Read More Introduction Japanese barberry (Berberis thunbergii) is a perennial and woody shrub that grows in different regions of Iran and the world. The Berberidaceae family has a high potential in food and pharmaceutical industries due to its antioxidant, antimicrobial and anticancer activities. Elicitors are biotic or abiotic stimulators that can induce responses in plants. Abiotic elicitors are used to increase secondary metabolites in plants. Nanoparticles have minimum dimensions between one and 100 nanometers, which have specific physical and chemical properties. Like many nanoparticles, zinc oxide and cerium dioxide nanoparticles are toxic to living organisms. The mechanism of toxicity of nanoparticles is generally contributed to the induction of oxidative stress which lead to the formation of free radicals. Nanoparticles of zinc oxide and cerium dioxide can increase the antioxidant capacity of plants by stimulating oxidative stress. Material and methods In order to study the changes in the physiological and biochemical characteristics of Japanese barberry under the influence of abiotic elicitors, an experiment was conducted in a completely randomized design with three replications in the greenhouse and laboratory of the Faculty of Agriculture and Natural Resources of Imam Khomeini International University in 2019. Treatments were included zinc oxide elicitors (0.1 g/L) and cerium dioxide nanoparticles (0.0002 g/L) and control (no elicitor). In this research, the amounts of protein, antioxidant enzymes and photosynthetic pigments in tissues of Japanese barberry were measured. Data analysis was done with SAS 9.1.3 statistical software. Comparison of means was done with Duncan's multiple range test and graphs were drawn using Excel software. Results and Discussion The results of the data analysis of variance showed that the effect of treatment with cerium dioxide and zinc oxide nanoparticle elicitors on the amount of leaf protein, antioxidant activity and also the amount of photosynthetic pigments was significant. Comparison of the means showed that these two elicitors increased the leaf protein content by 26.2% and 12.4%, respectively, compared to the control, but they did not have a significant effect on the root protein content. Cerium dioxide decreased the activity of guaiacol peroxidase enzyme by 77.1% in the root; But in leaves, the activity of this enzyme increased by 134.8% due to treatment with zinc oxide. The highest activity of leaf superoxide dismutase enzyme (25.2% increase compared to control) was related to zinc oxide treatment. The lowest content of catalase enzyme in leaves and roots was obtained with cerium dioxide and zinc oxide. Zinc oxide increased the activity of leaf ascorbate peroxidase enzyme by 111.8%; In the root, the highest and lowest activity levels of this enzyme were obtained in the control and cerium dioxide treatments, respectively. Both elicitors significantly increased the leaf peroxidase enzyme activity compared to the control (128.6% and 157.1%, respectively); However, only the treatment with zinc oxide caused a significant increase of 25.5% in the activity of this enzyme in the root. The highest activity of glutathione reductase enzyme in leaf and root was observed in zinc oxide treatment (23.3% and 12.5% increase compared to control). Nano zinc oxide increased the amount of total chlorophyll and carotenoid in the leaves compared to the control by 93.4 and 67.5%, respectively. Conclusion It was concluded that zinc oxide nanoparticles elicitor had the greatest effect in increasing the amount of photosynthetic pigments and enzyme activity. By using this elicitor in Japanese barberry culture, the amount of antioxidant enzymes can be increased and the extract can be used as a strong antioxidant in pharmaceutical products. .

Read More Introduction Secondary metabolites have a complex structure which is produced by chemical bio-synthesis. Therefor, they are mostly extracted from wild or cultivated plants. The essences are natural, complex and volatile secondary metabolites which are mostly produced in medicinal and aromatic plants. Peppermint (Mentha piperita L.), a hybrid from M. aquatica and M. spicata, is an herbaceous and perennial plant belonging to Lamiaceae family. It is widely cultivated in temperate regions of Europe, Asia, America, India and Mediterranean countries due to its commercial value and unique odor. Materials and Methods In order to investigate the influence of putrescine treatment at three levels including 0, 0.1 and 1 mM, and salicylic acid treatment at three levels including 0, 0.1 and 1 mM, on the yield performance and essence components of peppermint, a field factorial experiment was performed based on complete blocks design with three replications at Research Station of Tabriz University during 2019. Peppermint seedling were prepared from Agricultural Jihad Organization of Bostan Abad and cultivated at a distance of 30×30 cm. Foliar application of salicylic acid and putrescine was carried out twice during vegetative stage of plants (with one week interval). Harvesting and bio-chemical assessments were done at stage of 25 % flowering. The essential oil of peppermint was assessed to identify its main components. Results and Discussion The results of the current study showed that the average yield of essential oil in plants treated with 1 mM salicylic acid was significantly increased compared to the control. A total of 24 compounds were identified in the essential oil of peppermint aerial parts. The main components of the essential oil were included menthone, menthol, L-menthol and 1,8-cineole. Menthone and menthol constituted 26.14 and 23.21 % of essential oil, respectively. Different concentrations of salicylic acid and putrescine increased the production rate of menthone, menthol, l-menthol and methyl acetate compounds in the essential oil compared to the control. The concentration of 1 mM salicylic acid had the greatest effect on the production of essential oil compounds. The correlation results between essential oil compounds showed that menthone has a positive and significant correlation with methyl acetate. Similarly, foliar application of salicylic acid (10 mM) and indole-3-acetic acid (1.5 mg L-1) increased the content of main component of peppermint and lemon balm. In the other study, foliar application of lemon balm plants with salicylic acid and putrescine caused a considerable increase in monoterpenes and sesquiterpenes content which was in line with obtained results of the present study. Conclusion The plant growth regulators used in this research increased the yield and composition of the essential oil of aerial parts of peppermint, which shows that both salicylic acid and putrescine have a high potential in increasing the potency and chemical composition of the essential oil in the peppermint plant and probably in other aromatic crops. Therefore, foliar application of salicylic acid and putrescine during vegetative growth of peppermint could be introduced as influential technique to improve qualitative and quantitative characteristics of this aromatic and medicinal plant.

Read More Introduction Salinity stress is one of the most important problems in the cultivation and production of agricultural products, including medicinal plants. The expansion of saline lands following continuous droughts in recent years increases the importance of the tolerance level of medicinal plants determination, their type of reaction, and the necessity of finding a solution to reduce the damage of mentioned stress on plants and increasing the productivity of the above lands. Materials and Methods To study the effect of salinity stress and salicylic acid hormone on biochemical traits of Denak (Oliveria decumbens), a factorial pot experiment was conducted as randomized complete block design with three replications in the research farm of Shahid Chamran University of Ahvaz in 2020-2021. The experiment factors included irrigation water salinity in four levels (1.1 (tap water), 2.5, 5, 7.5 dS/m) and foliar application of salicylic acid hormone in four levels (0, 100, 200, 500 mg/L). Biochemical traits included total soluble carbohydrates, proline, malondialdehyde, sodium and potassium elements, the activity of superoxide dismutase, catalase, guaiacol peroxidase, ascorbate peroxidase, polyphenol oxidase and phenylalanine ammonia lyase enzymes were measured at flowering stage. Results and Discussion The interaction effect of salinity stress and foliar application of salicylic acid hormone significantly changed all the traits of Denak. The foliar application of 200 mg/L of this regulator in plants under 5 dS/m salinity stress increased the amount of total soluble carbohydrates, proline, the activity of superoxide desmutase, catalase, guaiacol peroxidase, ascorbate peroxidase, polyphenol oxidase enzymes compared to the control treatment. The highest level of activity of ascorbate peroxidase and phenylalanine ammonialyase enzymes was also obtained in the foliar spraying of plants under 5 dS/m salinity stress by the concentration of 500 mg/L of salicylic acid hormone, but no significant difference was observed between this concentration and the application of 200 mg/L of this hormone. The maximum concentration of potassium element and the minimum amount of malondialdehyde and sodium element of Denak leaf were also recorded in the treatment of plants under 2.5 dS/m salinity stress by the concentration of 200 mg/L of salicylic acid hormone. The salicylic acid regulator as a very important messenger in the plant's defense system through increasing the stimulation of antioxidant enzymes in Denak reduced the negative effects of free radicals, membrane destruction and the production of malondialdehyde compound in a 2.5 dS/m salinity stress. On the other hand, the application of this hormone reduced the damage caused by the secondary stress of dehydration by increasing the ability of the plant to produce osmotic pressure regulating compounds including soluble carbohydrates and proline. Conclusion Due to its ability to increase the production of osmolyte compounds including soluble carbohydrates and proline, activation of antioxidant enzymes and maintaining the stability of cell membranes, Denak is able to withstand salinity stress up to the level of 5 dS/m. The application of 200 mg/L of salicylic acid hormone is beneficial and recommended due to its positive effect on antioxidant capacity and increasing the production of osmotic protective compounds in the plant as a solution to increase the resistance of Denak against the salinity stress.

Read More Introduction Iranian Kakoti (Ziziphora persica Bunge) which belongs to the mint genus is widely distributed in Iran. In some parts of Iran, it is used as a spice and condiment in food. Iranian Kakoti is also a medicinal plant with antioxidant and antimicrobial properties. Iranian Kakoti is used in traditional medicine as a stomach disinfectant and cold remedy. Since the effect of stresses on the increase of secondary metabolites of plants has been proven, the aim of this study was to investigate the effect of drought stress on the secondary metabolites of Iranian Kakoti in comparison with normal conditions by assessment of the expression of genes which are related to the most important secondary metabolites of the studied plant. In previous studies, it was found that pulegone has the largest share among the metabolites produced in this plant. Materials and Methods Using the KEGG database, the metabolic pathway and genes involved in the production of the pulegone include the four key genes Isopiperitenol dehydrogenase, Limonene synthase, limonene hydroxylase and Isopiperitenone reductase were identified. Due to the lack of genomic information of Iranian Kakoti, the sequence of the four studied genes was predicted by the sequences of these genes in families close to this plant. The predicted sequences were used for primer design. The plants were cultivated under drought stress and normal conditions. The stress condition was achieved by reaching 30% Field Capacity. After observing phenotypic changes due to stress, the amount of proline and also Leaf relative water content (RWC) measured for the plants in both control and stress condition to confirm the accruing of the stress. Then leaf samples were collected. RNA extraction and cDNA synthesis were performed according to the manufacturer's instructions. The quantity and quality of the obtained RNA were evaluated by Nanodrop and gel electrophoresis. The qualified RNA samples were used for cDNA synthesis according to the manufacturer's protocols. Then the comparing the genes expression under drought stress with the control sample was performed. Results and Discussion In response to drought stress, leaf RWC was decreased to 31% in the plants under stress condition and proline levels increased by 1.9-fold compared to control value. Analysis of real-time PCR data showed that. The expressions of limonene synthase, limonene hydroxylase, Isopiperitenon dehydrogenase, Isopiperitenon reductase were increased 115, 400, 50 and 120 folds compared to control respectively. Conclusion The measurement of leaf RWC and proline were found to be beneficial methods to detection of incidence of drought stress in plant. The expression of genes involved in pulegone synthesis compared to the control plants changes with drought stress. The implement of drought stress is a useful method to increase essential oil in Iranian Kakoti.

Read More Introduction Hyoscyamine and scopolamine alkaloids are valuable secondary metabolites that are mainly produced by plants of the Solanaceae family, especially the henbane genus. The production of these alkaloids in henbane is influenced by genetic processes, but environmental factors play a more prominent role in the economic synthesis of these alkaloids. Materials and Methods This research was carried out in 2020 to investigate the effect of environmental factors on the quantity and quality of hyoscyamine and scopolamine alkaloids on 10 henbane populations in northwest of Iran. The seeds of H.niger, H.reticulatus and H.pusillus species were collected from Ardabil and East Azarbaijan provinces and dried in shade. After extracting alkaloids from the populations, the amount of these alkaloids was measured by high performance liquid chromatography (HPLC). To study the physical and chemical properties of the soil, soil sampling was done from the root development depth of each plant (30 cm) in the studied habitats. Results and Discussion The results of variance analysis showed that the effect of above mean sea level factor on the amount of hyoscyamine and scopolamine alkaloids is significant at the 1% probability level. The results of variance analysis showed that soil nitrogen and phosphorus have a significant effect on the increase of hyoscyamine and nitrogen on the increase of scopolamine at the probability level of 1%. While the effect of potassium on the amount of hyoscyamine and the effect of phosphorus and potassium on the amount of scopolamine is not significant. The research results showed significant phytochemical diversity in terms of hyoscyamine and scopolamine among species, populations, and genotypes of henbane. Based on this, H.pusillus species in terms of scopolamine content and H.reticulatus species in terms of hyoscyamine content were at the top of the studied species, and Meshkinshahr population was at the top in terms of scopolamine and hyoscyamine, while Tabriz and Givi populations were at the bottom in this respect. The high amount of alkaloid in some of the studied ecotypes is affected by the high amount of nitrogen, phosphorus, organic carbon in the soil and due to the low potassium in the studied habitats. Conclusion Maximum similarity was observed between the populations of Ahar with Herris, Shabester with Azarshahr, Givi with Tabriz, and Shabester with Khalkhal in terms of the amount of hyoscyamine and scopolamine. In this case, in addition to genetic factors, the similarity of the ecological characteristics of the studied habitats can also be involved.

The Study on the Hexane and Ethanol Extracts Components in Difference Varieties of Narcissus (Narcissus tazetta L.)

Read More IntroductionNarcissus (Narcissus tazetta L.) belongs to the family of Amaryllidaceae is one of the most important ornamental and aromatic plants which its secondary metabolites are being used in cosmetic, sanitary and perfume industries. Behbahan district belongs to Khuzestan province is one of the well-known narcissus production centers in Iran. Narcissus plants not only have ornamental value, but also have medicinal compounds such as alkaloids that have antiviral and anti-tumor properties. Numerous experiments have been identified the components of Narcissus essential oil and the most significant of aroma constituents have been reported as trans-β-Ocimene, Benzyl acetate, 1, 8-cineole, linalool and phenyl ethyl acetate. However, there is little information about Narcissus extract and the current study was performed to investigate the compounds of ethanol and hexan extracts in different varieties of narcissus. Materials and MethodsThis research was conducted in randomized complete design to evaluate components of hexane and ethanol extracts of five narcissus varieties (Shahla, Meskinack, Porpar, Panjegorbai and Golsefied) which are grown in Behbahan district. The fresh flowers of narcissus were extracted using tow solvents; hexane and ethanol. In the hexane extraction method, 10 g of the fresh flowers were weighed from each variety and poured into Erlenmeyer, and then 100 ml of hexane was added to each one and left at room temperature for 10 days. After extraction, the solvent was filtered and the extract was concentrated by vacuum rotary and absolute was obtained. Then 10 times of ethanol was added to the absolute of each variety and kept at -4 °C to precipitate the wax. After precipitation, the supernatant was separated and concentrated again by rotary and then the components were detected by GC/MS. In the ethanol extraction method, also 10 g of the fresh flowers were also weighed from each variety and poured into Erlenmeyer, and then 100 ml of ethanol was added to each one and left at room temperature for 10 days. After extraction, the solvent was filtered and the extract was concentrated by vacuum rotary and absolute was obtained. The rest of the steps are similar to the hexane method which is described above. Results and DiscussionAccording to the results, the maximum amount of hexane extract (1.3%) and the minimum amount (0.9%) were observed in variety of Shahla and in Porpar, respectively. According to the results obtained from the analysis of the extract by GC/MS, approximately 7 to 12 compounds have been identified in narcissus varieties. Benzyl acetate, Linalool, Benzyl alcohol, trans -β-Ocimene, Hydrocinnamyl alcohol, Phenyl ethyl alcohol, 1, 3 Dimethyl ethyl benzene were the most important components. In ethanol extract, the maximum amount of ethanol extract (6.2%) and the minimum amount (4.8%) were recorded in variety of Shahla and in Porpar, respectively. Interestingly, more compounds were identified in ethanol than hexane extract. The most important compounds were 2-butoxy ethanol, 1, 8-Cineol, Benzyl alcohol, trans-β-Ocimene, Linalool, Phenyl ethyl alcohol, Benzyl acetate, Benzene ethanol and Benzene which were recorded in all varieties of narcissus. ConclusionIn general, the results showed that there is a significant difference in terms of yield, number and percentage of chemical compounds of hexane and ethanol extracts of different varieties of narcissus. According to the results, the maximum amount of hexane and ethanol extract was obtained in Shahla while the minimum amount was observed in Porpar. Benzyl acetate, Linalool, Benzyl alcohol, trans-β-Ocimene, Phenyl ethyl alcohol, 1, 3 Dimethyl ethyl benzene are the most important components of both extracts. In order to isolate extract components and report exclusive compounds, further experiments are recommended. Read More ABSTRACT Introduction:Adiantum capillus-veneris Linn is a medicinal plant belonging to Polypodiales order. Pare-siavashan is the name of Adiantum capillus-veneris Linn in pharmaceutical textbooks of Iranian Traditional Medicine. A. capillus-veneris L. as a medicinal plant was utilized to treat various infectious diseases. In this study, the extract of A. capillus-veneris L. from Kurdistan province is investigated for the first time. Materials and Methods:The plant is collected from southeast of Marivan in October 2017. Theaim of the current study was to investigate cytotoxicity, antimicrobial, antioxidant properties and phytochemical screening of methanol extract and polar and nonpolar subfractions of the leaves and stem of A. capillus-veneris L. separately. The antioxidant activity, total phenolic content (TPC) and total flavonoid content (TFC) of the samples were determined using inhibition of free radical 2,2-diphenyl-1-picrylhydrazyl (DPPH), Folin-Ciocalteu, and aluminum chloride colorimetric methods, respectively. Gentamicin, rifampin and nystatin were used as positive controls in an antimicrobial method. Both 50% lethal concentrations (LC50) and 50% inhibitory concentration (IC50) of the samples were identified using regression analysis. Results and Discussion:Theleaves and stem extractsindicated the highest antioxidative activities in DPPH test that IC50 values =56.23 ± 0.85 and 44.66 ± 1.22 μg/ml respectively, which was higher than the synthetic antioxidant butylated hydroxyl toluene (BHT: IC50= 19 ± 1.03 μg/ml). The total phenolic contents of leaves and stem extracts, as Gallic acid equivalents, were 83.62 ± 1.87 and 147.39 ± 2.35 mg/g, respectively. The amounts of total flavonoids compounds of leaves and stem extracts were also determined 58.50 ± 0.78 and 35.63 ± 0.39 mg, respectively. Alkaloids, triterpenes, flavonoids, saponins, tannins were also identified in all the extracts and glycosides were detected only in water and hexane fraction extract of the stem, also hexaneextraction fraction of stem was devoid of triterpenes and the stem and leaves which contain no tannins. The leaves water extract of A. capillus veneris L. showed maximum zone of inhibition against bactrial strains. The hexane extracts of leaves and stem of A. capillus-veneris L. showed the weakest antimicrobial activity. The brine shrimp lethality bioassay was applied for the isolation of anitumour and cytotoxic agents. The leaves and stem extracts of A. capillus-veneris L. showed significant cytotoxic activity with LC50 values of 125.893 and 97.7237 (μg/ml), respectively. Vincristine sulfate (LC50 0.751 μg/ml) was used as the reference standard of brine shrimp lethality bioassay. Conclusion: This study suggested that the extracts of the A. capillus-veneris L. may be a promising source for novel anticancer agents. The extracts showed moderate to acceptable antibacterial activities.

Read More AbstractIntroductionAmong all major genera and species in the mint family, different types of organic compounds or metabolites such as phenolic compounds, flavonoids, anthocyanins, ascorbates, and carotenoids with antioxidant, antimicrobial, and anti-inflammatory effects are produced by means of metabolic processes. Research studies in this field have established that planting distance can have a significant effect on essential oil yield as well as the quantity and quality of secondary metabolites in medicinal plants. The main purpose of spacing between plants is also to provide a suitable mix of environmental factors (i.e. water, climate, light, and soil) to achieve optimum quality performance. Materials and MethodsThe present study was conducted using a factorial experiment based on randomized complete block design (RCBD) with three replications at Ilam University Research Farm, Iran, during 2016-2017. Planting density treatments at three levels (i.e., 20, 14, and 10 plants per m2) were determined based on changes in plant spacing on rows (i.e., three distances of 20, 30, and 45 cm). Two ecotypes were accordingly included: Malekshahi and Sumar. The characteristics measured were photosynthetic pigments (chlorophyll a/b, total chlorophyll, and carotenoid), total phenol and flavonoid content, anthocyanin, as well as essential oil content and yield. Results and DiscussionThe results of this experiment revealed that the amount of antioxidant compound, photosynthetic pigments (i.e., chlorophyll a/b, total chlorophyll, and carotenoid), anthocyanin, and essential oil yield had been significantly affected by ecotype and plant density. Therefore, the highest amounts of total phenol (91.64 mg GAE/g DW) and flavonoid (5.20 mg QUE/ g DW) were observed at 20 and 30 cm row spacing. Total phenol and flavonoid also decreased by 7.2% and 12% as plant density was reduced. Moreover, row spacing of 30 cm increased the amount of chlorophyll a/b by 14 to 16% and carotenoid by 10%. The highest amount of anthocyanin (2.5 μg/g FW) was noticed at 20 cm row spacing. In the same vein, the highest yield of the essential oil was spotted at 20 cm row spacing (91.38 kg/ha) and for Malekshahi ecotype (87.5 kg/ha). Compared with the Sumar ecotype, the Malikshahi had higher content of photosynthetic pigments, total phenol and flavonoid, carotenoid, anthocyanin, as well as essential oil yield content and yield. Growth, quantity, and quality of medicinal plants are controlled by genetic processes as well as environmental and agronomic factors such as plant density. As plant density is enhanced to an optimum level, competition increases for absorption of water, nutrients, and quantity and quality of light received by leaves due to branching density and moderated penetration into canopy. As a result, the rate of cell division and leaf area development reduces and the content of photosynthetic pigments is diminished following the destruction of chloroplast structure and inhibition of biosynthesis of new chlorophylls. On the other hand, secondary metabolites such as phenolic compounds, flavonoids, and anthocyanin develope in the plant in response to environmental conditions in order to protect it against environmental stresses through suppressing oxygen free radicals. In line with the results of the related literature, it was observed that the content of photosynthetic pigments had diminished after increasing the density to 20 plants per m2, but the content of carotenoid, anthocyanin, total phenol and flavonoid, as well as the essential oil yield had improved in the wild thyme (Thymbra spicata L.). ConclusionThe results revealed that the leaves of the plant have good amounts of essential oils, phenolic compounds, and flavonoids. Since the highest percentage and yield of essential oil, phenol, flavonoids, and anthocyanins were observed in the treatment of density of 20 plants per square meter and Malikshahi ecotype, this level of density and Malikshahi ecotype is introduced for production and review in breeding programs.

The majority of the soils in the arid and semi-arid regions face salinity. This is a huge threat for agricultural sustainablitty. With salinity adding-up, plants growth and development greatly decline due to reduced osmotic potential, unbalanced nutrients available and the variations in the type and content of growth regulators which negatively influence the plant yield and productivity. Under salinity situations, using micronutrients plays a vital role in the tolerance of plant against ROS molecules. These nutrients motivate antioxidant enzymes and hence, improve plant responses to the ROS molecules generation. As a result, they improve the yield, quality and productivity of plants under stressful conditions. Materials and MethodsAn experiment was conducted to study the effects of NaCl salinity stress (0, 50, 100 and 150 mM) and Foliar application of common and Nono-form Zinc-oxide (0, 5 and 10 mg L-1) on Rosmarinus officinalis as factorial based on RCBD with three replications at 2017- 2018. Results and DiscussionThe results revealed the interaction effects of salinity levels and Zinc Foliar application on chlorophyll b content, N, P, K, Na and Zn content, leave relative water content as well as essential oil yield. The highest data for chlorophyll b was recorded with no stress × 10 mgL-1 Nono-zinc spray. Relative water content for both Zn forms (0, 5 and 10 mgL-1) was maximum with no salinity treatments. The greatest data for essential oil yield was traced with NaCl0 × 10 mgL-1 Nano-zinc and NaCl0× 10 mgL-1 Zinc Oxide. NaCl0× 5 mgL-1 Nano-Zinc increased K+content in plant. For Zn compositional content, the highest data was recorded with no-salinity × 5 and 10 mgL-1of Nano-Zinc as well as with no-salinity × 10 mg L-1 of common zinc source. Aerial parts dry weight and Fe and Mg content were affected by independent effects of salinity and Foliar application of Zn. Aerial parts dry weight and Mg content increased at no salinity stress and 50 mM salinity. Chlorophyll a and Fe content were influenced by the non salinity levels. Foliar application with 0 and 10 mg L-1nano-Zn increased Mg content in plants. Salinity is one of the predominant stressors influencing the plants throughout all their life cycle. The results revealed that salinity drastically influenced rosemary dry weight, photosynthetic pigments content, as well as Mn, Cu, and Fe content. Zn Foliar application improved aerial parts fresh weight and the content of Mg, Cu, and Fe. In general, rosmary is salinity tolerant up to 50 mM without significant reduction in yield component. Therefore, the Foliar application of Nano-Zinc as Oxide form improved yield and growth characteristics. ConclusionSalinity drastically influenced the growth and physiological responses of the plants. Using Foliar treatments was able to partially ameliorate the salinity adverse effects and, promoted the growth and productivity of rosemary. However, economical administration of Foliar treatments needs more in-depth studies.

Read More Abstract Background and ObjectivesNettle (Urtica dioica L.) is considered a valuable plant with high level of various beneficial phytochemical, which has been used centuries in traditional medicine. Since it contains various secondary metabolites and shows noticeable activity against different Gram-positive and Gram-negative bacteria, nettle has been identified as an appropriate choice for wider applications in the food and pharmaceutical industries in the recent years. This plant is native to Asia, Europe, Africa and North America and grows naturally in different regions of Iran. Although nettle is an important medicinal plant worldwide, there is no information about optimization of its tissue culture conditions. This study was carried out to investigate the effects of different plant growth regulators, explant types and light condition on the callogenesis properties of nettle plant under in vitro condition. Materials and MethodsA factorial experiment was carried out on the base of completely randomized design with three replicates. Different explants including young leaves, old leaves, nodes and seeds of nettle plant were cultured in the solid MS medium supplemented with 30 g l-1 sucrose and different concentrations of plant growth regulators of 2,4-D (1, 2 and 3 mg l-1) and BA (0.1, 0.5 and 1 mg l-1). Callus-related characteristics, including time of callus induction, callogenesis percentage, and their color and texture were analyzed. Analysis of variance was carried out by SAS software ver. 9.3.1 and mean comparison was conducted using Duncan multiple range test. ResultsAccording to the results, callus induction was significantly influenced by explant type. The highest callus induction was attained from young leaves, nodes and old leaves, respectively. The results also showed that callus induction and its properties were significantly influenced by light condition. Preservation of cultures in dark condition resulted in higher callus induction than light condition. In addition, the effect of plant growth regulators was significant on callus induction and its properties. The highest percentage of callus induction was obtained from young leaf explants in the medium containing 1 mg l-1 2,4-D and 1 mg l-1 BA in dark condition followed by node explants in the same plant growth regulator treatment. However, medium supplemented with 1 mg l-1 2,4-D and 0.1 mg l-1 BA showed to be more effective for old leaf explants. Based on the results of this research, young leaf explants in the MS medium containing 1 mg l-1 2,4-D and 1 mg l-1 BA in dark condition are suggested for callus induction from nettle plant. DiscussionType, color and amount of produced callus in different plant species are highly dependent on the genotypes, compositions of tissue culture media as well as the levels of both endogenous and exogenous plant growth regulators. Auxin and cytokinin are two main plant growth regulators playing important roles in callogenesis of different plant species. There is an interaction between the level of plant growth regulators and color of produced callus. Based on the results of present study, it seems that in the nettle plant, node explants endure a higher level of plant growth regulators and hence produce a brighter callus than the leaf segment explants when they are treated with high concentrations of these chemicals.

Methanol spraying, as a biological stimulant on three carbonaceous plants, can increase carbon dioxide stability and reduce respiration, resulting in more dry matter in these plants. Materials and Methods This experiment was carried out to investigate the effect of methanol spraying on seed yield and mucilage variation of D. sophia based on randomized complete block design with five treatments and three replications in a field experiment in Khoy, Iran. The treatments consisted of the control treatment (distilled water without methanol application) with 4 concentrations of methanol spraying (containing 10, 20, 30 and 40%) carried out during the growing season three times at intervals of 12 days (36, 48, 60 days after planting). After the last spraying at the time of full maturity, the plants in each plot were taken individually. The measured morphophysiological characteristics included plant height, plant leaf number, plant stem number, plant dry weight, number of pods per plant, number of seeds per pod, 1000 seed weight and seed yield. Also, the measured phytochemical traits included seed ash content, mucilage percentage and yield, and inflation index. Results Results showed that foliar application of different concentrations of methanol had a significant effect on number of plant stems, number of plant leaves, plant dry weight, number of pods per plant, number of seeds per pod, grain yield, seed mucilage yield, seed ash, seed inflation index (p ≤ 0.01), plant height and 1000 seed weight (p ≤ 0.05). Foliar application of different concentrations of methanol had a significant effect on all morphological and functional components. To be more precise, methanol spray had a significant effect on plant height, number of stems, plant leaf number, shoot dry weight, number of pods per plant, number of seeds per pod, 1000 seed weight and seed yield, but the effect of year on none of the measured traits was meaningful. Also, the interaction between the year and methanol spraying on the traits of plant leaf number, number of seeds per pod and 1000 seed weight had a significant effect, but the remaining traits were not affected considerably. Combined analysis of variance showed that the year effect had no noticeable effect on seed ash. While methanol concentration had a significant effect (p≤ 0.01) on the seed ash content, the interaction of methanol in different years of experiment was not significant on this characteristic. Discussion Regular methanol spraying, as an active bioavailable and a new method in the production of medicinal plants, improved seed yield and mucilage of Descurainia sophia seed. Generally, the highest values for the evaluated properties were obtained in the induction of 40% volumetric methanol solution.

Read More Abstract Background and Objectives An important requirement of agronomic planning is to evaluate different plant nutrition systems in order to achieve high yield and quality, especially in medicinal plants. With the correct method of increasing soil fertilization and plant nutrition, it is possible to reduce the erosion and conservation of biodiversity, preserve the environment, and increase the efficiency of inputs. This research was carried out with the aim of improving the quantitative and qualitative performance of lemon balm with bio fertilizers such as mycorrhiza fungi, biophosphate fertilizer, and vermicompost. Materials and Methods The experiment was conducted in spring and summer of 2016 in a field covered by Agriculture Jihad of Guilan, located in Khoshkabijar District of Rasht, Iran. In this research, the experiment was conducted considering three factors: factorial contains mycorrhizal inoculation (M1 = non-inoculated and M2 = inoculated), phosphate biofertilizers (p < sub>1 = 0, p < sub>2 = 50 and p < sub>3 = 100 kg ha-1) and vermicompost (V1 = 0, V2 = 5 and V3 = 10 tons ha-1) layout based on a randomized complete block design with 18 treatments and three replications. Also, one plot was allocated to control in each replication and only chemical fertilizers (use of 90 kg ha-1 of fertilizer of urea, triple super phosphate 90 kg ha-1 and potassium sulfate 90 kg ha-1) and data obtained from control plot, mycorrhizal, phosphate biofertilizers, and vermicompost were used based on a randomized complete block. Results According to the results of this experiment, the highest number of leaf yield, essential oil percentage, biological yield, essential oil yield, total chlorophyll and carotenoids in inoculation with mycorrhiza was obtained. The application of 100 kg of phosphate biofertilizers had a significant effect on all measured biofertilizers. Fertilizer treatment of 100 kg ha-1 had the best result in all measured traits. Also, the best result was obtained with the use of 10 tons of vermicompost per hectare in biological yield (4808.2 kg ha-1), essential oil (0.13%), and total chlorophyll (1.54 mg gr-1FW). There were positive and synergic interactions between factors. For example, positive interaction between inoculation of mycorrhiza and phosphate biofertilizers on leaf yield and essential oil yield, and synergic interaction between mycorrhiza and vermicompost on biological yield were obsereved. Discussion The results of this study show that the treatment of inoculums with mycorrhiza, consumption of 100 kg of phosphate biofertilizers and 10 tons of vermicompost per hectare could provide the best situation to achieve the highest quantitative (5290 kg ha-1) and qualitative yield (0.26% essential oil) of lemon balm in a sustainable agricultural system.

Read More Abstract Background and Objectives Melissa officinalis is a medicinal plant belonging to the Lamiaceae family. The essential oil of this plant is used in many fields. Although more than 100 types of chemical substances have been identified in this plant, the most important compositions of its essential oil include citral, citronella, geraniol and linalool. Heavy metals are one of the main causes of non-biotic stress for living organisms due to increased use in the field of industrial and agricultural development and its high accumulation and toxicity. Cadmium is an unnecessary heavy metal, which, due to its high mobility and low concentration, easily enters the food chain from the soil. Cadmium induces oxidative stress by stimulating the synthesis of free oxygen radicals in the plant. In this study, the effect of cadmium chloride on oxidative stress induction in lemon balm was investigated. This experiment was carried out in 2016-2017 at Shahrekord University. Materials and Methods This research was conducted in a completely randomized design with three replications in vitro condition. We investigated the effects of different concentrations of cadmium chloride (0, 10, 20 and 40μm) on the biochemical parameters of the lemon balm. In this study, sterile stems of lemon balm propagated on the medium (1/2 MS) were used. After 60 days, seedlings were removed from solid MS medium and the roots were cut and cultured in liquid medium of 1/2 MS with different concentrations of cadmium. The cultivation was carried out in Erlenmeyer flask (250 cc) and was kept in an incubator shaker device. Sampling for the experiment was conducted one week after the growth of the stems in the medium. Results Based on the results obtained in this study, fresh weight of shoots grown in 10 and 20μM of cadmium chloride increased compared to the control. This heavy metal significantly reduced the amount of chlorophyll a, b and carotenoids, while the total chlorophyll content decreased only at 40μm concentration relative to control. Cadmium significantly increased hydrogen peroxide levels in all treatments. Also, it increased the activity of catalase and superoxide dismutase enzymes. Malondialdehyde increased in all treatment concentrations, but this increase was not significant at 40μm of cadmium chloride. Chloride cadmium reduced the absorption of molybdenum and iron, yet increased the adsorption of manganese and zinc. Discussion The results of this study revealed that the cadmium chloride in lemon balm shoots may have the potential to accumulate compounds such as hydrogen peroxide which, as a messenger molecule, produces antioxidants and can help the plant tolerate stressed conditions. It seems that two concentrations of 10 and 20 μm of cadmium chloride have no toxic effects for the stems and the plant has managed to withstand this tension with defense mechanisms.

On the other hand, the combined application of chemical and bio-fertilizers can improve yield and quality of medicinal plants. Basil (Ocimum basilicum L.), a member of Lamiaceae family, is an annual plant and is widely used as a vegetable and as an aromatic plant. Considering the importance of medicinal herbs and the global trend for their production and reproduction in sustainable and low-input agricultural systems as well as a lack of research on the response of Basil's medicinal plant to the combined nutritional systems (chemical and biological), the aim of this study was to compare the effects of various nutritional systems with chemical and biological nitrogen and phosphorus fertilizers as high-quality nutrients on some quantitative and qualitative characteristics of Basil. Materials and MethodsThis research was conducted as a factorial experiment based on a completely randomized design with three replications at the Research Center of Faculty of Agriculture, Shahed University, Tehran, in 2014. The experimental factors included nitrogen and phosphorus fertilizer combinations, 100% chemical based on a soil test, 100% biological (for nitrogen, Azotobacter crocococcus and Azospirillium lipophorum, for phosphorus, and Pseudomonas fluorescence) and combining a system of 50% chemical fertilizers with biofertilizers. To provide nitrogen fertilizer from the source of urea containing 46% pure nitrogen and phosphorus fertilizer were used triple superphosphate containing 46% phosphorus oxide (P2O5). Biofertilizers used to provide nitrogen are a mixture of Azotobacter crocum (Azeto-5 strain) and Azopyrillium lipophorum (93 strain), and for phosphorus from the mixture of Pseudomonas putida (P-168 strain) and Pseudomonas fluorescence (P-169 strain), all of which have a concentration 107 CFU/ml provided by the National Institute of Soil and Water Research of Iran. ResultsThe results showed that different combinations of nitrogen and phosphorus fertilizers and also the interaction between these two had a significant effect on plant height, the number of leaves and shoots yields, percentage, and yield of essential oil. The highest number of leaves per plant was found in treatments N2 × P4 (100% chemical nitrogen with combined of bio-fertilizer and 50% chemical phosphorus) and N4 × P4 (combin of bio-fertilizer and 50% chemical hosphorus combined with bio-fertilizer and 50% chemical nitrogen) respectively with the mean of 54.72 and 54.44 leaves per plant. Interaction of nutrition systems showed that the highest leaves and essential oil yield was in combination N4 × P4 respectively with the average 596.33 kg.ha-1 and 6.56 lit.ha-1. The uses of combination system of nitrogen and phosphorus increased the mean composition of the oils constitutive such as Linalool, Carvacrol and Eugenol compared to control. DiscussionThe results of this study indicate that the use of bio-fertilizers phosphorus and nitrogen, either alone or in combination with 50% chemical fertilizer to improve the quantity and quality of basil had a positive impact. They can, instead of continuous use of chemical fertilizers, be used to optimize the biological inputs for sustainable agriculture and reduce the pollution caused by the use of nitrogen and phosphorous fertilizers.

Read More Abstract Background and Objectives Taxanes are a family of diterpene alkaloids which is currently known as the most important anticancer compounds to suppress and inhibit cell growth, differentiation and proliferation in indefinitely known cancer cell lines. The most well recognized member of the taxane family is taxol (paclitaxel), a minor cytotoxic component derived originally from yew bark (Taxus brevifolia). 10-Deacetylbaccatin III (10-DAB), which is a homolog of taxol, can be extracted at high yields from the needles of Taxus baccata and is currently considered the most appropriate precursor for semi-synthesizing taxol and its analogue, taxotere (docetaxel). The natural source of both compounds is the inner bark and needles of several Taxus species, especially T. baccata and T. brevifolia. Plant cell culture is an environmentally sustainable source of taxol and offers several advantages as it is not subjected to weather, season or contamination. The aim of the present study was to evaluate taxol and 10-DAB content of two Taxus species and their ability in production of both taxanes through cell culture. Materials and Methods The plant materials of T. baccata and T. berevifolia were collected from Shahid Beheshti University (35° 48' N, 51° 23' E at an altitude of 1785 m) and botanical garden of Faculty of Agriculture, University of Tehran, Karaj (35° 48' N, 50° 59' E at an altitude of 1238 m), respectively. Callus culture of both species was initiated from internodal segments on B5 medium supplemented with plant growth reaulators. Cell suspension culture was established on the medium supplemented with 2.0 mg L−1 naphtalene acetic acid (NAA) and 0.1 mg L−1 benzyl amino purine (BAP). Cell viability was determined by Evan’s blue staining test. After 21 days, the amount of taxol and 10-DAB was analysed by HPLC. Results The results showed that mother stocks of T. baccata and T. brevifolia contain 45 and 27 µg/g DW of taxol and 40 and 4 µg/g DW of 10-DAB, respectively. Cell growth was estimated by measuring fresh weight (FW) and dry weight (DW). Results showed that the maximum growth was reached after 21 days of cultivation. Cell cultures of T. baccata and T. brevifolia produced 540 and 20 µg/l taxol and 1 and 110 µg/l of 10-DAB, respectively. Taxol production in T. baccata cell culture was 26-fold higher than in T. berevifolia cell culture. Discussion As far as our literature survey could ascertain, growth and taxol production in T. baccata cell cultures have previously been reported. Although, T. baccata cells are more potent for the production of taxol, T. brevifolia cell culture can be considered for the production of 10-DAB, a homolog of taxol, as an appropriate precursor for semi-synthesizing taxol and its analogue, taxotere (docetaxel). However, it is essential to study and quantify the effect of selected key medium components on growth as well as product accumulation and strike a balance between the two to enhance the yield and productivity.

With this boom in popularity has also come a rise in the need for commercial use, and very little academic literature exists on how to best utilize the preservative solutions (here 8-HQC and essential oils) to prolong cut flowers vase life keeping their full quality. The vase life of Lily is usually short due to vascular occlusion. The use of synthetic chemical fungicides to protect cut flowers raises environmental and human health concerns because of chemical residue routinely found on food meant for consumption. Thus, biocides showing insignificant toxicity to human and organisms are more desirable and less likely to cause negative environmental effects. Materials and Methods The chemical composition of essential oils extracted from the endemic Iranian plants Satureja hortensis L., Thymus vulgaris L. and Citrus limon L. was studied in order to evaluate their efficacy as a substitute for synthetic chemical that was used as preservative-solutions. The effects of essential oils (250 mg L-1) individually and 8-hydroxyquinoline citrate (8-HQC) with sucrose 3% were evaluated. Cut flowers were pulsed for 12 h in different preservative solutions, followed by storage in dry and wet conditions (in distilled water) at 8˚C for 15 days, and then all treated cut flowers were kept in room temperature to evaluate commercial vase life. At the end of commercial vase life, parameters such as relative fresh weight, amount of chlorophyll, phenol, anthocyanin, protein and carbohydrate, vase life and antioxidant enzyme activities were measured. In this experiment, flowering stems were arranged using a completely randomized design. Results Limonene was identified as major component of C. limon L. essential oil, whereas Carvacrol was the major component in S. hortensis L. and Th. vulgaris L. Results showed that applying all essential oil treatments or 8-HQC increased positive characteristics and vase life compared with control. However, among all these treatments, the flowers treated with Thyme essential oil plus 3% sucrose or 8-HQC were kept in wet storage condition showed best vase life. Based on our results, new antimicrobial agents such as limonene, thymol and carvacrol in combination with 3% sucrose had a positive effect on the vase-life and relative fresh weight. The senescence of cut flowers is associated with a series of highly regulated physiological and biochemical processes, including breakage of water balance, degradation of photosynthetic pigment, decrease in metabolic constituents, and loss of membrane integrity. In our experiment, Thyme essential and 8-HQC oil treatments were reduced degradation of leaf protein and chlorophyll content which was seen in the controls. Essential oils derived from plants such as T. vulgaris are particularly valuable because of their antibacterial and antioxidant properties. Discussion In conclusion, using various concentrations of essential oils and 8-HQC in preservative solutions showed promising prospects for the utilization of natural essential oils or plant extracts in extending lily vase-life. The proposed mechanism of action, e.g. inhibition of the growth of bacteria in the vase water or inside the xylem vessels of the flower stem needs further elucidation.

In order to select suitable clones for commercial cultivation in Dezful climatic conditions, in this study morphological and biological production traits of 58 clones of Satureja rechingeri were evaluated. Materials and Methods After initial screening of plant populations, 58 genotypes were selected based on their adaptation into field conditions. To evaluate the morphological and biological production traits of these clones in Dezful climatic conditions, an experiment was conducted based on randomized complete block design with three replications. Morphological and biological production traits of 58 clones were evaluated in the second year of cultivation at the flowering stage. Results Analysis of variance showed significant differences for all studied traits (p< 0.01) among 58 clones. The most important traits varied in terms of plant height (27.83-71.50 cm), fresh weight (20.26-289.71 g/p), flower and leaf weight (4.05 to 83.62 g/p), and plant oil yields (0.07 to 1.54 g/p). Flowers and leaf dry weight had a significant and positive correlation with plant height, number of the main branches and lateral branches, leaf width, plant diameter, fresh weight, dry weight and essential oil yield. Cluster analysis grouped the 58 clones within two main groups. Factor analysis showed that three main factors justified 70.62 percent of the whole variation. Clones of Z28, Z19, K48, E7, H82, F14 and G48 were superior with regard to weight of dry leaves, flowers and essential oil yield. Discussion Superior S. rechingeri clones could be used for commercial cultivation in Dezful climate conditions. The most important biological production traits, yield of leaf and flower fraction, essential oil content and yield showed significant variation between clones. These variations could be used to breed high-quality and desirable cultivars for use in food and pharmaceutical industries.

Perovskia, with the common Persian name of "Brazemble", a small genus from Lamiaceae family, is distributed in various regions of Asia, as Iran, Afghanistan and Pakistan. The genus is represented in Iran by only three species as P. abrotanoides, P. atriplicifolia and P. artemisoides. The plant is an aromatic shrub which mainly grow in mountains at an altitude of 2200 to 4200 m from Northeastern across center to Southeastern of Iran. All parts of the plant are aromatic, but Perovskia is not edible. The plant is used in Iranian folk medicine as an analgesic in rheumatic pains, treatment of leishmaniasis, fever and headache. The present study, was planned to evaluate variations in essential oil composition and contents of different Plant Organs from P. abrotanoides. Materials and Methods In this study, different plant organs (leaf, flower and stalk) of P. abrotanoides were collected from natural habitat in North Khorasan province and their essential oil content and composition were studied. The experiment was arranged in a completely randomized design with three replications for the essential oil contents. Essential oils were analyzed by Gas chromatography (GC) and Gas chromatography–mass spectrometry (GC-MS) in respect to their chemical composition. Results Essential oil contents of leaf, flower and stalk were 1.0, 2.3 and 0.7% (w/w), respectively. The total number of compounds identified and quantified was thirty-nine in leaf, thirty-two in flower, and thirty-five in stalk, representing 99.4, 99.8, and 98.7 % of the total essential oil, respectively. The major compounds of the essential oil were α-bisabolol (2.6-29 %), 1,8-cineole (11.4-16.5 %), camphor (10.1-21 %), α-pinene (1.9-16.2 %) and δ-3-carene (7.4-11.3 %). Results showed that leaf essential oils of the plant characterized with high content of oxygenated sesquiterpenes (41 %). Oxygenated monoterpenes, were rich in flower (42.1 %) and stalk (49.5 %). The main volatile compound identified in the leave was α-bisabolol, which reached a concentration of 29%. The content of essential oil in the flower was higher than that in the leave and stalk. Discussion Results of the current study show that the main compounds and content percentage of essential oil were different in each plant parts. By using the hydrodistillation, 32 to 39 compounds were identified representing 98.7% to 99.4 % of the total oil components. The essential oil analysis revealed that α-bisabolol, 1, 8-cineole, camphor, α-pinene and δ-3-carene, were main essential oil constituents in all organs. The variation observed for essential oil content was 0.7% to 2.3% in stalk and flower, respectively. Chemical variation of P. abrotanoides essential oils from different plant parts, which is significant for conservation and breeding programs, can be considered by medicinal plants breeders and pharmaceutical industries for breeding and processing uses.

Seaweed extracts (SWE) are known to cause many beneficial effects on plants as they contain growth promoting hormones, trace elements, vitamins and amino acids. Humic acid plays a vital role in soil fertility and its application increases the plant growth and nutrient uptake. The present study was undertaken to investigate the effect of SWE, humic acid and chemical fertilizers on the morphological and biochemical characteristics of Trigonella foenum-graecum L. Material and methods The experiment was carried out in 2016 at the Agricultural Research Greenhouse at Imam Khomeini International University, Qazvin, Iran. The experiment was based on a completely randomized design with four treatments including: SWE, humic acid, chemical fertilizers and control and 7 replications. The pots were filled with 5kg of farm soil, which was evenly mixed with recommended level of chemical fertilizers (N; 0.08, P; 0.06 and K; 0.1 g/kg soil) for chemical fertilizer treatment. Seeds were sown at a depth of 1.5 cm in each pot. Seedlings were thinned to five plants per pot 10 days after emergence. In 4 leaves, appearance stage, SWE and humic acid treatments were applied as a foliar spray every 14 days, for three times before harvesting. Growth parameters were measured in correct time. The biochemical constituent pigments such as chlorophylls and carotenoids and also total ascorbic acid in leaf were estimated in control and treated plants using standard methods. Dried seeds of Fenugreekwere subjected to hydro distillation for 3h using a Clevenger-type apparatus to produce oil and essential oil percentage was measured. Results In relation to the mean comparisons, the maximum shoot length and the highest number of pods per plant was obtained in plants receiving SWE and NPK. Maximum plant fresh and dry weights were observed in plants received SWE. All fertilizer treatments increased the number of seeds per pod but there were no significant differences between the treatments. The highest chlorophyll b, total chlorophyll contents, total carotenoids, ascorbic aicd and essential oil percent were obtained in plants that received SWE. Discussion Foliar application of seaweed extract fertilizer on fenugreek plants significantly increased all the examined growth parameters of the plants compared to the control. These results agree with previous studies where growth and plant vigor of dill, coriander and fenugreek plants were enhanced by seaweed extracted treatment. Seaweed extracts create equilibrium in growth as a result of the present auxins which will increase vitamins and hormones produced in the treated plants. Results showed that treatments had a significant effect on total chlorophyll, chlorophyll 'a' and 'b', total carotenoids and essential oil content of fenugreek. Seaweed extracts contain cytokinins as well which induce the physiological activities and increase the total chlorophyll, carotenoids and ascorbic acid in the plant.

Read More Abstract Background and Objectives Organic fertilizers are carbon-based compounds that increase the productivity and growth quality of plants. Municipal waste compost is an organic manure that improves the physical, chemical and biological properties of soil and adds organic matter and exerts a positive effect on elements absorption, quality and yield of saffron. In addition, in saffron cultivation, it is possible to produce considerable amounts of stigma by using of standard mother corms with a minimum weight of 8 g. Therefore, the aim of this research was to investigate the interaction effects of municipal waste compost and mother corm size on chemical properties of saffron. Materials and methods This experiment was conducted with a factorial arrangement based on randomized complete block design with three replications at the Agricultural Research Station, University of Birjand during the growing season of 2014-2015. Treatments were four levels of municipal waste compost (0, 5, 10 and 20 t.ha-1) and three mother corm weights (0.1-4, 4.1-8 and 8.1-12 g). Flower of saffron was measured during autumn of 2015. Total phenol was determined using the Folin-Ciocalteu assay. Free radical scavenging activity of the samples was determined using DPPH method. Secondary metabolites of saffron (crocin, picrocrocin and safranal) were measured according to Iran’s national standard. Results The results showed that the highest total phenol and antioxidant activity (10.0789 mg.100 gr-1 and 65.66%, respectively) was obtained in treatment 20 t. ha-1 compost and the lowest (10.0785 mg.100 gr-1 and 62.58%) was recorded in control. Results showed that addition of the compost rate significantly increased secondary metabolites (crocin, picrocrocin and safranal). So, corm weight had a significant effect on antioxidant activity and the main secondary metabolites and the highest characteristics were obtained for corm with 8.1-12 g. As, the effect of interaction of municipal waste compost and corm weight significantly affected the main secondary metabolites. Discussion The results showed that fertilizer treatment and corm weight influenced chemical properties and the main second metabolites of saffron positively. It has been reported that plants cannot simultaneously allocate resources to growth and defence and that there is competition between proteins and phenolics in plants for the common precursors involved in their biosynthesis .These results led us to presume that saffron plants may utilise benefits from compost fertilizer for their protein synthesis and growth development. On the other hand, organic matter (e.g. compost) acts as precursors or activators of phytohormones and growth substances and secondary compounds in plants. The type of soil and the content of organic compounds in soil can have a decisive effect on increasing the content of organic compounds in soil and strengthening antioxidant activity.

Read More Abstract Background and Objectives Nowadays, the use of sodium nitroprusside as an antioxidant and plant growth regulators to decrease the negative effects of various stresses has been an in portant consideration. Materials and Methods This investigation was conducted as a factorial experiment based on a completely randomized design with five replications at the laboratory of Faculty of Agriculture, Ferdowsi University of Mashhad. The first factor was water stress in three levels (0, 0.5, 1 %) by polyethylene glycol (PEG), and the second factor was use of different concentrations of sodium nitroprusside (SNP) (0, 25, 50, 100 µM), in MS medium. In order to provide the explants, seeds of catmint hairless were disinfected and cultivated in MS medium. When the explants were grown, they were affected by polyethylene glycol and different concentrations of sodium nitroprusside in MS medium. After two weeks, growth characteristics, antioxidant activity, total phenol content and electrical conductivity were measured in explants. Results Water stress reduced the traits related to explants growth such as explants height, distance between internodes, fresh weight of explants, number of nodes and number of branches in each explant. Concentration of 25 µM of sodium nitroprusside had a great impact on improving these traits. The maximum amount of the measured traits was observed in water-stress-conditions using 25 µM of sodium nitroprusside and in without-water-stress conditions. Also, antioxidant activity (85.15%), total phenol content (5.15 mg/g FW) and electrical conductivity (74.92%) were in maximum amount in water-stress conditions by 1% polyethylene glycol application and non-use of sodium nitroprusside that decreased with the use of sodium nitroprusside. Discussion The results showed that using NO as an antioxidant compound reduces the effects of tension generally and the low concentration of nitric oxide (Concentration of 25 µM of SNP) seems to be the best concentration for catmint hairless seedlings under severe drought stress conditions which can reduces the adverse effects of stress and increases the probability of tolerance of seedlings in drought stress conditions by effecting on antioxidant activity of the plant.

Read More Background and Objectives Fennel is one the most important medicinal and aromatic plants which is widely used in food, medicine and cosmetic industries worldwide. This study was carried out in order to investigate the effects of different nitrogen nutritional systems including chemical, organic and biologic ones at different plant densities on yield, yield components and seed essential oil content and yield of fennel in south western Iran climatic conditions. Materials and Methods To do this research, a field experiment was carried out in 2013-2014 agricultural season at Islamic Azad University of Shoushtar agricultural college in split plot design with 18 treatments in 4 replications based on completely randomized blocks design. Different nitrogen system treatments consisting of N1: Uniformly spreading urea fertilizer in the plot, half at planting time and other at stem elongation, N2: Uniformly spreading half of urea fertilizer in the plot at planting time and spraying the other half at stem elongation on fennel foliage, N3: cow manure, N4: Inoculation of fennel seeds with Azotobacter and Azospirillum, N5: Applying half of required nitrogen fertilizer from cow manure and other half from urea, N6: Applying half of required nitrogen fertilizer from cow manure + inoculation of fennel seeds with Azotobacter and Azospirillum were applied to the main plots. Three fennel densities consisting of FD1: 60 plant/m2, FD2: 80 plant/m2 and FD3: 100 plant/m2 were applied to subplots. Results Based on the results, utilization of the first integrated treatment which consisted of cow manure and urea as chemical fertilizer at 50:50 percent, along with using 100 plant per m2 as a proper crop density achieved production of 94.575 g/m2 seed yield containing 3.375 percent of essential oil. Other findings showed that Azotobacter+ Azospirillum had no positive effect on plant seed yield and essential oil. Discussions The findings of our study showed that replacing 50 percent of required nitrogen with cow manure in fennel could lead to a favorable quantitative and qualitative seed production as well as the treatments which use 100 percent of nitrogen as chemical fertilizers. It seems that nitrogen application with manure probably through increasing organic matter mineralization and mineral absorption, improved the effectiveness of manure. Therefore application of such a combination not only could lead to a desirable fennel quantity and quality, but also is more consistent with environment. Other findings of the present study showed that seed inoculation with bio-fertilizers (Azotobacter + Azospirillum) had no positive effects on plant seed yield and essential oil. For such a result, it could be said that climate and soil conditions and also plant species and varieties are important in bio-fertilizers usefulness and also in determining suitable bacteria species for application.

Read More Background and Objectives Satureja khuzestanica and satureja rechingeri are two valuable and endemic medicinal species that grow widely in southwest parts of Iran. These species are rich in phenolic compound caracole; therefore, they have considerable biological activities. In order to evaluate the quantitative and qualitative yields of two savory species (Satureja khuzestanica and S. rechingeri) in different irrigation intervals, an experiment was carried out in the west of Lorestan (Kashkan) during the growing season of 2012-2013. Materials and methods The Experiment was conducted on the basis of split-plot design in completely randomized block design with three replicates. Irrigation treatments (15 and 30 days interval and rainfed) as the main-plot and species in two levels (Satureja khuzestanica and satureja rechingeri)as the sub-plot were carried out. The characteristics measured were leaf and flower yield, essential oil content, essential oil yield, essential oil compositions, leaf area index and plant height. Results Results showed that the effect of irrigation treatments on all traits except for essential oil compositions was significant. The effect of species on essential oil content and essential oil yield was significant. Mean comparison of traits showed that by increasing in irrigation intervals, leaf and flower yield, essential oil content, essential oil yield, leaf area index and plant height of both summer savory species were decreased amount of by 73, 23, 78, 80 and 33%, respectively compared to control (Irrigation interval of 15 days). Between species, essential oil content (3/7%) and essential oil yield (100/2 kg.ha-1) of rechingeri species were more than essential oil content (2/03%) and essential oil yield (62/2 kg.ha-1) of khuzestanica species. Discussion The leaf and flower yield, essential oil content, essential oil yield, leaf area index of both summer savory species were decreased by increasing irrigation intervals and rainfed cultivation. Trend of changes in essential oil yield was similar to leaf and flower yield, In other words, the essential oil yield was further affected by the herb yield (leaf and flower yield).
