids, acetic acid was dominant in Tamar stage, while in the earlier stages malic acid was considered the main organic acid. Other major investigated compounds, including flavonoids, chlorophylls, carotenoids, starch and vitamin C decreased during the development. Different compounds including p-cinamic, gintistic, gallic, chlorogenic, vanilic, caffeic, syringic and p-cumaric acids were detected by HPLC evaluation of phenolic acids. Among them, gallic acid was the dominant, and the trend was incremental during the development of fruit. Discussion Comparing the current results with published data for other date cultivars shows that there are considerable differences in biochemical compounds among different cultivars. In this cultivar, the increasing trend of accumulation of soluble sugars, organic acids, phenolic compounds and antioxidant capacity during ripening stages from late Kimri to Rutab and Tamar stage makes the fruit an important source of essential nutrients.

Olive is an economically important species of the Mediterranean area, so understanding the mechanisms by which olive plants face drought stress under environmental conditions is essential for the improvement of olive yield and oil quality. Olive (Olea europaea L.) is one of the fruit trees which become important in the Iranian fruit industry at the near future. It seems that olive tree has potential for resistance to drought conditions of semi-arid regions of Iran. There is a high production potential for olive tree in many regions of Iran. Regulated deficit irrigation is an optimizing strategy under which crops are allowed to sustain some degree of water deficit and yield reduction. During regulated deficit irrigation the crop is exposed to certain level of water stress either during a particular period or throughout the growing season. The main objective deficit irrigation of is to increase water use efficiency (WUE) of the crop by eliminating irrigations that have little impact on yield, and to improve control of vegetative growth (improve fruit size and quality). The resulting yield reduction may be small compared with the benefits gained through diverting the saved water to irrigate other crops for which water would normally be insufficient under conventional irrigation practices. This study was aimed to investigate the effect of regulated deficit irrigation regime on vegetative and pomological characteristics and yield of table olive konservolia cultivar in field condition. Materials and Methods This experiment was conducted in Dallaho Olive Research Station (Geographical characters was longitude of 45˚, 51΄ E and latitude of 34˚, 30΄ N and the height of sea level 581m) located in Kermanshah province. An experiment was used based on a randomized complete block design with three replications. Adult table olive Konservolia cultivar was uesd. Each experiment unit consists of Three trees. Vegetative and reproductive traits were evaluated according to I.O.O.C. descriptors. Six irrigation regimes including of full irrigation (as control), regulated deficit irrigation (100% of full irrigation during growing season plus 25% irrigation during pit hardening, regulated deficit irrigation (100% of full irrigation during growing season plus 75% irrigation from Start pit hardening to harvesting), irrigation in three stage (before flowering, pit hardening and before Harvesting , 60% of full irrigation (continuous deficit irrigation) and no irrigation (Rainfed). To elevate the effect of irrigation regimes, some growth vegetative traits measured at the end of growth season including current-season shoot growth and current-season shoot diameter as well as some fruit traits including fruit and oil yield, fruit weight, fruit length and diameter, pulp fresh and dry weight, fruit moisture percent, pulp percent were measured. Collected data were analyzed with SAS program. Results Obtained results showed that the highest fruit yield, oil yield, fruit weight, fruit diameter, pulp fresh and dry weight, pulp percent were observed at full irrigation and regulated deficit irrigation (100% of full irrigation during growing season plus 25% irrigation during pit hardening, but the lowest one found at Rainfed. There was no significant difference between deficit irrigation in three stage and 60% of full irrigation (continuous deficit irrigation) in fruit yield, oil yield, fruit weight, fruit diameter, pulp fresh and dry weight. The water use efficiency of fruit yield of T4 and T2 was higher than 100ETc and other treatments. Overall, the results showed that RDI during fruit pit hardening could increased water use efficiency, whitout reduce fruit yield, oil yield, fruit weight, fruit diameter and pulp fresh and dry weight. Discussion In the arid and semi arid as well as sub-tropical regions, water shortage is a normal phenomenon and seriously limits the agricultural potential.Therefore, under irrigation or rain-fed conditions, it is important for the available water to be used in the most efficient way. Softening, chilling injury and the incidence of different diseases are limiting factors for postharvest life of persimmons fruits during cold storage. Ultraviolet-C (UV-C) light radiation has recently suggested to controlling decay and reducing chilling injury of some fruits. Therefore, this study was conducted to evaluate the effects of UV-C treatments with different doses on alleviating chilling injuries and maintaining qualitative attributes of persimmon fruits during storage.Materials and methodsThe persimmon fruits cv. `Kashan´ were harvested from a commercial orchard near Kashan city. UV-C treatments was carried out at 0 (as control), 5 minutes irradiation from 15 and 30 cm distance with 3.2 and 0.8 kJm-2, and 10 minute irradiation from 15 and 30 cm distance with 16.4 and 4.1 kJm-2. After 1, 2, 3 and 4 months, fruits were removed from storage and chilling injury indices, polyphenol oxidase (PPO) activity and some quality attributes were measured. ResultsThe effect of UV-C treatments on weight loss, chilling index, electrolyte leakage, lipid peroxidation and PPO activity was effective during storage. High percent of weight loss (6.84) and chilling symptom (40) was observed in irradiated fruits with 16.43 kJm-2 and low level of mentioned parameters was seen in 3.2 kJm-2 treatment. High level of electrolyte leakage (80.49) and lipid peroxidation (1.78 nM g-1 FW) were detected in treated fruits with 4.1 and 16.43 kJm-2 and low level of above parameters were observed in non-treated and treated fruits with 3.2 kJm-2. Application of UV-C with high dose increased PPO activity (9.7 u gr-1 FW) in comparison to other treatments during storage. DiscussionOur results suggest that application of UV-C irradiation in low level after harvest could increase postharvest life of `Kashan´ persimmon by controlling chilling injuries and preserving fruit quality. However, it is worth mentioning that chilling symptoms of persimmon was reduced by low doses of UV –C irradiation. In fact, UV-C treatment with high doses caused high percentage of injured fruits. In conclusion, the use of UV-C radiation in controlling chilling injuries and maintaining quality of persimmons fruits offers an interesting research area and considering the low risk of the treatments.

In order to increase the quality of flowers, all plants should be fertilized correctly. Potassium is one of the elements that play a very important role in the quality and performance of ornamental flowers. Nano fertilizers increase nutrient efficiency, reduce soil toxicity, minimize the potential negative effects associated with overdosage, and are an effective step toward achieving sustainable and environmentally sustainable agriculture. The present study was undertaken to investigate the effect of potassium and nano-potassium fertilizers on the physiological and biochemical characteristics of Asiatic Lilium Hybrid cv. Tresor. Materials and Methods In the current study, the effect of foliar spraying of different concentrations of potassium (0, 1, 2, and 4 mM) and nano-potassium (0, 0.5, 1, and 2 mM) was investigated on qualitative and quantitative traits of lily cut flower "Tresor" at the four stages of pre-harvest. The traits included a number of buds and leaves, bud length, bud diameter, stem length, stem diameter, length and width of leaves, fresh and dry weight of stems and leaves, flower longevity, peduncle length and diameter, dry matter percent of stems and leaves, ionic leakage, chlorophyll index and content of carotenoids, Flavonoids, anthocyanins, and chlorophyll a, b, and total. Results The result showed that there are significant differences between different spraying treatments and the number of buds, stems fresh weight, and ionic leakage. The highest stem fresh weight was observed by the 0.5mM nano-potassium, while the largest number of buds, the highest cells stability, and the lowest ionic leakage were achieved by the 4mM potassium. In regard to the traits of chlorophyll content, Carotenoid, Anthocyanin, and Flavonoid, there were no significant differences between the treatments of potassium and the nano-potassium. Also, the 4mM potassium and the 2mM nano-potassium showed the longest vase life. Considering the fact that the number of buds in the Lily is the most important trait, the 2 mM nano-potassium is recommended as the best treatment for spraying to feeding potassium. Discussion The foliar application of potassium and nano-potassium enhanced stem fresh weight and vase life of cut flower lilies and reduced ionic leakage compared to the control. Also, the largest number of buds was achieved by the 4 mM potassium. These results are in agreement with previous studies on carnation, Narcissus, and lily. Potassium is vital to many plant processes. Potassium plays a major role in the transport of water and nutrients throughout the plant in the xylem. Potassium is required for every major step of protein synthesis. Also, the enzyme responsible for the synthesis of starch (starch synthetase), which is activated by K. Briefly potassium, plays signiﬁcant roles in enhancing the crop quality. High levels of available K improve the physical quality, disease resistance, and shelf life of crops.

Recently, the remediation of these metals has been considered in plants. Plant in vitro culture is a key tool in phytoremediation research. In general, Plant tissue culture and in vitro selection techniques are used to increase the tolerance and the accumulation of heavy metals which have been reported in numerous plant species and populations. The application of ornamental plants for phytoremediation in metal-contaminated soil is a new alternative. Ornamental kale is an important bedding plant in many landscapes of cold temperate regions. The present study was aimed to evaluate the Pb resistance and remediation of Brassica OleraceaVar. Acephala affected by in vitro culture as a bedding plant. This experiment was carried out in 2016 at Arak University. Materials and Methods In this study, the stalk explants of Brassica OleraceaVar. Acephala was used for callus induction. Callus and plantlet were exposed to different concentrations of lead in the media. The Callus induction medium was MS (Murashige and Skoog) supplemented with 0.1 mgL-1 2,4-D and BA. The regeneration medium was MS medium supplemented with 0.5 mgL-1 NAA in combination with 1 mgL-1 BA. The rooting medium was half-strength MS medium without plant growth regulators and sucrose. All of the media containing 30 gL-1 sucrose, 7 gL-1 agar, and pH were adjusted to 5.8. All of the regeneration stages from callus induction to rooting and the acclimatization stages were done by different concentration of Pb(NO3)2 (0, 10, 25, and 50 mgL-1). The traits, such as callus browning, fresh and dry weight, callus formation, survival rate, root and shoot length, leaf number, and the lead accumulation, were evaluated during various stages of callus regeneration and rooting in plantlets. A Completely Randomized Design (CRD) arrangement with three replications was used in this experiment. Data were analyzed using the ANOVA procedure of SAS statistical software (version 9.2). Results Based on the results, not only did different concentrations of lead not have the toxic effects but also they had a stimulation effect in some concentrations on callus growth and regeneration. Lead concentrations increased leaf number and length of shoots in the regenerated ornamental kale. Lead accumulation in cultures was increased by increasing the concentration of lead in the medium at all stages of the regeneration. The highest accumulation of Pb was obtained 2395.7 ppb in callus treated by 25 mg L-1 Pb in media. Discussion The results of the present study showed the occurrence probability of the somaclonal variation due to 2,4-D supplemented in the media. In the regenerated plant, results revealed that the growth traits and Pb uptake were affected by Pb concentrations. Some in vitro derived plantlets showed an increase in uptake potential of lead in their organs (6.1 to 2.7 times compared to the control) and that it is remarkable and suitable for phytoremediation studies.

Read More Abstract Background and Objectives Persian walnut (Juglans regia L.) belongs to the family of Juglandaceae and is one of the most important nut crops in Iran. Until recently, all of the fruitful walnut trees grown in Iran are seedling originated and thus, they exhibit a signiﬁcant variation, especially in nut and kernel characteristics. Therefore, the identiﬁcation of the promising genotypes based on the phenotypic traits is essential in breeding programs. The aims of the current work were to evaluate 119 Persian walnut trees in different western regions of Iran based on nut and kernel characteristics, and then to detect genetic relationships among desirable genotype using ISSR marker. Materials and Methods In this research, 119 Persian walnut genotypes collected from different parts of the west of Iran were evaluated based on nut characteristics. Based on the primary evaluation, 30 superior genotypes that had desirable characters in nut and kernel properties, such as nut and kernel weight, kernel percentage, kernel removal from the nut, kernel plumpness, and kernel ﬁlled were selected and then, the genetic variation among them was evaluated using 10 ISSR primers. Results The preliminary evaluation of 119 walnut genotypes illustrated that most of the evaluated genotypes showed high variability for the measured traits related to nut and kernel. Among the nut and kernel characters, the kernel shriveling showed the highest coefficient of variation (114.02%), while the lowest CV was related to the nut diameter (8.68%). Among walnut genotypes, both nut and kernel weight varied from 7-19.8 g and 2.8-9.20 g, respectively. The genetic relationship among 30 promising genotypes with ten ISSR primers indicated a considerable level of variability. All of the ISSR primers were polymorphic and produced a total of 79 alleles among the 30 genotypes, which 77 alleles were polymorphic. The size of the ampliﬁed fragments ranged from 200 bp to 1700 bp. The number of the observed alleles for each locus ranged from 3 (HB10) to 13 (UBC-807), with an average of 7.7 alleles per locus. Polymorphic index content (PIC) was observed to be highest (0.82) in the UBC-807 locus, while the HB10 locus had the lowest value (0.51) with an average of 0.75 among ISSR locus. The Jaccard’s genetic similarity coefficient ranged from 0.31 to 0.85 among the genotypes. The cluster analysis performed based on ISSR data using UPGMA, divided the genotypes into seven major groups. Discussion Finally, the results of this study showed that there was high variability among walnut genotypes in terms of quantitative and qualitative characteristics of the nut and the kernel. In this study, the average nut and kernel weight among walnut genotypes varied. The difference in nut and kernel properties of germplasm under the same geographical conditions may be the result of genotypic factors. The results indicated that some genotypes (16, 4, 24, 26, 11, and 17) had desirable traits in the nut and kernel. Moreover, the Persian walnut is of most important horticultural crops grown in Iran. Therefore, these genotypes can be propagated according to the vegetative methods and used for commercial cultivation or utilized for traditional breeding and advanced biotechnology studies to achieve superior progenies.

Improving Quality and Antioxidant Characteristics of Pulp and Peel of Citrus During Storage by Pre-Harvest Spray of Calcium Chloride on Fruit

Pre-harvest foliar application of calcium chloride (CaCl2) is an efficient and safe strategy to maintain or improve the fruit quality during storage. Also, calcium significantly increases the membrane and cell wall density and reduces the physiological disorders of the fruit. Thus, the aim of this study was to evaluate the effect of pre-harvest CaCl2 sprays of citrus trees at three fruit development stages on improving fruit quality and antioxidant capacity during storage. Materials and Methods To conduct this research, citrus trees of three different varieties (‘Thomson’, ‘Moro’ and ‘Page’) were sprayed with different concentrations of CaCl2 (0, 1, 2 and 4%) at three fruit development stages (120, 140 and 160 days after full bloom). After harvesting, citrus fruits were stored at 5°C and 85% relative humidity for 60 days. Then, weight loss and juice percentage, total carotenoids, superoxide dismutase (SOD) and ascorbate peroxidase (APX) activity, and total antioxidant capacity (IC50 content) of the fruits peel and pulp were determined on 0 (harvesting time), 30 and 60 days. Data were analyzed using SAS (Ver.9.1) and significance of the differences between the means was computed using Tukey test. Results Results showed that weight loss percentage in fruit treated with CaCl2 (especially in 2 and 4% treatments) of all three cultivars was significantly less than control during storage. Total carotenoid of pulp of Thomson and Moro cultivars increased with increasing of CaCl2 concentrations compared to control during storage. In Page mandarin, the carotenoid content of pulp was not affected by calcium chloride treatment. Results also showed that SOD activity in peel and pulp of treated fruits was higher than control at harvest time. At the time of harvesting, the peel APX activity of Thomson and Moro cultivars (control) was higher than those treated by calcium. Antioxidant capacity (based on IC50 content) of peel and pulp varied depending on the cultivar and the treatment. That is, the highest antioxidant capacity was determined in ‘Thomson’ at harvest time and in treated fruits with high level of CaCl2. Discussion In the present study, the CaCl2 pre-treatment was sprayed on the citrus trees and the results indicated that pre-harvest calcium chloride significantly decreases weight loss percentage and increases antioxidant capacity, SOD activity and total carotenoid contents in citrus fruits during storage. It seems that the accessibility of sufficient CaCl2 during plant growth has a strong influence on concentrations of bioactive compounds and overall antioxidant properties of citrus fruits. Overall, it is suggested that an appropriate CaCl2 pre-treatment of citrus tree can help improve or maintain pulp and peel quality during storage. It is further suggested, using chlorine-free calcium sources in southern Iran with alkaline soils.

Statistics shows that this fruit tree is cultivated in more than of 102 thousand hectares of territory of Iran. Due to severe water shortages, it is necessary to make use of methods with objectives to reducing the negative effects of drought on plant and to increasing water use efficiency. Previous studies stated that plant growth regulators such as salicylic acid could apply to reducing the adverse effects of environmental stresses. Salicylic acid (SA) influences various physiological and biochemical functions in plants and has diverse effects on the tolerance to biotic and abiotic stress. The objective of this study was to investigate the effects of exogenous SA application on biochemical traits of ‘Konservalia' olive leaf under 100 %, 66 % and 33 % ETcrop by analyzing chlorophyll content, total soluble carbohydrate and proline. Materials and Methods The study was conducted in order to investigate the effect of different levels of water deficit (100, 66 and 33 percent of evapotranspiration (ETcrop)) and four concentrations of salicylic acid (0, 0.5, 1 and 2 mM) on fresh and dry root weight, stem and leaf and leaf number, leaf area, stem diameter, leaf proline, total soluble sugars, chlorophyll content as a factorial experiment based on a randomized complete block design with three replications. Two- year old olive trees (Olea europaea cv. ‘Konservalia’) were used. Seventy- plants were grown in 11 L pots, containing a mixture of field soil (73.2 % sand, 13.3 % silt and 13.5 % clay) and manure. Plants were sprayed with four concentrations of salicylic acid and irrigated every ten days, according to the amount of evatranspiration of plants (ETcrop). Parameters such as leaf proline and total soluble carbohydrates content, chlorophyll content were measured three times at intervals of once every 30 days after treatment. Data analysis was performed using MSTATC and SAS software and mean comparison was done by Duncan's multiple range test at 5 and 1 % probability Results The results showed that leaf proline and total soluble carbohydrates increased while chlorophyll content (chlorophyll a, b and total) decreased with increasing levels of water deficit. Results also showed that the interaction of salicylic acid and irrigation had a significant effect on fresh and dry root weight, stem and leaf, leaf area and plant height at p Discussion In this study, the role of SA in plant tolerance to abiotic stresses such as heat, heavy metal, and osmotic stress was reported. The results showed that 2 mM SA application alleviated the adverse effects of drought stress (66 and 33 % ETcrop) in young olive trees through the improvement of chlorophyll content, enhancing leaf total soluble carbohydrate and proline content which can lead to osmotic adjustment.

Read More Abstract Background and Objectives Chrysanthemum, due to numerous applications in floriculture and medicinal industries has been considered one of the most important ornamental plants and medicinal plants in the global area. In addition to being used as a cut flower, it also has a special place among potted flowers and gardens. Also, it is used as a medicinal plant with strong healing properties in traditional Chinese medicine and considered to treat eye diseases, headaches, colds, and etc. However, only a few reports have been presented in the field of genetic diversity of Chrysanthemums by morphological and molecular markers. Therefore, there is a need for further identification of germplasm and cultivars of Chrysanthemums in Iran. This study aimed to assess the genetic diversity of 15 cultivars of Chrysanthemums based on morphological characteristics and selection of cultivar adapted to Khorramabad region. Materials and Methods This experiment was carried out in a randomized complete block design with three replications in the area of Beiran-Shahr, Lorestan province in 2017. Twenty-one morphological characteristics were determined from fresh materials. These traits in accordance with the given instructions describing World UPOV Chrysanthemums, were measured. Quantitative measurements were carried out on 10 leaves per cultivars. Some variables including diameter (branches, petioles, discs, buds and flowers) were measured by digital caliper. The traits such as length and width of leaf, plant height, length and width of the tab florets with ruler were measured. Results Based on data analysis most of the traits in cultivars showed significant differences (p≤0.01). Results showed that the correlation between leaf area with leaf width, petiole diameter and number of leaves has a positive and significant correlation with the probability level of 5%. The most significant and negative correlation (r = 0.99) was observed between number of flowers per plant and number of florets. Based on the results obtained from principal components analysis, seven independent and major components with Eigen values greater than one could explain 89 percent of the total variation. The first component, verified 24.4 percent of the total variation. Cluster analysis of cultivars in Euclidean distance of 26.68, was categorized into two main groups. The cultivars "Avadis" and "Norooz3" in the first group were distinguished from the rest of the cultivars with the lowest height. The "Fariba"; "Ashraf" and "Andiea" cultivars were placed in the group and most abundant cultivars. Discussion Based on the results, leaf size, plant height, number of branches and number of flowers in plant traits were most important in recognizing the diversity of cultivars Chrysanthemum. By using some cultivars belonging to the first cluster with less height and cultivars in the second cluster with more leaves and flowers as a parent, a new cycle of Chrysanthemum breeding program can be initiated.

Read More Abstract Background and Objectives Production of pure lines is one of the most important tools due to production of high-yield seeds. Pure lines can be produced in a short time using in vitro techniques and therefore, reduce several years in required time for conventional plant breeding programs. The purpose of this study was investigation of different concentrations of TDZ on production of haploid plants in cucumber ovule culture. Materials and Methods The experimental layout was conducted in factorial arrangement in randomized complete design with three replications in greenhouse and laboratory of Razi university during 2015-2017. Factors were two cucumber genotypes (Esfahani and Beit Alfa) and different concentrations of TDZ (0, 0.01, 0.02, 0.03, 0.04, 0.05, 0.06, 0.07, 0.08 mg/l). Unfertilized ovaries were harvested 1 day before an thesis and they were sliced into 1 mm cross sections under sterile conditions and placed on solid MS medium. Immediately after placing the unfertilized ovary slices of each genotype on induction medium, they were exposed to a thermal shock pretreatment at 35±1°C for 3 days. The first visual structures formed after 3 days in culture. After two weeks of culture, the frequency of embryo formation was recorded. After embryogenesis, the embryos were sub-cultured in medium containing 1.5 mg/L-1 GA3 and finally cultured in medium containing 0.05 mg/L-1 NAA and 1.5 mg/L-1 BAP for organogenesis. Results Results indicated a statistically significant effect at 1% among TDZ concentrations. Genotypes and interaction between genotype and TDZ concentration did not have a significant effect on embryo induction. According to the results of means comparison, M8 medium with the average of 23.33 for Esfahani genotype and 20.66 for Beta Alfa genotype had the highest embryo induction and M1 showed the lowest embryo induction for both genotypes. Ploidy level of calli and embryos were identified by Flucytometry assay. Haploid embryos were transferred to the regenerative medium. The chromosome content of haploid plants was doubled spontaneously in all regenerated plants. Double haploid plants originated from megaspore and they were equivalent to haploid, which relates to homozygosity and can be used directly in breeding program. Discussion The high number of embryos was obtained in high concentration of TDZ (0.08 mg/l). Flucytometry assay is a way to recognizing ploidy level in plant tissues; thus it could be useful in choosing haploid tissue. Spontaneous chromosome doubling may occur via somatic cell fusion, endoreduplication, endomitosis and possibly many other mechanisms. In conclusion, the higher rates of embryo formation frequency achieved and doubled haploids obtained in the present study are promising for future work.

The harmful effects of high salinity on plants can be observed in senescence, necrosis of plant organs and decrease productivity. Major processes such as photosynthesis, protein synthesis, and energy and lipid metabolism affect plants during the beginning and development of salinity stress. Calendula officinalis L. belongs to the Asteraceae family; it is an annual with bright or yellow orange daisy-like flowers which are used for medicinal or culinary purposes. Organic fertilizers develop favorable physical, chemical and biological environment in the soil. They stimulate plant root growth, increase nutrient uptake and soil water-holding capacity, decrease evaporation from the soil and surface water runoff, facilitate drainage, regulate soil temperature and provide a rich substrate for soil microbes. Materials and Methods This study was conducted to determine the effects of soil salinity and organic amendments on some growth characteristics, absorption of nitrogen, phosphorus and potassium and peroxidase enzyme activity in pot marigold plant (Calendula officinalis L). In a factorial experiment and completely randomized design (CRD), five levels of organic amendments (control, 0.5 and 1 g.L-1 algae extract, 20% v/v of pot volume cow manure and 20% v/v of pot volume vermicompost) and three levels of salinity (3.5, 7.5 and 10.5 dS.m-1) with three replications per treatments were applied. In this experiment, media without organic amendment were considered as control. Results Results showed that increasing soil salinity levels progressively decreased the growth characteristics and nutrients uptake. The maximum leaf number, fresh and dry weight of flowers and nitrogen, phosphorus and potassium absorption were obtained in cow manure treatment and EC = 3.5 dS.m-1, While the highest flower diameter was observed in sea algae treated plants (0.5 g.l-1) in media with EC = 7.5 dS.m-1. Salinity causes growth reduction due to the low osmotic potential of the medium and by a specific ion effect as a secondary cause in several vegetable crops. Discussion The results of the present study showed that the organic media can improve leaf number, crown diameter and fresh and dry weight of flowers. This can be due to increased media moisture storage and enhanced nutrient absorption. All treatments significantly increased growth characteristics of pot marigold compared to control. The positive effect of cow manure to nutrients uptake could be because of rich N, P, K compared to the others. According to the results, in normal conditions, pot marigold plant could tolerate salt stress until 7.5 dS.m-1 but by suitable media culture its threshold tolerate will be increased up to 10.5 dS.m-1. Also it was revealed that cow manure compared to other treatments could increase plant tolerate to salt stress and growth characteristics.

On the other hand, the combined application of chemical and bio-fertilizers can improve yield and quality of medicinal plants. Basil (Ocimum basilicum L.), a member of Lamiaceae family, is an annual plant and is widely used as a vegetable and as an aromatic plant. Considering the importance of medicinal herbs and the global trend for their production and reproduction in sustainable and low-input agricultural systems as well as a lack of research on the response of Basil's medicinal plant to the combined nutritional systems (chemical and biological), the aim of this study was to compare the effects of various nutritional systems with chemical and biological nitrogen and phosphorus fertilizers as high-quality nutrients on some quantitative and qualitative characteristics of Basil. Materials and MethodsThis research was conducted as a factorial experiment based on a completely randomized design with three replications at the Research Center of Faculty of Agriculture, Shahed University, Tehran, in 2014. The experimental factors included nitrogen and phosphorus fertilizer combinations, 100% chemical based on a soil test, 100% biological (for nitrogen, Azotobacter crocococcus and Azospirillium lipophorum, for phosphorus, and Pseudomonas fluorescence) and combining a system of 50% chemical fertilizers with biofertilizers. To provide nitrogen fertilizer from the source of urea containing 46% pure nitrogen and phosphorus fertilizer were used triple superphosphate containing 46% phosphorus oxide (P2O5). Biofertilizers used to provide nitrogen are a mixture of Azotobacter crocum (Azeto-5 strain) and Azopyrillium lipophorum (93 strain), and for phosphorus from the mixture of Pseudomonas putida (P-168 strain) and Pseudomonas fluorescence (P-169 strain), all of which have a concentration 107 CFU/ml provided by the National Institute of Soil and Water Research of Iran. ResultsThe results showed that different combinations of nitrogen and phosphorus fertilizers and also the interaction between these two had a significant effect on plant height, the number of leaves and shoots yields, percentage, and yield of essential oil. The highest number of leaves per plant was found in treatments N2 × P4 (100% chemical nitrogen with combined of bio-fertilizer and 50% chemical phosphorus) and N4 × P4 (combin of bio-fertilizer and 50% chemical hosphorus combined with bio-fertilizer and 50% chemical nitrogen) respectively with the mean of 54.72 and 54.44 leaves per plant. Interaction of nutrition systems showed that the highest leaves and essential oil yield was in combination N4 × P4 respectively with the average 596.33 kg.ha-1 and 6.56 lit.ha-1. The uses of combination system of nitrogen and phosphorus increased the mean composition of the oils constitutive such as Linalool, Carvacrol and Eugenol compared to control. DiscussionThe results of this study indicate that the use of bio-fertilizers phosphorus and nitrogen, either alone or in combination with 50% chemical fertilizer to improve the quantity and quality of basil had a positive impact. They can, instead of continuous use of chemical fertilizers, be used to optimize the biological inputs for sustainable agriculture and reduce the pollution caused by the use of nitrogen and phosphorous fertilizers.

Olive is an economically important species of the Mediterranean area, so understanding the mechanisms by which olive plants face drought stress under environmental conditions is essential for the improvement of olive yield and oil quality. Drought is one of the most important environmental stresses that affect plant growth and development negatively. This study was aimed to investigate the effect of drought stress on some morphological characteristics of some superior local olive genotypes in pot condition was conducted in Dallaho Olive Research Station (geographical characters was longitude of 45˚, 51΄ E and latitude of 34˚, 30΄ N and the height of sea level 581m) located in Kermanshah province. In order to select drought resistant or tolerant local olive genotypes an experiment in pot conditions was conducted. Seven olive genotypes (D1, Dd1, GW, Ps1, Bn3, Bn6 and Ds17) used as plant materials. Pot experiment based on completely randomized design with three replications and two factors (irrigation regimes and genotypes) was conducted in 2015. Irrigation Ttreatment were including of 100% (control), 80%, 60% and 50% of evapotranspiration respectively. Some vegetative characteristics such as plant height, trunk diameter, leaf number, leaf, shoot, and root fresh and dry weight were recorded. Collected data were analyzed with MSTATC program. Obtained results showed that Bn3 genotype had the highest height and D1 had the lowest height. Bn3 genotype had the highest number of leaf and D1 had the lowest but D1, Dd1 and Gw were in a statistic class. Genotypes showed significant differences in leaf fresh and dry weight so that Bn3 genotype had the highest leaf fresh and dry weight and D1 had the lowest. Genotypes indicated significant differences in pot in shoot and root fresh and dry weight so that Ds17 genotype had the lowest shoot fresh and dry weight and Gw had the lowest root fresh and dry weight. and Bn6 had the lowest root dry weight/ shoot dry weight ratio. irrigation treatments significant differences in recorded vegetative and physiological characteristics so that 100 percent irrigation treatment increase relative water content percent (RWC), total chlorophyll. among olive genotypes Bn3 and Ds17 had the highest, relative water content percent (RWC), total chlorophyll and mda less produced. The results indicated that olive genotypes had different resistance to drought and vegetative growth status of Bn3 and Ds17 genotypes were better in drought stress conditions. It can be concluded that Bn3 and Ds17 genotype had the relatively high resistance to drought stress.

Read More Abstract Background and Objectives Gladiolus grandiflora Hort. belongs to Iridaceae family and it is one of the most important cut flower and bulb in the world. Fragrance, long vase life and color variation make it different in floriculture. Breeding and introduction of new cultivars with innovation in ornamental are important. The aim of this research was of introducing new hybrids of gladiolus through hybridization. Four cultivars in Amsterdam, White prosperity, Advance red and Rose supreme were chosen for hybridization. Materials and Methods Progenies were evaluated in a randomized complete block design with three replications. The Research was conducted in Ornamental Plants Reserch Center (OPRC) in Mahallat from 2015 to 2016. Quantitative traits included leaf length, leaf width, stem diameter, number of leaf, days to germination, weight and diameter of cormelet. Finally, statistical parameters such as coefficients of correlation, mean, standard deviation, coefficient of variability, principal component analysis and cluster analysis were estimated. Results Analysis of variance among progeny (hybrids) showed that the progeny has significant differences (p≤0.01) in all traits and a wide variation in progenies was observed for all traits. NIOP9 progeny in leaf length, leaf width, stem diameter, number of leaf, days to germination, weight and diameter of cormelet was superior progenies compared to other cormelet. Lilcewise, progeny NIOP4 with 2.49 per seed was superior to cormelet. Correlation coefficients among the different progenies revealed that the most positive and significant correlation was between the number of leaves and crown diameter (r= +0.93) and the lowest level (r= -0.17) of correlation was found between the number of days to germination and number of cormelet. The highest coefficient of phenotypic variation of 57.17% was obtained for the number of cormelet and the lowest of 11.15% for the number of days to germination. The highest heritability in traits was estimated as 98.46 and 96.46% for the number of cormelet and cormelet diameter, respectively and the lowest as 12.90% for the crown diameter. Discussion All traits showed significant differences in progeny. These results indicate that the major traits in the phenotypic and genetic diversity coefficient were very low, indicating that they had less environmental effects, since the progeny was cultivated under similar and controlled conditions (greenhouse).
