Read More IntroductionThe endosperm (3n) is the food source for the embryo, which has received two polar nuclei from the maternal parent 2n and one sperm cell from the paternal parent 1n. Traditionally, triploid plants are created by crossing between tetraploid plants and diploid plants. To carry out this method, it is necessary to create tetraploid lines, and it is possible to double the chromosomes of diploid species by using substances that inhibit the formation of the division spindle. The advantage of endosperm culture over the traditional method is that the production of triploid plants in endosperm culture is done in a single step, and the time required to generate a triploid plant is less than in the traditional method. Increasing the level of ploidy of the nucleus can affect the plant's physiological and biochemical processes as well as altering the rate of photosynthesis, respiration, activity of genes, enzymes, and expression of isoenzymes compared with diploid types. They will have a new phenotype and can surpass the range of traits of their diploid ancestors such as increasing biomass and changing the quality and concentration of plant active ingredients, increasing resistance to drought stress, and resistance to pests. Therefore, the chance of their selection will increase.Materials and methodsImmature seeds were collected in late May, about 35 to 45 days after pollination, and mature seeds were harvested after fully ripening approximately in late July, and disinfection using a stereomicroscope with 60 times magnification under a laminar hood. The endosperm tissue was separated from the seed coat and embryo. In this study, mature and immature endosperm tissues of the mooseer plant were cultured onto hormonal compounds for callus induction in dark conditions for three months. In further experiments, the effects of sucrose were investigated at two levels of 30 and 50 gl-1 and the combination of BAP and NAA treatments on mooseer production. The flow cytometry method and chromosome counting were used to determine the ploidy levels of regenerated plants. ResultsIn this study, the treatment of 1 mg L-1 of NAA and 1 mg L-1 of BAP indicated the highest percentage of callus formation in both mature and immature tissues of mooseer endosperm. Seedlings produced in the treatment of 0.5 mg L-1 of NAA and 3 mg L-1 of BAP in culture medium with 50 g of sucrose caused the production of bulblet with greater length and diameter. The present study identified the cytogenetic properties, chromosome counts, and stomatal sizes of triploid plants regenerated from both mature and immature endosperm tissue of mooseer, using flow cytometry. A total of 12 triploid seedlings were obtained from mature tissue, while 8 seedlings were obtained from immature tissue.ConclusionTraditionally, triploids are produced by hybridization between induced superior tetraploids anddiploids, but endosperm culture is a one-step protocol. In this study, according to the sexual and asexual propagation (propagation through daughter bulbs) of mooseer, the modification and production of the triploid plants in mooseer allow the propagation of this plant through daughter bulbs. Generally, triploid plants were successfully produced through the endosperm culture method.

Mutation induction is a method to increase genetic variety that is used in plant breeding along with selection, recombination or a combination of the both. Rosa persica Michx can serve as a valuable genetic resource for breeding programs, especially due to the market demand for variety in rose cultivars as well as its drought resistance. In two independent experiments, this study examined the effect of ethyl methane sulfonate (EMS) as a chemical mutagen on seed germination and seedling characteristics of Rosa persica. Materials and Methods The seeds of Rosa persica Mich.seeds were collected from an adult shrub on the Arak University campus, Karbala St., Sardasht (34° N, 49.6° E) and an altitude of 1872 m above sea level during mid-summer. The seeds were separated from the hips by a sharp scalpel and the damaged and abnormal seeds were removed. Before applying the cold stratification, the seeds were washed for several hours and then placed in layers of wet sand at 4 ° C for 60 days.First, the effects of 0, 0.2, and 0.3% EMS at 4, 8 and 12 hours on seed germination characteristics were examined. Second, 0, 0.2, and 0.3 concentrations of EMS at 4 and 8 hours were studied on reproductive buds. Seed germination characteristics such as final germination percentage, germination rate index, coefficient velocity germination, germination rate, mean daily germination, daily germination speed, mean germination rate, germination index, seed vigor, seed dormancy and seedling vegetative traits such as stem lengths, root lengths, and leaf number are evaluated at first experiment. In second experiment, flower diameter, flower opening rate, number of necrotic buds and the number of seeds in each hip were measured. Results and DiscussionThe results showed that increasing the concentration of EMS and the time of seed treatment caused a further decrease in germination indices. In 0.3% EMS treatment for 8 hours on seeds, traits such as germination percentage, germination rate index and daily germination were reduced to zero. According to the results of the second experiment, leaf and seedling lengths were longer at 0.2 and 0.3% EMS concentrations and at 4 h incubation. There was a decline in the number of seeds per hip and an increase in necrotic buds with increasing concentration and time of EMS application. In order to succeed in a breeding program using mutagenic methods, the appropriate concentration and time must first be optimized. To determine the best concentration of mutagens, there are two important factors to consider: first, that the concentration should not be so high as to destroy plants, and second, that the applied concentration should be selected moderately. Although, that the frequency of mutations occurs sufficiently. ConclusionIn order to induce a positive mutation and diversity in Iranian roses for the purpose of introducing a new cultivar, seed treatment with a concentration of 0.3% EMS and an incubation time of 4 h is recommended. It is also recommended to apply a concentration of 0.2% EMS on the reproductive buds for 4 h to induce mutations. Keywords: Seed, breeding, germination, mutation, ornamental plant

Today, agricultural machinery is one of the most important sources of environmental pollutants in mechanized farms and pollution caused by them can disrupt the germination process and plant growth. Therefore, in addition to a variety of environmental challenges, such as salinity and drought stress, contamination from agricultural machinery can also be added to germination and seedling growth as a threat. Materials and MethodsBased on the gray correlation coefficient between the comparison series and the standard series, a relative series can be created. If γ (x_0, x_i) ≥γ (x_0, x_j), which indicates the degree of correlation between x_i and x_0, is greater than x_j and x_0; In this case, the relationship between potential causes and the optimal amounts of decision factors is shown. In the sense that a larger degree of connection has a smaller effect. Therefore, increasing the degree of prioritization of risk indicates the potential reasons that need to be improved. To analyze the risk of gaseous pollutants leaving the tractor exhaust, the treatment with the lowest gray correlation coefficient should show the highest risk priority. So that the highest degree of gray connection determines the best state of seed treatment. The aim of this study was to investigate the effect of pollutants from tractor exhaust in three different initial life, useful life and burn on canola seed germination. This factorial experiment was performed based on a completely randomized design with three replications. Experimental treatments include three different tractor life periods including initial life, useful life and wear as the first factor, and treatment for the duration of seeds exposed to tractor exhaust pollution including zero, 30, 60, 90, and 120 minutes as the factor. The second was considered. Results and DiscussionThe results showed that the highest values of root and stem length, germination rate coefficient, germination index, germination rate index and canola seed vigor were obtained from the absence of exhaust pollution; while the lowest amount of these traits was related to the treatment of exposure to contamination for 120 minutes and worn tractor. Conversely, the amount of seed protein and the activity of antioxidant enzymes catalase and peroxidase increased with increasing tractor wear and duration of contamination. Calculations based on gray theory showed that the treatment with a lower rank is the preferred treatment and has a lower risk according to all growth indices considered for rapeseed. In this study, zero time for different tractor lives without treatment, the risk was identified. Therefore, it is likely that increasing the exposure time to the tractor exhaust contaminant will gradually reduce the seed tolerance and poisoning threshold. ConclusionGray's theory has shown that the duration of exposure is more important than the amount of contamination. The severity of exposure also varied depending on the degree of infection. Rapeseed seeds were more sensitive to contaminants from worn-out tractors than new tractors. The sensitivity of these seeds doubles the need to replace worn-out machines, service and repair tractors in a timely manner, use clean fuel, and use new or serviceable tractors.
