Read More Abstract Background and Objectives Determination of genetic diversity level is fundamental for identification of desirable parents to be used in different breeding programs, and molecular markers have been succssfully taken for the analysis of genetic diversity in various crops. The objective of this study was to inversitgate genetic diversity among 28 genotypes of barley using root and shoot characters and 14 ISSR primers through two separate experiments (greenhouse and molecular experiments). Materials and Methods The greenhouse experiment was carried out as randomized complete block design with two replications in the research greenhouse of Faculty of Agriculture, University of Tabriz, Iran. The traits measured in the greenhouse were root dry weight, root volume, shoot dry weight and ratio of root to shoot dry weight. The molecular experiment was conducted to study the diversity of barley genotypes using 14 ISSR primers. The polymorphic information content and marker index were calculated for each ISSR primer. Results Analysis of variance showed significant differences among the genotypes under study for all traits. According to the mean comparisons, genotypes 3, 5, 9 and 20 had higher mean in terms of all characters. The cluster analysis, based on Ward's algorithm and Euclidean distance, grouped genotypes in four clusters. Group 3 had the highest mean in terms of all of the studied characters. Out of 14 ISSR primers used, 11 primers generated scorable and appropriate banding pattern. A total of 559 polymorphic bands with 80-3000 bp were produced. Polymorphic information content was estimated to be between 0.116 and 0.252 with the average of 0.187. The marker index ranged from 3.528 to 27.972 with the mean of 9.704. Classification of the studied barley genotypes was conducted by molecular data using neighbor joining algorithm based on distance coefficient of number of differences, which assigned the genotypes into four groups. There was a concordance between the grouping of genotypes based on molecular data and the characters in the greenhouse, but this concordance was not complete. Association analysis of ISSR markers with measured characteristics of barley genotypes showed that ISSR1 and ISSR5 had significant relationship with most of the root and shoot traits. Discussion Highgenetic diversity was observed among barley genotypes with respect to root and shoot traits. The results showed that ISSR primers have the ability to separate barley genotypes from each other. Also, it seems that ISSR markers under study can be used in marker assisted selection of barley genotypes in breeding programs.

The reduction of genetic variation in tomato through domestication and breeding has resulted in the need for conservation and utilization of all existing genetic resources. Local populations are among the most important genetic resources. They are often heterogeneous and composed of different genotypes which are mostly homozygous. Therefore, assessment of their diversity requires a different approach of common methods. The objective of this study was estimating the phenotypic variation among and within the tomato local populations of Kalaybar area using Nei’s genetic diversity index. Materials and Methods Plant material consisted of 8 tomato local populations collected from Kalaybar area in the East Azerbaijan province.The experiment was carried out as a randomized complete block design with three replications. A total of 36 plants of each population in three blocks were planted with the distance of 140 cm between rows and 80 cm between plants. Seven traits, including fruit yield and some of its related traits, was measured. All continuous traits were divided into three classes with equal width and the frequency of classes was used to estimate total diversity index (HT), phenotypic diversity within each population (Hs) for each trait. Then, mean diversity index () for all traits, proportion of phenotypic diversity among populations (GST) for each trait, and mean diversity index for each population across all traits () were calculated. Results The total diversity index (HT) for each trait ranged from 0.46 to 0.53 with an average of 0.49. Most of values were around the mean. All traits had relative high value of mean diversity index ( > 0.40). The mean phenotypic diversity among populations (GST) was 0.10. For the majority of traits, little diversity was observed among populations. The greatest value of GST (0.16) was relevant to the mean of fruit weight per plant. Most of population had similar magnitudes of mean phenotypic diversity within each populations () across all traits (0.42 to 0.49). The fruit yield (0.51) and mean of fruit weight per plant (0.51), the number of fruit per plant (0.56), the number of node on stem (0.54), the number of floret (0.54), the number of days to flowering (0.44), and the number of days to fruit (0.54), had the highest diversity index (Hs) values in Gharavanloo, Rostamkandi, Sheykhlan-sofla and Kalantar, Sheykhlan-olya, Alisharloo, and Sheykhlan-olya populations, respectively. Discussion The results showed a relatively wide phenotypic diversity present in tomato local population of Kalaybar area for most of the studied traits. The highest diversity was within the populations and there was little variation among the populations. The traits that contributed the most to within populations’ heterogeneity differed from population to population. However, the Sheykhlan-olya population had the highest fruit yield (4.83), the number of fruits (18.4), the number of nodes on stem (20.7), the number of flowers (83.5), and it was concluded that population could be used as a genetic source for future tomato breeding programs in Kalaybar region.

They are rich in organic matter and nutrients, and thereby can be used in agriculture as an organic fertilizer. Their application will lead to the production of healthy and qualitative foods. To the best of our knowledge, so far the effect of application of marine algae has not been evaluated on saffron qualitative parameters. Therefore, the aim of present study was to investigate the effect of algae in different levels of mother corm weight on qualitative indices of petals and stigmas in saffron. Materials and MethodsTo investigate the effect of growth promoting fertilizer containing seaweed extract and mother corm weight on qualitative parameters in petals and stigmas of saffron (Crocus sativus L.), an experiment was conducted at Research Field of College of Agriculture at University of Birjand in 2017. The experimental factors included seaweed extract (0, 15 and 30 l ha-1) and corm weight [0.1-4 (small), 4.1-8 (medium) and 8.1-12g (large-sized)], which were tested in a RCBD with three replications. ResultsThe use of algae extract had a significant effect on the petal yield, carotenoid content and antioxidant activity of petals (antioxidant and anthocyanin content) and quality of stigma (picrocrocin, safranal and crocin content). The highest amounts of dry petal yield (0.027 g plant-1), carotenoid content (1.83 mg g-1 FW), anthocyanin (24.49 mg 100g-1 DW), antioxidant (29.80%), safranal (42.43), crocin (198.78) and picrocrocin (74.57) were obtained when seaweed extract was used at the rate of 30 l ha-1. However, no significant difference was found between the two levels of algae extract in terms of all mentioned parameters. The lowest values of petal yield (0.023 g plant-1), carotenoid (1.04 mg g-1 FW), anthocyanin (22.29 mg .100 g DW), antioxidant (25.54%), safranal (40.61 λ), crocin (176.64 λ ), and picrocrocin (72.60 λ ) were obtained from control treatment. Mother corm weight also exerted a significant effect on antioxidant activity, petals anthocyanin content and safranal content in stigma and the highest of them were gained when large-sized mother corms were planted. The interaction effect of experimental factors was not significant on measured parameters. DiscussionOverall, the results of this study indicated the beneficial effects of seaweed extract on improving biochemical traits and effective ingredients, which is appropriate for organic production of saffron. The positive role of algae on plants is related to providing micro- and macro-nutrient as well as production of biologically active biotical substances against plant-infecting pathogens. In addition, they have a significant role in improving soil physical and chemical properties, which lead to better growth and development of the plants. Better quality of saffron produced from larger mother corms may also be related to more nutrient reservoir in such corms.

In addition to plant genetics, environmental variability plays a key role in plant yield. Drought is one of the most important environmental stresses that reduce the productivity of sunflower and other crops. Understanding the nature of phenotypic traits that improve performance under stress conditions and identifying the complex physiological and genetically mechanisms is fundamental to plant stress. Improving drought tolerance and increasing water productivity in plants is one of the most important goals of the breeders. Gaining knowledge of the amount of phenotypic and genetic parameters in the plant species for genetic and plant breeding studies is very important for the exact selection of the genotypes according to the objectives of the breeding programs. Understanding the nature of morphological, physiological, genetic and molecular mechanisms under the influence of drought tolerance can play an important role in the development of drought tolerant genotypes for cultivation in arid and semi-arid areas. The main aims of this study included investigation of genetic diversity of sunflower germplasm (to understand the relationships between morphological traits and their heritability) and accurate identification of the most important traits that affect the sunflower yield in each of the normal conditions and drought stress. Materials and MethodsAn experiment was conducted in a randomized complete block design with three replications in normal and drought stress conditions in 2014 growing season on 12 sunflower genotypes in a research field of Islamic Azad University, Karaj Branch. Each block consisted of 12 plots and each plot contained five rows. Each row was 5 meters length with 60 cm spacing, and plant spacing on rows was 20 cm. The studied traits included grain yield, seed length, seed width, seed diameter, leaf length, leaf width, plant height and stem diameter. ResultsThe results of combined analysis of variance confirmed the significant diversity among genotypes for most traits. Grain yield showed higher values for parameters of genetic efficiency, genotypic and phenotypic variation coefficients under drought stress and non-stress conditions compared to other traits. While the highest percentage of heritability in the normal condition belonged to grain yield, it belonged to plant height in the stress conditions. Plant height, leaf width, stem diameter and leaf length showed the highest change in terms of traits in stress conditions compared to normal conditions. Dendrograms derived from cluster analysis classified genotypes into three groups based on studied traits under both drought stress and non-stress conditions. According to the cluster analysis, under drought stress condition, the Sor and Progress genotypes were identified as tolerant genotypes and, in contrast, genotypes of Favorite, Record, SHF81-90, Master and Lakomka were recognized as sensitive genotypes. DiscussionIn the combined analysis of variance the significance of the environmental effect shows that drought stress has significantly affected all traits. Therefore, the applied stress reveals the differences between the cultivars and the evaluation of the cultivars in the stress environment can lead to the selection of suitable cultivars. The effect of genotype was not significant for some traits, which suggests that the selection of cultivars for these traits is complex because the differences between the cultivars are not clearly defined. Selection is more reliable for traits that are significant, such as grain yield. The interaction between genotype and environment also increases the complexity of selection, which was significant only for plant height in this study. The high coefficient of phenotypic variation for traits indicates that the expression of these traits is greatly affected by the environment. Also, the high coefficient of genotypic and phenotypic variation for some traits indicates the wide range of changes for these traits. In general, the correspondence of the phenotypic coefficient and the genotypic coefficient in some traits indicates that the environmental effects on trait expression are negligible. However, when the phenotypic variation coefficient is higher than the genotypic variation coefficient, it indicates a high environmental effect. The low difference between the phenotypic and genotypic coefficient of variation for some traits suggests that these traits are more controlled by genetic factors. Therefore, the selection of parents for these traits is appropriate for hybridization programs.

Read More Abstract Background and Objectives GF677 is a widely used rootstock for peach, nectarine and almond. It is vigourous and adapts well to limestone soils and drought. Due to low efficiency of propagation trough cutting, tissue culture is a good and fast method for propagation of disease-free plants of GF677. In vitro multiplication efficiency in GF677 is widely dependent on the type of culture medium and growth regulators. Therefore, the aim of this study was to determine the conditions required to optimize micropropagation methods for GF677 rootstock from nodal explants. Materials and Methods This experiment was conducted during 2015 at the laboratory of plant tissue culture at Agricultural and Natural Resources Research Center of Kermanshah, using a factorial in a completely randomized design with three replications. Sterile nodal explants were cultured onto different media of MS, WPM and B5 supplemented with benzyl adenine (BA) at concentrations of 0.25, 0.5, 1, 2 and 4 mg/L and indole-3-butyric acid (IBA) at concentrations of 0, 0.1, 0.25 and 0.5 mg/L. Elongated shoots of GF677 were cultured on MS medium supplemented with 0.25, 0.5, 1.0 and 2.0 mg/l IBA and 0.0, 0.1, 0.2 and 0.5 mg/l BA for rooting. Factorial analysis of variance was carried out and differences between means were scored with LSD tests. Results The effect of different culture media (MS, B5 and WPM) and plant growth regulators (BA and IBA) on a number of shoots per proliferated explant of GF677 proved that the highest rate of adventitious shoot initiation, percentage of egeneration, shoot length and diameter, number and length of nodal, leaf number and multiplication was obtained in MS medium containing 1 mg/l BA + 0.5 mg/l IBA. Cytokinin stimulates the initiation and activity of axillary meristems, which result in shoot formation. This study showed that the number of shoots increased as concentration of BA increased to a certain amount. As concentration of BA increased to 1 mg/l, the number of shoots increased, too. It seems that there is a positive correlation between concentration of BA and the number of shoots to a certain concentration of BA. At concentrations higher than 1 mg/l BA the number of shoots increased. One of the possible reasons can be the reductive effect of higher concentrations of BA. Apparently, a certain amount of BA is required to obtain the best effect. Higher concentrations of BA cause formation of high amount of callus, which is not appropriate in tissue culture. The results of this experiment confirmed the positive influence of the growth regulator on the number of roots per shootlet. Among the various plant growth regulators used, the highest rate of rooting (33%) and the number of roots per shootlet (1.62) were obtained on MS medium containing 1.0 mg/l IBA and 0.5 mg/l BA. The concentration of auxin during rooting period strongly influenced the quality of root system during the rooting period. Rooted plants were transferred to a combination of terrestrial environments, including perlite, sand and soil in the ratio of 1: 2: 1, respectively. Among rooted plantlets that were acclimatized and transferred to the potting mix successfully, 90% survived and grew naturally after strengthening and transferring to the soil. Discussion The shoot multiplications are influenced by the media and growth regulators. The MS medium gave the best results for the proliferation of cultures from explant among the tested media (MS, B5 and WPM). Growth regulators compounds have significant effects on different traits and these changes depend on type and concentration of hormone. Both cytokinin and auxins are important in micropropagation for GF677 rootstock.

Read More Abstract Background and ObjectivesPomegranate (Punica granatum L.) is a fruit tree that originated from Iran and has been cultivated since ancient times. Classical propagation methods of pomegranate like stem cutting and air-layering have seasonal timing and a higher risk of pathogens contamination. Micropropagation by tissue culture could be considered a method for production of steady and pathogen free plants in this species. However, factors such as lack of rooting and browning due to the oxidation of phenolic components under invitro culture can limit its micropropagation efficiency. To overcome these problems, the effects of various factors, including media, iron-chelating agent, and plant growth regulators, were examined on the pomegranate propagation through in vitro conditions. Materials and MethodsTo obtain sterile explants, stem cuttings of the pomegranate cultivar Shishe Cap Ferdos were treated with different disinfection treatments. Then, the effect of DKW, MS and WPM basal media and two types of iron chelating agents (EDDHA and EDTA) were evaluated to decrease the secretion of phenolic compound from explant and subsequent browning. In the next step, the effect of plant growth regulators treatments, including 1-2 mg/l of BA in combination with 0.2 mg/l of various auxins (IAA, NAA or IBA) on shoot development and proliferation was studied. After that, rooting of regenerated plantlets was explored at WPM medium supplemented with 0.5 or 1 mg/l IAA, IBA or NAA. Finally, pomegranate plantlets were transferred to different soil mixtures, including eat moss and perlite (2:1), cocopeat and perlite (2:1), peat moss and vermiculite (2:1) and cocopeat and vermiculite (2: 1) for acclimation. ResultsThe results indicated that sterilization with 2% sodium hypochlorite for 15 minutes followed by washing with 1g/l carbendazim for 10 minutes, and finally culturing the explants in the medium containing 1 g/l carbendazim had the best effect on reducing explant contamination. Among different basal media and iron chelating agents, WPM resulted in the lowest explant browning. Also, EDDHA was more efficient compared to the EDTA so that explant browning in WPM media containing Fe-EDDHA reduced by 55.8% compared to DKW containing Fe-EDTA. In addition, WPM basal medium supplemented with 2 mg/l BA and 0.2 mg/l IAA induced higher shoot proliferation in node explant of pomegranate. For rooting treatments, after 45 days, WPM supplemented with 1 mg/l IAA induced higher rooting percentage, root length and number in pomegranate shoots. Among different acclimatizing treatments, plantlets that grew on peat moss and perlite (2: 1) had the highest leaf number and stem height. DiscussionOur results suggested that the medium composition and chelating agent affected pomegranate explants browning. EDDHA appears to have a positive effect on reducing browning by chelating copper, which is a phenol oxidase cofactor. Also, EDDHA was more efficient compared to the EDTA.

Read More Abstract Background and Objectives Currently, there is an immense attention towared medicinal plant and secondary motabiltes. Yarrow (Achillea millefolium L.) is a medicinal plant which consists of two important compounds; including penene and linalool. Since these two compounds have the importance values, the researchers significantly focused on them. The rate of transcript expression of penene synthesis and Linalol synthesis induced at the end of 2-c-methyl-D-erythritol-4-phosphate (MEP) pathway that caused to produce Penene and Linalol. In order to evaluate the synthesis of Penene and Linalol, a research study was carried to exame the exogenous treatment (zero, 25 and 50 mg/l) of gibberellic acid (GA3) at different times (24, 48 and 74 hr.) on Yarrow at the greenhouse on 1395-1397. The results of R-PCR indicated that significant rate of the transcript expression was related to the Penene synthesis at 25 mg concentration of GA3 at the 48 hr after treatment. In the most samples as the rate of the Penene synthesis increased the rate of Linalol synthesis decreased. The evidences suggested that the exogenous treatment of GA3 induced a transduction signal in the plant. In which at the end, the plant molecular process respond to it and showed in overall that evaluation of GA3 implemented different encoding gene expression for theses compounds, and illustrated that when the Penene synthesis inceased in the treated plant the Linalol synthesis dcreased simontenously. Materials and Methods A greenhouse pot trial was conducted at the Experimental Research Station of College of Agriculture at Shahid Chamran University of Ahvaz. The plants used in this study were provided from the National Center for Genetic and Biological Resources of Iran, with access code P1000093. Two months after seed germination, the desired amounts of gibberellic acid were prepared with distilled water (25 mg/l and 50 mg/l), and then each potted plant was sprayed uniformly with 10cc of prepared solution. Sampling was carried out by collecting the leaves of treated plants at 24, 48 and 72 hours after the treatment. The phenol-chloroform method (Wang and Ghabrial, 2002) was conducted with some modifications to extract RNA of collected leaves at the two biological replications. Quantity and quality of extracted RNAs were measured using Nanodrop and electrophoresis of %1 agarose gel. Synthesis of cDNA was performed using the Takara cDNA synthetase kit according to its manufacturer's instructions. Partial sequences of pinene synthase and linalool synthase genes were obtained in an early experiment in our laboratory (data is not published). The coding sequence of the target genes in other plants within the same family aligned with our obtained sequences to identify conserved segments for each gene using CLUSTAL W program. The standard housekeeping actin for yarrow with accession number JX679606.1, which is available in the public GenBank database, used to normalize the expression of the genes of interest. The primers of all genes (pinene synthase, linalool synthase and actin) were designed from the conserved sequences by PrimerQuest Tool, and then verified with oligoanalyzer v.3.1. Relative expressions of target genes were determined using real-time PCR with SYBR green fluorescence detection. Relative differences in target gene expression were calculated using REST software. Results The acquired result of real-time PCR revealed that in pinene synthase gene, the highest amount of transcription occurred at 48 hours after the treatment with the concentration of 25 mg of gibberellic acid and, interestingly, the lowest expression of the transcript for linalool synthase gene observed at the same level of concentration and time. In most of the samples, with the increase in pinene synthase transcriptase, the amount of linalool synthase transcript decreased. Discussion Taking into account the fact that both pinene and linalool are synthesized from the common substrate geranyl diphosphate. Therefore, an increase in the expression one leads to reduction in the transcript expression of the other one in the same pathway. Our results also confirmed that the transcription level of pinene synthase gene has been increasingly altered in response to the applied concentrations of exogenous gibberellic acid, resulting in a reduction in the expression of linalool synthase gene. Evidence suggests that the application of gibberellic acid triggers a cascade of molecular events which ultimately constitutes the plant response to the elicitor.

Benzylisoquinoline alkaloieds (BIAs) are a very large and complex group of plant alkaloids. Today’s various methods of genetic engineering are applied to improve the biosynthetic pathways in medicinal plants. For successful implementation of techniques such as gene silencing and over-expression, it is important to get accurate and complete information about genetic characteristics of related genes in the biosynthesis of secondary metabolites. DBOX is one of the important genes in biosynthesis of sanguinarin and papaverin alkaloids of poppy. According to the cause, the present study was performed on DBOX to explain genetic characteristic and gene expression pattern. Materials and Methods Specific primers were designed based on databases, and complete sequences of DBOX gene were amplified using DNA and cDNA as a template, and then were cloned in pTZ57R/T plasmid. After sequencing of cloned fragments, CDD, CELLO and ProtParam tools were used to determine ORFs, domains and the location of protein accumulation. Phylogenetic relationship between the gene in this plant with other plants was determined. Finally, the pattern of gene expression in different tissues was assessed using real time RT-PCR technique and RNA-seq data derived from SRA data bank. Results Sequencing results showed the 1614-bp fragment that has 100% identity with the DBOX gene of P. somniferum. The protein encoded by this gene was analyzed by bioinformatics methods. Three domains including FAD binding, Berberine and FAD/FMN-containing dehydrogenase in the protein sequence were identified and the existence of an N-terminal signal peptide was also determined in this protein. The results also showed that the highest concentration of protein was present in the plasma membrane and highest function of this protein was in the extracellular. In order to analyze and determine the expression pattern of DBOX gene in different tissues, 5-gene libraries of RNA-seq data of opium poppy (with ERX651037, ERX651023, ERX651056, ERX651082 and ERX651062 access numbers) derived from NCBI-SRA were analyzed. The numbers of sequences from the alignment between different tissues were compared using the Fisher exact test and Chi-squared 2X2 tests. Results of the gene expression analysis showed that the highest level of gene expression of this gene performed in roots and the lowest level of gene expression occurred in the stems. In addition, results of the real-time RT-PCR experiment were similar to in silico tests and the present results demonstrated that DBOX gene transcript levels in root were significantly more than other plant tissues. Discussion Results showed that DBOX gene is an intron less gene. According to existing domains, this gene is a member of a FADOX gene family. Due to the presence of FAD and berberin like domains in protein sequence of this gene, its key function in biosynthesis of BIA alkaloids was confirmed. Different expression patterns of DBOX gene in root confirmed accumulation of high value sanguinarin alkaloid in this tissue.

Read More Abstract Background and Objectives Satureja avromanica Maroofi is a medicinally perennial herb which is native to Iran, exclusively restricted to Uramanat mountains of Kurdistan province. It has various pharmaceutical and biological activities such as antibacterial, antifungal, antioxidant, anti-inflammatory, antiviral, anticancer and antiprotozoal properties. The experiment was conducted to study the biochemical characteristics of S. avromanica Maroofi extract by two extraction methods as solvent and solvent-ultrasonic. Materials and Methods The study was carried out as a factorial based on a completely randomized design. The investigated factors were the type of solvent (water, 75% water, 50% water, 75% methanol and methanol) and extraction method (solvent and solvent-ultrasonic). For this purpose, the plant samples were collected from Uraman Mountains of Kurdistan province in July of 2016. Samples were immediately transferred to Herbarium of Agricultural Research Center and Natural Resources of Kurdistan province. The samples were dried in shade for 15 days, and then crushed into pieces of 0.5 to 1 cm by a laboratory mill (Molyneux, Spain). The content of phenol and flavonoid as well as antioxidant activity was measured. Besides, the components of phenol and flavonoid for the best treatment were determined by HPLC apparatus. Results The results showed that the compounds extracted by water-ultrasonic were significantly more than those extracted by water. Inhibition percent in water-ultrasonic and water treatments were 16.62% and 11.92%, respectively. The content of phenol (45.36 mg/g, 40.29 mg/g) in water-ultrasonic was higher than that obtained in water treatment (40.29 mg/g). The flavonoid content (15.33 mg/g) in water-ultrasonic was more than that in water treatment (13.29 mg/g). Therefore, water-ultrasonic treatment was more efficient in extracting S. avromanica Maroofi compared to water treatment. On the other hand, the 75% water treatment had more compounds compared to other solvents. The chromatogram of essential ingredient revealed that S. avromanica Maroofi had more Caffeic acid, Gallic acid, Chlorogenic acid, Quercetin, p-Coumaric acid, Ferulic acid. Also, Rutin as the highest amount was obtained in Ferulic acid to be 9.12 mg/g. Discussion We found that water is the best solvent for biochemical extraction of S. avromanica Maroofi. We found a 12% increase of phenol and 14 % increase of flavonoid in water-ultrasonic relative to water treatment. Due to the wide structure of hydrogen bonds, water is a very polar solvent with a dielectric constant at room temperature and atmospheric pressure. Ultrasonic is a type of ultrasound that increases the polarity of the liquid phase. Ultrasonic waves cause mechanical fluctuations in the liquid medium. The mechanical effect of ultrasound improves the penetration of solvent into cellular materials and mass transfer. Hence, ultrasonic waves are more efficient for producing the biochemical compounds like phenol and flavonoid.

The romans enlarged its cultivation from the Greek islands throughout the Mediterranean basin, mainly along the African and European coasts as well as in the areas where large volumes of water have improved the climate. In the past, cultivar distinguishing approaches were based on morphological characteristics of leaf, fruit and stone. Recently, morphological traits with isozyme and DNA- based markers have been applied to the classification of cultivars. The aim of this study was the evaluation of morphological and molecular similarities between the new and the known cultivars of olive trees in the Fadak garden in Qom province in Iran. Materials and Methods In order to analyze genetic variations of olive tree genotypes in Qom province, the olive genotypes (Manzanilla, Sevilana, Mahzam, Arbequina, and Conservalia) and two superior cold resistant genotypes (Fadak and Tooba) were studied using morphological traits based on distinctness, uniformity and stability (DUS) guideline in the Fadak garden (Qom province, Iran) in 2013. In addition, a total genomic DNA was extracted from the leaves, and the PCR was carried out by a set of six inter simple sequence repeats (ISSR) markers for amplification. Then, amplification products from ISSR primer tests were characterized on 2% agarose gels immersed in 1X TBE buffer. The gels were stained with ethidium bromide. The PCR fragments were scored for the presence or absence as 1 and 0, respectively. Results In this study, analysis of variance was performed on 12 quantitative traits. Results showed there were significant differences (p < 0.01, p < 0.001) between all the traits and Fadak and Tooba separated from other genotypes by comparing the means of the tests (Duncan multiple range method). According to the biplot graph (the first two components) in the PCA method by quantitative traits, Arbequina, Manzanilla and Tooba were included in one group, and Fadak and Tooba were distinctly separated from other genotypes. Also, according to the biplot graph (the first two components) in the PCA method by qualitative traits, the distance between Fadak and Tooba was low. In the molecular analysis, all ISSR markers (six primers) showed polymorphism. The total band range was 8-21 (with 14.66 as the average) and the PIC varied from 0.231 to 0.447 (0.309 average). Based on the PCA method, Tooba was utterly separated from other genotypes. Discussion In this study, Fadak and Tooba were completely different from other varieties using morphological traits and ISSR molecular markers. Fadak was found to be similar to Sevilana and Conservalia. There was a high similarity between Fadak and Tooba. Also, no similarity was observed between the results of morphological and molecular traits.

Read More Abstract Background and Objectives Plant cells often increase cold tolerance by reprogramming their genes expression, which results in adjusted metabolic alternations, a process enhanced under cold acclimation (CA). In this study, responses of Carbohydrate alteration and delta12 and delta15 genes expression to cold stress (CS) phases were comparatively studied in three genotypes of bread and durum wheat differing in sensitivity. Two of them (Norstar, bread wheat and Gerdish, durum wheat) were tolerant to CS and the other one, SRN (durum wheat) was sensitive to CS. Materials and Methods Seeds of Norstar (hexaploid, bread wheat) and the two genotypes of Gerdish and SRN (tetraploid, durum wheat) provided by Dryland Agriculture Research Institute (DARI) of Iran were soaked in distilled water and then germinated in Petri dishes on filter paper for 72 h at 25 C in a thermostat. Subsequently, the seedlings were planted in pots. The cooling regime adopted in our experiments allowed us to differentiate the examined genotypes in terms of their tolerance to CS. In our experiment, the plants were moved from control conditions immediately into the acclimated temperature of 4–5 ºC for 14 days with the same photoperiod and irradiance. Leaf samples of genotypes were harvested and analyzed after 14 days under these conditions. After 14 days of CA, the plants were placed into a climatic chamber chilled preliminary to 0 ºC. During further treatments, the temperature was lowered gradually to -5 C (at the rate of 0.5 ºC min-1 ), and the plants were incubated at this temperature for 12 and 24 h. Total cellular RNA was extracted by Biozol method (Fersion Pooyesh, Tehran, Iran) using 80 mg FM leaflets. Applying fermentase reverse transcriptase enzyme instruction, the first strand of cDNA was produced after DNase treatment. Primers were designed using primer 3 to obtain 18–21 bp length. Carbohydrate extraction and determining the carbohydrate concentration were done by 80% ethanol and the AOAC method, respectively. Results These responses confirmed the existence of a wide range of genetic capacity in durum wheat to increase cold tolerance particularly in Gerdish. The findings of the present study showed that under experimental treatments, the carbohydrate content significantly changed so that cold stress in acclimated plants increased sucrose, glucose and fructose contents particularly in Norstar and Gerdish as compared to the SRN plants. Increasing expression of delta12 and delta15 genes under cold stress in Norstar and SRN genotypes in comparison with SRN indicates the capacity of cells in increasing cold stress. Discussion The results may be a sign for associating other metabolite or enzyme activities to create relative tolerance against cold-induced oxidative stress. Also, these responses showed high genetic diversity for cold tolerance in durum. Eventually, assessing the dynamics of cell responses after CS without CA phases could profitably be a novel path in plant stress response investigations in the short run.

Read More Abstract Background and Objectives Drought is a major abiotic stress that limits agricultural crop production. Thyme (Thymus vulgaris L.) is an aromatic and medicinal plant which is very important for the herbal industry. This study was aimed to investigate the variation some of the secondary metabolites and morphological traits as well as changes in expression of genes involved in the biosynthesis of thymol (including HMGR and TPS2) by real-time PCR. One of the main goals of the present study was to find out the relationship between the transcript of these genes and related metabolites in thyme. Materials and Methods This study was conducted in the greenhouse of Iranian Institute of Medicinal Plants of Jahad Daneshgahi (Karaj, Iran) in 2016. The experimental design was a randomized complete block design with 5 replication and 4 water deficit stress treatments, including 100% (T1, control), 70% (T2, mild stress), 40% (T3, moderate stress), and 20% (T4, severe stress) of field capacity (FC). Total RNA was extracted from leaves using TRIzol reagent following the manufacturer’s instructions. First strand synthesis of cDNA was performed immediately using 1 µg of total RNA with simultaneous use of oligo-dT (50 µM) primers and random hexamer and reverse transcriptase enzyme following the manufacturer protocol. In order to examine gene expression by real-time PCR, the method of fluorescence dye SYBR green (Fermentase, USA) was used. The amount of thymol and carvacrol were measured by HPLC. Results Results of morphological traits showed that increasing the water deficit stress caused a decrease in root biomass, shoot fresh weight, plant height, root length, and the number of side branches. Results of gene expression profiling showed that the highest expression of TPS2 and HMGR genes and the highest amount of thymol and carvacrol was obtained in treatment T2 (70% FC). Discussion TPS2 and HMGR gene expression changes were similar to the changes of monoterpenes thymol and carvacrol in all treatments and the highest number of monoterpenes and gene expression were obtained in mild water deficit stress treatment in comparison with control, moderate, and severe stresses. Altogether, according to the results of this study, mild stress had a significant effect on increasing the expression of HMGR and TPS2 and led to a higher production of monoterpenes.

The human population is expected to increase to over 8 billion by the year 2020. Therefore, providing adequate food and preventing crop yield reduction is essential. Plant breeding can solve this problem to some extent. Evaluating yield components and their inter-relationships as well as detecting suitable selection indices are very important in the safflower breeding program. The multivariate statistical analysis can provide more insights into the deep structure of data and traits’ relationships Materials and MethodsIn order to study the relationships between yield components of eight genotypes of safflowerunder normal irrigationand drought stress conditions and determine the high yield genotypes, a split-plot experiment was conducted in a randomized complete block design with three replications in the research field of Imam Khomeini International University, Qazvin. Irrigation regime included normal irrigation(irrigation after 60 mm evaporation from class A pan evaporation) and drought stress (watering until 50% flowering similar to normal irrigationand 50% flowering to maturity and irrigation after 100 mm evaporation from class A pan evaporation) were considered as main plots. Eight genotypes, including Kuseh Local, Sina, Isfahan Local, Mexican 88, Faraman, Soffeh, Goldasht, and Mexican 11 were used too. ResultsThe analysis of the variance showed that the irrigation effect was significant for traits 1000 grain weight, flower weight, harvest index, and grain yield. There were significant differences among the genotypes for most of the measured traits, except boll diameter, boll number per plant, relative water content, and relative water loss. Plant height, shoot number, boll number, stem diameter, chlorophyll value, flower weight, biological yield, and harvest index positively correlated with the grain yield in both normal irrigation and drought stress conditions. Based on the results of stepwise regression and path analysis, biological yield and harvest index in both normal irrigation and drought stress conditions had the greatest effect on grain yield. Due to the high correlation between yield and harvest index and biological yield under stress conditions, the selection of varieties with high harvest index and biological yield can be very effective in achieving maximum yield under stress conditions. Factor analysis revealed that 4 factors accounted for approximately 82% variance changes in both normal irrigation and drought stress conditions. These factors were called yield, seed, boll size, and leaf water, respectively under normal irrigation, andplant vigor, yield, seed, boll size, and leaf water, respectively under drought stress conditions. Based on the seed yield factor and the biplot display, Faraman, Mahali Isfahan, Sina, and Mahali Kuseh were determined as high yield genotypes both in normal and drought stress conditions. DiscussionGrain yield is a quantitative trait with a low broad sense heritability. Therefore, identifying the traits correlated with grain yield is a suitable strategy for the improvement and the indirect selection of grain yield. Stepwise regression and path analysis are efficient and useful statistical approaches for reaching the above aim. According to the results, the traits of the harvest index and biomass were recognized as the best suitable traits for indirect grain yield selection under normal and drought stress conditions.

Sequential Path Analysis Based on Yield and Morpho-Physiological Characteristics in Squash Landraces from Northwest of Iran

Read More Abstract Background and Objectives Squash is an annual herbaceous plant and grows in tropical and subtropical regions of Europe and America. Edible species are divided into three groups of zucchini or courgette, gourd and pumpkin squash. The seed of pumpkin squash has high nutritional value due to its high protein and oil content. Iran is ranked fourth in terms of production and cultivation area of squash in the world after China, India and Russia. The present study was conducted in order to determine the most important morphological and physiological characteristics influencing yield in squash landraces from Northwest of Iran. Materials and Methods Eighteen squash landraces comprising 11 confectionery landraces, 5 gourd landraces and 2 ornamental landraces were evaluated in a randomized complete block design with three replications in Saatlou research station of West Azerbaijan. The landraces were evaluated for morphological and physiological characteristics based on International Board for Plant Genetic Resources (IBPGR)) descriptor. Results The results of phenotypic correlations revealed that the characteristics including number of fruit, fruit weight, 100 seed weight, fruit diameter, seed thickness, plant height, internodes length, primary branch and fruit pH and characteristics including seed weight, peduncle lengths, sepal’s length of female flower and fruit total soluble solids have positive and significant correlations with fruit yield at 1 and 5 percent probability level, respectively. In stepwise regression, four features including internodes length, sepal’s length of female flower, chlorophyll percentage and fruit pH were entered in model. The results showed that fruit pH and internodes length have the most direct and positive effect on the fruit yield, and the percentage of chlorophyll has the highest indirect effect through internodes length on the fruit yield. Discussion Sequential path analysis showed that selection of genotypes based on only correlation coefficient may not be effective. Several characteristics were identified to explain squash yield during path analysis. Selection based on identified characters would be more effective in improving yield in squash breeding programs.

Since the retrotransposesons have transferable motive they tend to move along the genome. The movement is a key figure to induce alteration at the genomic structure. Thus, this reinforces the genome to boost the expression of some genes and/or silence the other genes. Furthermore, retrotranspostions are able to affect the activity of genes transcription at the organelles and biotic and abiotic stresses. Materials and Methods In the present study, retrotranspostion iPBS marker was used to assess genetic characterization of 130 inbred lines of rice which were derived from hybridization of Neda and Hashmi rice cultivars. In this study, 4 primers were used which were able to produce 2990 bands and among them1449 were polymorphic. Results According to amplification of polymorphic primers analysis, R2020 primer showed totally 443 polymorphic bands and had 56.79% bands with the highest polymorphism among all the primers. None of primers showed any single polymorphic band. Discussion Cluster analysis classified inbred lines of rice into two groups. Parental lines along with 146, 148, 149, 151 and 154 lines were in one group and 67, 68 and 82 lines were fit into another group with close distance to the parental lines. In terms of the presence of retrotransposition genes these lines possess the most genetic similarity to the parental lines. The information which was revealed by the findings of the presented study indicated that iPBA markers can be assigned to trace the genetic mapping in order to assess the molecular genetics of the characteristics of the line within a plant population in the breeding program. Read More Background and Objectives Dracocephalum kotschyi Boiss. (Labiateae) is an endemic perennial herbaceous plant known in Iran as Zarrin-Giah. Recent pharmacological studies have confirmed some of the methoxylated flavonoids in plant’s parts having anti-cancer properties. Excessive harvesting of wild plants and limited distribution areas are the main reasons why D. kotschyi is now listed as an endangered plant. Polyploidy induction is an effective tool in medicinal plant breeding. Chromosome duplication and polyploidization may affect plant morphology and breeding systems, ultimately enabling the release of improved genotypes. Materials and Methods In this study, an efﬁcient procedure was established for successful induction of tetraploid D. kotschyi by treating diploid explants with colchicine in horticulture laboratory of Urmia University during 2014-2015 years. Seedlings apical meristem treatment was carried out in two growth phases including, the two and four-leaf plants through presoaking manner. Colchicine at a concentration of 0, 0.05, 0.1, 0.2 and 0.5 % (w/v) was applied in each of these stages. Cytological and morphological evidence conﬁrmed the results of ﬂow cytometry analysis. Results Cytological analyses showed the increase of chromosome numbers from 2n=2x=20 to 2n=4x=40. The results of this study demonstrated for the ﬁrst time that chromosome counting in D. kotschyi of a total of 165 surviving seedlings, 7.27% was found to be tetraploids, 13.3% was chimers while the remainder were diploids. Tetraploid plants demonstrated signiﬁcantly longer stomata and a higher stomatal index compared with diploid control plants. Negative correlation between stomata size, plants height, leaf number and lateral shoot number was obtained in treated plants. The total content of flavonoids increased from 1583.28 in diploids to 1890.07 (μg/g DW) in stable tetraploids. Discussion It seems that 0.5% of colchicine can be used as an effective treatment for polyploidy induction in D. kotschyi.

Read More AbstractBackground and ObjectivesIn order to increase yield in apple orchards, multiplication of rootstocks is essential, and it depends on the availability of a simple and easy proliferation method of rootstocks. The study investigated the effect of disinfection, culture condition and plant growth regulators on the rate of contamination, number of lateral branches, stem length, number of leaves, callus creation and the rate of branch creation of rootstocks MM106, MM111 and B9.Materials and MethodsA factorial experiment was carried out as a completely randomized design with three replications. Surface sterilization of explants with different concentrations of mercuric chloride and sodium hypochlorite (mercuric chloride 0.1% for 3 minutes and ethanol 70% for 30 seconds; mercuric chloride 0.1% for 5 minutes and ethanol 70% for 30 seconds; NaOCl 0.75% for 15 minutes and ethanol 70% for 30 seconds) was done and then explants were cultivated in modified MS, WPM and DKW mediums. The evaluated characters were infection percent of explant, number of lateral branches, stem length, percentage of callus induction and number of leaves. Data were analyzed using SAS software. The analysis of variance on the test data was performed at 5% level and comparison to the middle of the Duncan test.ResultsResults showed that using a mercuric chloride (0.1%) and ethanol (70%) respectively for 3 Minutes and 30 Seconds achieved the least contamination. Results showed significant differences between plant hormone and rootstocks for traits. In this test, the modified MS medium with 0.1 mg/L BA hormones with the largest percentage growth had more successful establishment. Average comparisons showed that rootstock MM106 in terms of all traits had a significant difference with other rootstocks. The highest stem length, number of leaves and callus creation were shown in rootstock MM106. In order to investigate the effect of hormone levels on branch creation, different levels of BAP (6-Benzylaminopurine) and IBA (Indole-3-butyric acid) were used. The best medium was the culture containing 0.1 milligrams per liter IBA plus 2 milligrams per liter BAP.DiscussionsIn general, the present results showed that genotypes respond differently to in vitro conditions. The rootstock B9 by the above method showed better response to branch creation trait than other genotypes.

Read More Background and Objectives Mulberry (Morus spp.) is a tree of the family of Moraceae. There are 24 species of Morus and one subspecies, with 100 known varieties. The various methods used in classification of Morus were mainly based on the conventional systematic studies and agronomic characters. Despite the widespread genetic diversity in Iranian mulberry tree, so far, the identification of these trees has not been selection, introduction and cultivation. In order to identify mulberry trees during 1387-1392, they were collected from different provinces (Tehran, Mazandaran, Kurdistan, Markazi, Khuzestan, Fars, West Azarbaijan, Isfahan, Hamedan, Khorasan, South Khorasan, Semnan, Ardabil, Kohgiluyeh and Boyer-Ahmad, East Azerbaijan) according to the IPGRI (International Institute for genetic reserves). The aim of this study was evaluation of genetic diversity and mulberry clustering genotypes, on the basis and quantitative characteristics and to determine the genetic distance between them. Material and Methods Mulberry trees are widely distributed in Iran. After reviewing the population of mulberry according to descriptor of IGPRI throughout the country, 118 specimens of mulberry from genotypes were selected and they were evaluated for their morphological characteristics. Some of these characters are quantitative and some others were measured and recorded based on coding UPOV. After collecting the data, descriptive statistics were extracted and analyzed. With the assumption of variance analysis distinguished genotypes into treatment and regions within each area as replication, design used nested. Mean comparison of quantitative traits LSD. Discussions Analysis of simple correlation coefficients showed a significant positive correlation during the harvest time of fruit ripening r=0.82, the shape of inflorescence at the time of fruit ripening r=0.71, shape of inflorescence at the time of fruit ripening r=0.71 at 1% level. Brix with the shape by time of fruit ripening r=0.60 Brix of fruit shape r=0.28 fresh weight at the time of ripening r=0.81, significant positive correlations were calculated at 5% level. The results of mulberries ripen over an extended period of time unlike many other fruits. Using cluster analysis to the “Ward” based on morphological characterization at a distance of 15; genotypes were divided in three main groups. In order to determine the most important morphological characters to differentiate among the population of principal component analysis was used (PCA).The first components to 0.65 percent of total respectively. Overall, the results showed that great morphological variation among the plants population.
