Read More Introduction Wheat is one of the plants that is important in terms of diet and business, and the global demand for it is increasing. One of the harmful and dangerous stresses is ultraviolet light, which affects the growth and production of plants and leads to the production of reactive oxygen species, which are very toxic. The present study was evaluated in order to investigate the effect of C type ultraviolet light on seedlings of three varieties of wheat in order to identify the systems involved in transmitting the defense message of this plant from biochemical and molecular aspects. Materials and Methods The experiment was carried out in a factorial manner based on a completely randomized design with three replications at Mohaghegh University of Ardabil. The investigated factors included the first factor of Control and treatment with ultraviolet light (type C with a wavelength of 254 nm) and the second factor of wheat cultivars (Sardari, Morvarid, and Gaspard). Type C ultraviolet light stress was applied at the stage of three-four leaves for 20 minutes and then sampling of the treated leaves at zero, 1, 2, 4, 6, 10, 18, 24 and 48 hours later. The treatment was done. Results and Discussion The results showed that the quantitative activity of peroxidases in Sardari and Gaspard cultivars was not significantly different in front of type C ultraviolet light, while in Morvarid cultivar, this enzyme increase had an upward trend from the very first hours. Also, the results of the qualitative examination of peroxidase enzymes against C type ultraviolet light showed the number of 5, 6 and 9 peroxidase isoenzymes in three of Sardari, Morvarid and Gaspard wheat cultivars. Qualitative analysis of catalase enzymes in Sardari and Gaspard cultivars identified 4 isoenzymes and 3 isoenzymes in Morvarid cultivar. On the other hand, in DNA electrophoresis, the effect of type C ultraviolet light on the DNA of three wheat cultivars showed that DNA breakage occurred in Sardari and Gaspard cultivars 48 hours after treatment compared to control samples and in Morvarid cultivar, from the first hours after treatment. Investigation the changes in expression of genes involved in metabolism such as hexokinase and plant defense such as PR1 in three wheat cultivars as a result of treatment with type C ultraviolet light shows that the expression changes of these two genes in Sardari and Gaspard cultivars follow almost the same pattern and 10 hours after From the treatment, they have shown a decrease in expression, but in Morvarid cultivar, the expression of HK and PR1 genes increased from the very first hours. Conclusion In general, the results show that the increase in the activity of peroxidase and catalase enzymes and changes in the expression of PR1 and HK genes in the path of transmission of ultraviolet light C type in three cultivars Sardari, Morvarid and Gaspard gives. DNA breakage along with other changes in wheat indicate the possibility of programmed cell death in wheat.

Read More AbstractIntroductionLentil is used mainly for human consumption as a source of protein and carbohydrates in soups, stews and vegetarian dishes. It is grown to improve economical returns to producers, diversify and lengthen crop rotations and reduce the requirement for nitrogen fertilizer. Lentil, a member of the legume family, Leguminosae, can supply a significant part of its Nitrogen requirement by fixing Nitrogen from the air when inoculated with the appropriate rhizobial inoculant. The pattern of nutrients in the proximal composition of lentil is similar to that of other grain legumes, but seed protein content (19.5–35.5%) is less than in soybean. Fiber concentration is low and is largely within the seed testa, so the fiber in lentil meal can be reduced if it is de-hulled before grinding. In addition to high-quality protein, essential amino acids, and major minerals, the seed contains iron up to 505 mg kg-1 and zinc up to 330 mg kg-1 on a whole seed basis. Amino acids are help in tissue protein formation. Some amino acids are not synthesized in the body and it is necessary to take them in diet. Lentils contain different amino acids that can be used by most people. This research was carried-out to study effect of different amino acids on the activity of antioxidant enzymes, Proline content and seed yield of lentil in delayed planting. Materials and MethodsThe research was conducted as a factorial experiment based on a randomized complete block design with three replications in the Research Farm of Agricultural Faculty, Tarbiat Modares University, through May to July, 2018. The studied factors included mode of application (priming, spraying and priming + spraying) and type of amio acids (Arginine, Aspartic acid, Proline, trade amino acid (proamin) and distilled water (control). Analysis of variance (ANOVA) and also mean comparisons were accomplished using the general linear model (GLM) procedure. LSD procedure at a probability level of 0.05 was used to determine statistically significant differences among treatment means. Results and and DiscussionThe results showed that the highest total dry weight was obtained from the application of Arginine spraying (2330 kg ha-1), priming + spraying aspartic acid (2388 kg ha-1) and trade amino acid (2219 kg ha-1). The highest amount of catalase (CAT) activity was observed in the treatment of distilled water by foliar application and its least effective activity was observed in the application of Arginine as priming. The application of aspartic acid as acidic acid with 888.7 kg ha-2 produced the highest yield. As a result, it is more than three times the performance of using distilled water (control). The application of Arginine (555.1 kg ha-2), commercial amine (444.0 kg ha-2) and Proline (327.8 kg ha-2) were established in the next rank, respectively. According to the results of this study, amino acids can reduce the stress of heat. In the absence of amino acid, yield reduction will be higher. Although amino acids are a Nitrogen source, the routine concentrations involved in exogenous supply are so low that its positive effects can be attributable to an increase in Nitrogen availability. ConclusionAspartic acid and Arginine due to their role in the Nitrogen cycle and also, due to the fact that amino acids are the building blocks of proteins, improved lentil yield. Therefore, it is recommended to use amino acids to increase the yield of lentils. The application of aspartic acid via both priming and spraying is suggested as the best treatment.

Read More Abstract Background and ObjectivesGerbera is one of the important cut flowers in the floriculture industry, but it is sensitive to early wilting and neck bending which reduce its postharvest vase life. In cut flowers, water deficits are created when transpiration is more than water absorption. Therefore, petal wilting and neck bending increase in these flowers. These conditions are mainly due to occlusion of the vascular bundle, which can occur in different ways, such as the activity of pathogenic bacteria and other microorganisms in vascular bundle. In recent years, due to the adverse effects of chemical antimicrobial compounds on human health and the environment, many studies have been conducted to find replacement materials to maintain the quality and vase life of cut flowers in postharvest. Therefore, in this study, the effect of different concentrations of harmel aqueous-alcoholic extract (Peganum harmala L.) on bacterial population and vas life of ‘Stanza’ and ‘Pink Elegance’ cultivars of Gerbera cut flower were studied. Materials and MethodsThe effect of concentrations 0, 0.25, 0.5, 0.75 and 1% of the aqueous-alcoholic extract of Harmel plant was studied in a factorial experiment based on completely randomized design with four replications. At harvest time, 3, 6 and 9 days after harvest (at 20 °C and 70% relative humidity), the effect of treatments on the bacterial population of the vase solution and end stem, vase life and some characteristics of ‘Stanza’and ‘Pink Elegance’ Gerbera cut flowers were studied. ResultsThe results showed that the inhibitory effect of harmal extracts on gram-positive bacteria was higher than gram-negative bacteria. Also,‘Stanza’cultivar had better quality and longer vase life than ‘Pink Elegance’ cultivar. By increasing the vase life of flowers, the quality of these flowers decreased. In the first place, however, concentrations of 0.25% and then 0.5% of the harmal extract performed better than other treatments in various aspects: first, they reduced the bacterial solution, stem end, and electrolyte leakagein stem and petals; second, they preserved quality (discolored of vase solution, discolored of petals and stem), relative fresh weight, stem diameter, totalsolublesolids, catalase activity and phenylalanine ammonia lyase activity; and finally, they increased vase life of gerbera flowers in both cultivars. Concentrations of 0.75% and 1% increased allelopathic substances in cut flower. While these treatments decreased the bacterial population in vase solution and stem end, they did not have a significant effect on the quality and vase life of gerbera cut flowers. DiscussionThe results of this study showed that the different concentrations of harmal extract significantly reduced the growth of microbial agents, including the bacterial population, but by increasing the concentration of the harmal extract, the allelopathic effects of this plant extract on gerbera cut flowers were harmful and did not affect their vase life and quality. Therefore, the concentration of 0.25% then 0.5% reduced the bacterial populations in vase solution and stem end of cut flowers and preserved the quality and vase life of ‘Stanza’ and ‘Pink Elegance’ cultivars. Therefore, the use of low concentrations of the harmel aqueous-alcoholic extract is recommended to reduce the microbial population of the vase solution and maintain the quality and vase life of the gerbera cut flowers.
